Sympathetic nervous system-mediated fibro-adipogenic progenitor mobilization drives stroke-related sarcopenia.

Patients who survive stroke usually experience rapid muscle wasting and an increased risk of physical disability. Although multifactorial in … external perturbations. In the present study, we report that FAPs rapidly respond to the overactive sympathetic nervous system (SNS) and egress from the muscle niche into circulation during the acute phase of stroke. FAP-specific ablation of adrenoceptor beta 2 (Adrb2) markedly ameliorated stroke-related sarcopenia, highlighting the central role of SNS-mediated FAP loss in …

PET imaging of cardiac sympathetic nervous system dysfunction using meta-[18F]fluorobenzylguanidine.

PURPOSE: Cardiac sympathetic nervous system (SNS) dysfunction is a hallmark of and contributor to the development of heart failure (HF) and cardiac arrhythmias. Positron emission tomography (PET) radiotracers targeting the norepinephrine transporter (NET) have demonstrated utility in assessing global and regional cardiac sympathetic innervation. To date, the scarcity of fluorine-18 containing SNS radiotracers demonstrating NET dependency in rodents has precluded research in commonly used preclinical cardiac disease models. We demonstrate the utility of [18F]mFBG cardiac imaging by performing P…

Motor innervation directs the correct development of the mouse sympathetic nervous system.

The sympathetic nervous system controls bodily functions including vascular tone, cardiac rhythm, and the "fight-or-flight response". Sympathetic chain ganglia develop in parallel with preganglionic motor nerves extending from the neural tube, raising the question of whether axon targeting contributes to sympathetic chain formation. Using nerve-selective genetic ablations and lineage tracing in mouse, we reveal that motor nerve-associated Schwann cell precursors (SCPs) contribute sympathetic neurons and satellite glia after the initial seeding of sympathetic ganglia by neural crest. Motor nerv…

Ventromedial Hypothalamus Activation Aggravates Hypertension Myocardial Remodeling Through the Sympathetic Nervous System.

Background: The ventromedial hypothalamus (VMH) is an important nuclei in responding to emotional stress, and emotional stress is a risk factor for cardiovascular diseases. However, the role of the VMH in cardiovascular diseases remains unknown. This study aimed to investigate the effects and underlying mechanisms of VMH activation on hypertension related cardiac remodeling in two-kidney-one-clip (2K1C) hypertension (HTN) rats. Methods: Eighteen male Sprague-Dawley rats were injected with AAV-hSyn-hM3D(Gq) into the VMH at 0 weeks and then randomly divided into three groups: (1) sham group (sha

Angiotensin II, Oxidative Stress, and Sympathetic Nervous System Hyperactivity in Heart Failure.

In congestive heart failure (CHF), sympathetic nervous system is hyperactive. This article reviews current understandings about central and peripheral neural mechanisms underlying sympathetic hyperactivation in this pathological condition. During the development of CHF, renin-angiotensin system (RAS) activities and angiotensin II-mediated oxidative stress become enhanced. Here, on the basis of findings obtained from animal studies, it is examined how RAS overactivation and oxidative stress in central and peripheral nervous systems of CHF mediate sympathetic hyperactivation. Mechanisms by which…

The sympathetic nervous system in polycystic ovary syndrome: a novel therapeutic target?

Polycystic ovary syndrome (PCOS) is a common endocrine condition associated with long-term health risks, including type 2 diabetes and vascu … d pressure in two PCOS subjects with hypertension, accompanied by improved insulin sensitivity. The sympathetic nervous system may thus offer a new therapeutic target in PCOS but larger and longer-term studies are needed before these treatments can be considered in clinical practice.

Uncoupling of sympathetic nervous system and hypothalamic-pituitary-adrenal axis in cirrhosis.

BACKGROUND AND AIM: The hypothalamic-autonomic nervous system (HANS) axis and the hypothalamic-pituitary-adrenal (HPA) axis are stimulated in parallel in response to stress factors under healthy conditions. This physiological synergism of the axes aims at optimizing anti-inflammatory actions. Therefore, we investigated whether this synergism is altered in patients with liver cirrhosis. METHODS: As a typical marker of the HANS axis neuropeptide Y (NPY is a neurotransmitter of the sympathetic nerve terminal) and of the HPA axis, cortisol together with adrenocorticotropic hormone (ACTH) and cort

Sympathetic nervous system exclusion following experimental subarachnoid haemorrhage prevents vasospasm in rabbits.

BACKGROUND: The objective of this study was to establish whether the exclusion of the effect of the sympathetic nervous system prevents vasospasm of cerebral arteries after experimental subarachnoid haemorrhage in rabbits. METHODS: The effect of sympathetic exclusion on vasospasm was studied in 29 New Zealand rabbits under conditions similar to human subarachnoid haemorrhage: 1. The activity of the sympathetic nervous system was excluded only after subarachnoid haemorrhage. 2. The effect of this exclusion was evaluated on the eighth day after subarachnoid haemorrhage. 3. The single haemorrhag…

Hemodynamic and sympathetic nervous system responses to stress during the menstrual cycle.

