Structurally tuned drug release from hybrid manufactured tablets via overprinting and overmolding.

Hybrid manufacturing, which integrates three-dimensional printing and injection molding, presents a scalable solution for producing personalized oral dosage forms. This study presents a systematic comparison between overprinting and overmolding, two hybrid manufacturing strategies, to assess their influence on tablet microstructure, drug release dynamics, and processing performance. Using a Taguchi L9 experimental design, formulations containing polycaprolactone, poly(vinylpyrrolidone-co-vinyl acetate), and theophylline (5-30% w/w) were processed via hot-melt extrusion and fabricated into bila

3D-printing of lipid formulations: Going slow in a fast-paced field.

Lipid-based excipients (LBE) are a crucial class of functional excipients with significant potential for pharmaceutical formulation and product development. The wide variety of molecules in this class provides opportunities for designing novel dosage forms. However, the utilization of LBEs for 3D-printing applications is currently limited, thereby hindering the full potential of lipids and impeding the advancement of 3D-printing technologies. Notably, only a few publications report the 3D-printing of lipid-based formulations. This review provides a detailed discussion and analysis of these pub

Optimization of the manufacturing process of a complex amphotericin B liposomal formulation using quality by design approach.

In this work, the manufacturing process of a complex liposomal amphotericin B (AmB) product was optimized using quality by design (QbD) approach. A comprehensive QbD-based process understanding and design space (DS) to the critical process parameters (CPPs) is essential to the drug development and consistent quality control. The process was based on the acid-aided formation of drug-lipid complexes in a methanol-chloroform mixture (step I) followed by spray drying (step II), hydration and liposome formation by microfluidization (step III), and lyophilization (step IV). Firstly, the risk assessm

Ultrasonic evaluation of permanent moisture impact on microstructural and micro-viscoelastic properties of pharmaceutical tablets: a proof of concept study.

Moisture exposure can significantly affect the critical quality attributes of pharmaceutical tablets by altering their internal microstructure and micro-viscoelastic properties. In compressed granular materials, moisture-induced changes in chemical state, interparticle bonding, porosity, and residual stress distribution can alter both mechanical integrity and performance-related attributes. In this study, irreversible moisture-induced changes in tablet bulk, microstructural, and micro-viscoelastic properties were investigated and quantified using ultrasonics. As a result of uniform vapor diffu

Navigating translational research in nanomedicine: A strategic guide to formulation and manufacturing.

Over the past two decades, extensive research has focused on both the fundamental and applied aspects of nanomedicine, driven by the compelling advantages that nanoparticles offer over their bulk counterparts. Despite this intensive research effort, fewer than 100 nanomedicines have been approved by the U.S. Food and Drug Administration and the European Medicines Agency since 1989. This disparity highlights a substantial gap in translational research, reflecting the disconnect between the prolific research in nanomedicine and the limited number of products that successfully reach and sustain t

Non-aqueous formulations for ram and screen extrusion-spheronisation.

The use of non-aqueous cellulose-based formulations for extrusion-spheronisation (E-S) is investigated. A 10 wt% hydroxypropyl cellulose/isopropyl alcohol solution (HPC/IPA) was identified as a suitable sticky liquid binder for preparing non-aqueous pastes. Preliminary tests were performed on a series of pastes using a ram as well as a laboratory roller screen extruder, since the former is commonly used in batch testing and the latter replicates the shear range in a manufacturing screen extruder. Pellets with acceptable size and shape distributions were obtained with Avicel® HFE-102 NF/HPC/IPA

Holistic Considerations in Optimizing a Sterile Product Package to Ensure Container Closure Integrity.

A new major chapter dealing with container closure integrity was released by the United States Pharmacopeial Convention. Chapter <1207> provides a significant amount of education and guidance concerning test methodologies to prove that a system is integral and safe for use. The test method used is only one of the major considerations in approaching the challenge of proving an integral system. This paper takes a holistic review of all the major considerations needed in qualifying a new vial system for container closure integrity. There is substantial interplay among many aspects in the process

Formulation of paracetamol-containing pastilles with in situ coating technology.

The focus of this research was to apply the in situ coating technology for producing paracetamol- (PCT-) containing pastilles for paediatric use from a eutectic of two sugar alcohols (sorbitol, xylitol) in one step. This type of melt-technology is more cost-efficient and simpler than other conventional tableting technologies, whereby the formation of the pastilles and their coating occur upon the same fabrication step. We managed to produce pastilles having a softer core and a harder, resistant shell in one cooling step. Adding polyethylene glycol (PEG) 2000 or 6000 to the PCT-containing eutec

[Early achievements of the Danish pharmaceutical industry--8. Lundbeck].

The article series provides a written and pictorial account of the Danish pharmaceutical industry's products from their introduction until about 1950. Part 8 deals with products from Lundbeck. Lundbeck which today is known as a considerable international pharmaceutical company could in 2015 celebrate its 100 years' jubilee. Among the early Danish medicinal companies H. Lundbeck & Co. is in many ways an exception as the company was not originally established as a pharmaceutical company. Not until several years after the foundation the company began to import foreign ready-made medicinal product

PCA-CR analysis of dissolution profiles. A chemometric approach to probe the polymorphic form of the active pharmaceutical ingredient in a drug product.

A simple chemometric approach to differentiate among the three crystalline polymorphs of the model drug Furosemide (FUR) in a pharmaceutical dosage form is presented. The proposed method is based on the principal component analysis with confidence regions (PCA-CR) comparison of the dissolution profiles of the test pharmaceutical formulation, and formulations containing the different polymorphs, employed as the corresponding references. For the elaboration of the references, FUR polymorphs I, II and III were prepared, characterized and compounded with the excipients found in the test commercial

Advanced Pharmaceutical Applications of Hot-Melt Extrusion Coupled with Fused Deposition Modelling (FDM) 3D Printing for Personalised Drug Delivery.

Three-dimensional printing, also known as additive manufacturing, is a fabrication process whereby a 3D object is created layer-by-layer by depositing a feedstock material such as thermoplastic polymer. The 3D printing technology has been widely used for rapid prototyping and its interest as a fabrication method has grown significantly across many disciplines. The most common 3D printing technology is called the Fused Deposition Modelling (FDM) which utilises thermoplastic filaments as a starting material, then extrudes the material in sequential layers above its melting temperature to create

Three-dimensional printing as a transformative platform for patient-centric pediatric drug delivery: technologies, formulation strategies and clinical translation.

BACKGROUND: Pediatric patients require age-appropriate dosage forms because developmental differences in physiology, pharmacokinetics and swallowing ability can significantly affect therapeutic outcomes. Conventional dosage forms are often unsuitable for children and practices such as tablet splitting, crushing or extemporaneous preparation may lead to dose inaccuracy, altered stability, poor palatability and reduced adherence. These limitations highlight the need for innovative formulation strategies that provide precise dosing while improving acceptability and therapeutic effectiveness. In t
