Pathways from the superior colliculus and the nucleus of the optic tract to the posterior parietal cortex in macaque monkeys: Functional frameworks for representation updating and online movement guidance.

The posterior parietal cortex (PPC) integrates multisensory and motor-related information for generating and updating body representations and movement plans. We used retrograde transneuronal transfer of rabies virus combined with a conventional tracer in macaque monkeys to identify direct and disynaptic pathways to the arm-related rostral medial intraparietal area (MIP), the ventral lateral intraparietal area (LIPv), belonging to the parietal eye field, and the pursuit-related lateral subdivision of the medial superior temporal area (MSTl). We found that these areas receive major disynaptic p

Single-nucleus RNA sequencing of developing superior colliculus identifies neuronal diversity and candidate mediators of circuit assembly.

The superior colliculus (SC) is a sensorimotor structure in the midbrain that integrates input from multiple sensory modalities to initiate motor commands. It undergoes well-characterized steps of circuit assembly during development, rendering the mouse SC a popular model to study establishment of neural connectivity. Here we perform single-nucleus RNA-sequencing analysis of the mouse SC isolated at various developmental time points. Our study provides a transcriptomic landscape of the cell types that comprise the SC across murine development with particular emphasis on neuronal heterogeneity.

Synaptic inputs and their pathways from fixation and saccade zones of the superior colliculus to inhibitory burst neurons and pause neurons.

The caudal part of the superior colliculus (SC) plays an important role in the generation of saccades, whereas the rostral part of the SC is … g from PNs revealed that they receive convergent excitation from the rostral parts of the bilateral superior colliculi and that the rostral SC inhibits IBNs on both sides via PNs. The neural connections determined in this study support the functional independence of the rostral SC and are consistent with the notion that the "fixation zone" is localized in the rostra …

The mosaic architecture of the superior colliculus.

The superior colliculus is a midbrain structure serving visual, multisensory and sensorimotor processing. Throughout various collicular layers, visual afferents are linked together with afferents related to other sensory modalities as well as with afferents from sources not easily subsumed under the term 'sensory'. These inputs are orchestrated in a topographic fashion and led to premotor neurons that are important elements in generating saccadic eye movements and orientation movements of other kinds. Using immunocytochemical techniques to chart the distribution of various substances serving n

Changes in retinal arbors in compressed projections to half tecta in goldfish.

In adult goldfish, electrophysiological studies have shown that the retinotectal projection reorganizes, following removal of half of the tectum, to form a complete but compressed projection over the remaining half tectum. As a result, each fiber terminates more rostrally than normal. Electron microscopic studies suggest a competition between retinal fibers for a fixed number of synaptic sites. The current study examines whether retinal arbors in the compressed projection are smaller than normal in extent or branching and whether the fiber paths in the tectum show the rostral movements and the

The effect of binocular lid suture on auditory responses in the guinea-pig superior colliculus.

Guinea-pigs were deprived of visual experience from soon after birth until the terminal auditory mapping experiment (performed between 57 and 96 days after birth). The visual deprivation was effectuated by binocular eyelid suture. Multi-unit responses were obtained from the superior colliculus (SC) in response ot free field presentation of bursts of broad band noise. In lid sutured animals a topographically ordered auditory map of azimuthal space was demonstrated in the SC. The auditory responses were, however, not as precise or tuned when compared with normal animals. Nevertheless, lid suture

The organization of descending tectofugal pathways underlying orienting in the frog, Rana pipiens. II. Evidence for the involvement of a tecto-tegmento-spinal pathway.

In the frog, identical orienting deficits, involving a failure to turn toward stimuli in the ipsilateral hemifield, can be produced by small white matter lesions either in the caudal mesencephalon (Kostyk and Grobstein, 1987a) or in the caudal medulla (Masino and Grobstein, 1989). These findings suggest that descending turn signals may run uninterrupted from the midbrain to the spinal cord, and that something other than tectospinal axons may carry such signals. We here report studies to determine whether there is a tecto-recipient structure whose axons pass through the known critical lesion si

After-effects of moving textured background in motion-sensitive neurons of anuran optic tectum.

Motion-sensitive neurons in anuran optic tectum were shown to respond to a stationary object centered in the excitatory receptive field, if a textured background moved for a while and then stopped ('motion after-response'). This motion after-response was attributed to a post-inhibitory rebound activation derived from effects of masking the excitatory receptive field center surrounded by an antagonistic inhibitory region. It was suggested that a similar rebound activation mechanism may also be involved in a certain type of perceptual motion after-effects in humans.

Topographical projections from the posterior thalamic regions to the striatum in the cat, with reference to possible tecto-thalamo-striatal connections.

Projections from the posterior thalamic regions to the striatum were studied in the cat by the anterograde tracing method after injecting wheat germ agglutinin-horseradish peroxidase conjugate (WGA-HRP) into the caudalmost regions of the lateroposterior thalamic nucleus (caudal LP), suprageniculate nucleus (Sg) and magnocellular division of the medial geniculate nucleus (MGm). The results were further confirmed by the retrograde tracing method after injecting WGA-HRP into the regions of the caudate nucleus (Cd) and putamen (Put) where afferent fibers from the caudal LP, Sg and MGm were distrib

[Experimental anatomic study of the topography of the retinofugal projections in Discoglossus pictus (Amphibia, Anura). II. The tectum: existence of a double-system of primary optic projections].