In this study, the impact of menstrual cycle phase on hemodynamic and sympathetic nervous system responses was examined during exposure to a battery of laboratory stressors. Participants were 40 healthy premenopausal women, aged 26 to 51. Impedance cardiography was used to measure stroke volume, heart rate, and cardiac output. Systemic vascular resistance was derived on the basis of concurrently recorded blood pressure and cardiac output. The menstrual cycle's effect on the sympathetic nervous system response was explored by evaluating plasma catecholamine responses during stress. In luteal co…

Cocaine acts in the central nervous system to inhibit sympathetic neural activity.

Cocaine was administered i.v. to decerebrate cats while monitoring cardiac preganglionic sympathetic nerve activity (SNA), arterial blood pressure (BP) and heart rate (HR). Cocaine, 4 mg/kg i.v., reduced SNA by 55 +/- 6%, but did not significantly affect BP or HR. Cocaine, in doses that were ineffective by the i.v. route, was administered into the vertebral artery and produced decreases in SNA, BP and HR in anesthetized cats. Administration of cocaine into the carotid artery was without effect. Topical administration of cocaine to the intermediate area of the ventrolateral medullary surface (2

The Vestibular System: A Newly Identified Regulator of Bone Homeostasis Acting Through the Sympathetic Nervous System.

The vestibular system is a small bilateral structure located in the inner ear, known as the organ of balance and spatial orientation. It senses head orientation and motion, as well as body motion in the three dimensions of our environment. It is also involved in non-motor functions such as postural control of blood pressure. These regulations are mediated via anatomical projections from vestibular nuclei to brainstem autonomic centers and are involved in the maintenance of cardiovascular function via sympathetic nerves. Age-associated dysfunction of the vestibular organ contributes to an incre

Timing of Prenatal Stress Exposure Predicts Infant Sympathetic Nervous System and Affective Responses.

Prenatal stress has broad detrimental consequences for neurodevelopment, with potential sensitive periods within gestation affecting specific developmental systems. We examined the effects of prenatal stress timing, level, and fluctuations on three markers of sympathetic nervous system activity: infant salivary alpha amylase (sAA), fear, and anger responses. In addition, we explored whether the effects of prenatal stress differed for boys and girls. We assessed 195 mother-infant dyads (45% girls) from an ethnically diverse and economically disadvantaged community sample. Women reported perceiv…

Metabolic control of ovarian function through the sympathetic nervous system: role of leptin.

The link between metabolism and reproduction is well-known. Both undernutrition and obesity affect the reproductive system. Metabolic status … these hormones are leptin, insulin, and GLP-1 that act directly in the hypothalamus to activate the sympathetic nervous system. In this minireview, we propose that leptin could be an important regulator of sympathetic innervation in reproductive tissues. Leptin may affect the density or activity of sympathetic nerves, thereby affecting reproductive function. We also specula …

Effects of prenatal opioid exposure on infant sympathetic and parasympathetic nervous system activity.

Prenatal opioid exposure has been associated with developmental problems, including autonomic nervous system dysregulation. However, little is known about the effects of prenatal opioid exposure on the autonomic nervous system beyond the first days of life, particularly across both the parasympathetic and sympathetic branches, and when accounting for exposure to other substances. The present study examined the effects of prenatal exposure to opioid agonist therapy (OAT, e.g., methadone) and other opioids on infant autonomic nervous system activity at rest and in response to a social stressor (

The Sympathetic Nervous System Contributes to the Establishment of Pre-Metastatic Pulmonary Microenvironments.

Emerging evidence suggests that neural activity contributes to tumor initiation and its acquisition of metastatic properties. More specifically, it has been reported that the sympathetic nervous system regulates tumor angiogenesis, tumor growth, and metastasis. The function of the sympathetic nervous system in primary tumors has been gradually elucidated. However, its functions in pre-metastatic environments and/or the preparation of metastatic environments far from the primary sites are still unknown. To investigate the role of the sympathetic nervous system in pre-metastatic environments, we…

The sympathetic nervous system regulates skeletal muscle motor innervation and acetylcholine receptor stability.

AIM: Symptoms of autonomic failure are frequently the presentation of advanced age and neurodegenerative diseases that impair adaptation to common physiologic stressors. The aim of this work was to examine the interaction between the sympathetic and motor nervous system, the involvement of the sympathetic nervous system (SNS) in neuromuscular junction (NMJ) presynaptic motor function, the stability of postsynaptic molecular organization, and the skeletal muscle composition and function. METHODS: Since muscle weakness is a symptom of diseases characterized by autonomic dysfunction, we studied …