The possibility of retinotopic organization of bilateral retinotectal projection was studied by fotal retinal lesions followed by the Fink-Heimer technique. This work in Discoglossus pictus (Amphibia, Anura) shows two types of direct visual projections: a contralateral projection within the tectal layer 9 and a bilateral projection on tectal layer F of Potter (1969). The ipsilateral projection within tectal layer 9, earlier described after enucleation (Picouet et Clairambault, 1977), is not observed here. We discuss about reasons we failed to reveal it. Although appearing to be grossly in acco

Retinotectal map formation in dually innervated tecta: a regeneration study in Xenopus with one compound eye following bilateral optic nerve section.

Retinotectal map formation was studied during regeneration in young adult Xenopus. Right compound double-temporal eyes (TT) were formed in tailbud stage embryos by the fusion of two temporal halves of the eye blastema in the same orbit. In other animals right compound double-nasal eyes (NN) were prepared. In both combinations the left eye was kept intact. After metamorphosis the right and left optic nerves were sectioned to induce optic fiber regeneration from each eye to both tecta. The patterns of retinotectal projections from the compound and normal eyes were studied from 37 to 364 days aft

Retinotopic analysis of fiber pathways is amphibians. I. The adult newt Cynops pyrrhogaster.

The possibility that pathways of retinal fibers within the optic tract and the tectum of the adult newt are retinotopic was examined by selective labeling of the retinal fibers with horseradish peroxidase. Within the optic tract fibers from the ventral, temporal and dorsal retinal quadrants were ordered from the dorsal to ventral edges of th optic tract. The nasal retinal fibers exhibited two different pathways. The fibers from the dorsonasal retina ran along the ventral edge of the optic tract, while the fibers from the ventronasal retina ran along the dorsal edge of the optic tract. Segregat

Retinofugal and retinopetal connections in the upside-down catfish (synodontis nigriventris).

The retinofugal and retinopetal connections in the upside-down catfish Synodontis nigriventris were studied by use of the horseradish-peroxidase (HRP) techniques, autoradiography, and degeneration-silver methods. An unusual retinal projection to the torus semicircularis as well as projections to the retina from three different sources in the brain are described. After intra-ocular injections of HRP, labeled cells were found in the optic tectum, the dorsomedial optic nucleus and one of the pretectal nuclei. These new findings support the basic hypothesis (i) that neuronal connections are more e

Connections and visual-field mapping in cat's tectoparabigeminal circuit.

1. The aim of these experiments was to analyze the organization of the reciprocal connections between the cat's superior colliculus and parabigeminal nucleus. Both physiological and anatomical techniques were employed. 2. A population of cells in the superficial gray and upper optic layers of the colliculus was labeled retrogradely by horseradish peroxidase injections into the parabigeminal nucleus. No other sources of input to the nucleus were found in the brain stem or diencephalon. 3. A map of the visual field within the parabigeminal nucleus was reconstructed by plotting visual receptive f

Identification of cells of origin of tectopontine fibers in the cat superior colliculus: an experimental study with the horseradish peroxidase method.

The pontine projections from the superior colliculus in the cat have been studied by means of retrograde axonal transport of horseradish peroxidase (HRP). Following injections of HRP in the dorsolateral pontine nucleus, where the tectopontine fibers terminate, a fair number of labeled cells are found throughout the rostrocaudal extent of the ipsilateral superior colliculus. Relatively few of the labeled cells are of medium size (25-40 micron in diameter), more than 80% are small (10-25 micron), but no large cells are labeled. The cell bodies giving rise to tectopontine fibers are distributed i

Quantitative light microscopic autoradiographic localization of cholinergic muscarinic receptors in the human brain: brainstem.

We have investigated the localization of muscarinic cholinergic receptors in the brainstem of eight patients free of neurological disease following quantitative autoradiography of microtome sections of postmortem tissue labeled in vitro with N-[3H]methyl scopolamine as a ligand. Receptor densities were quantified by microdensitometry with the aid of a computer assisted image analysis system. Our results reveal a heterogeneous distribution of receptor sites. High concentrations of muscarinic cholinergic receptor sites were associated with many nuclei and areas of the brainstem including the nuc

The superior colliculus motor region does not respond to finger tapping movements in humans.

Electrophysiological studies in macaques and functional neuroimaging in humans revealed a motor region in the superior colliculus (SC) for upper limb reaching movements. Connectivity studies in macaques reported direct connections between this SC motor region and cortical premotor arm, hand, and finger regions. These findings motivated us to investigate if the human SC is also involved in sequential finger tapping movements. We analyzed fMRI task data of 130 subjects executing finger tapping from the Human Connectome Project. While we found strong signals in the SC for visual cues, we found no
