The emergence of glucagon-like peptide-1 agonists represents a notable advancement in the pharmacological treatment of obesity, yet complementary approaches are essential. Through phenotypic drug discovery, we developed promising nitroalkene-containing small molecules for obesity-related metabolic dysfunctions. Here, we present SANA, a nitroalkene derivative of salicylate, demonstrating notable efficacy in preclinical models of diet-induced obesity. SANA reduces liver steatosis and insulin resistance by enhancing mitochondrial respiration and increasing creatine-dependent energy expenditure in

Radix aconiti carmichaeli is a widely used traditional Chinese medicine that has been found to be effective in treating cardiovascular diseases and metabolic disorders. Patients with these diseases often experience a heat generation disorder, which is characterized by chilliness and can worsen the progression of the disease. This study established an in vitro screening model combining the examination of cellular mitochondrial membrane potential and mitochondrial temperature to screen drugs with thermogenic activity. After differentiation and determination of the content of characteristic metab

Currently, considerable attention is focused on exploring the potential relationship between herbal medicine (HM) and the gut microbiome in terms of thermoregulation, which is an important aspect of human health, in modern system biology. However, our knowledge of the mechanisms of HM in thermoregulation is inadequate. Here, we demonstrate that the canonical herbal formula, Yijung-tang (YJT), protects against hypothermia, hyperinflammation, and intestinal microbiota dysbiosis in PTU-induced hypothyroid rats. Notably, these properties were associated with alterations in the gut microbiota and s

Iodotyrosine deiodinase (IYD) is a type of deiodinase enzyme that scavenges iodide from the thyroid gland. Previously, we showed that H3 Ab acts as an agonist on IYD to induce migration of cells to the heart and differentiate human stem cells into brown adipocyte-like cells. To continue this study, we investigated the dual function of IYD in hypothyroidism by blocking IYD and in thermogenesis by looking at the induction of brown adipocyte-like cells by treatment with H3 Ab in a mouse model. Surprisingly, our results suggest H3 Ab acts on IYD as both an antagonist and agonist to reduce T4 and i

Overweight and obesity result from an imbalance between caloric intake and energy expenditure, including expenditure from spontaneous physical activity (SPA). Changes in SPA and resulting changes in non-exercise activity thermogenesis (NEAT) likely interact with diet to influence risk for obesity. However, previous research on the relationship between diet, physical activity, and energy expenditure has been mixed. The neuropeptide orexin is a driver of SPA, and orexin neuron activity can be manipulated using DREADDs (Designer Receptors Exclusively Activated by Designer Drugs). We hypothesized

As with other forms of psychological stress, conditioned fear causes an increase in body temperature. The mechanisms underlying this stress-induced hyperthermia are not well understood, but previous research suggests that nonshivering thermogenesis might contribute, as it does during cold exposure. The major source of nonshivering thermogenesis in the rat is brown adipose tissue (BAT), and the largest BAT deposit in that species is in the interscapular area just below the skin. BAT is also under sympathetic control via beta-adrenoceptors. If BAT contributes to fear-induced hyperthermia, then t

The uniqueness of UCP1 (as compared to UCP2/UCP3) is evident from expression analysis and ablation studies. UCP1 expression is positively correlated with metabolic inefficiency, being increased by cold acclimation (in adults or perinatally) and overfeeding, and reduced in fasting and genetic obesity. Such a simple relationship is not observable for UCP2/UCP3. Studies with UCP1-ablated animals substantiate the unique role of UCP1: the phenomenon of adaptive adrenergic non-shivering thermogenesis in the intact animal is fully dependent on the presence of UCP1, and so is any kind of cold acclimat

The effects of essential fatty acid (EFA) deficiency on energetic metabolism and interscapular brown adipose tissue (BAT) activity were examined in the cold acclimated rat. Weanling male Long-Evans rats were fed on a low fat semipurified diet (control diet, 2% sunflower oil; EFA deficient diet, 2% hydrogenated coconut oil) for 9 weeks. They were exposed at 5 degrees C for the last 5 weeks. In EFA deficient rats, compared to controls, growth retardation reached 22% at sacrifice. Caloric intake being the same in the two groups, it follows that food efficiency was decreased by 40%. Resting metabo

Several thermogenic pathways have been described in adipose tissue, including calcium-mediated thermogenesis in beige adipocytes. We have previously shown that adipocyte-specific deletion of the RNA binding protein human antigen R (HuR; Elavl1) results in thermogenic dysfunction. RNA sequencing revealed the downregulation of several calcium transport genes upon HuR deletion. The goal of this work was to define the HuR-dependent mechanisms of calcium driven thermogenesis in brown adipocytes. Brown adipose tissue (BAT)-specific HuR-deletion (BAT-HuR-/-) mice have no changes in their body weight,

Adipose tissue metabolic dysfunction plays a central role in the development of obesity and related metabolic disease. Luteolin is a natural flavonoid with antioxidant, anti-inflammatory, antidiabetic, and antiobesity activities, while it is limited by poor solubility, bioavailability, and stability. This study developed a whey protein isolate (WPI)-Pueraria lobata amylopectin (PLA) composite gel, characterized by a dense 3D network and enhanced mechanical strength. The WPI-PLA gel efficiently encapsulated luteolin in a stable amorphous form and exhibited uniform particle size and excellent co

BACKGROUND: Antibiotic-induced disruption of the gut microbiota is known to influence host metabolism; however, whether its metabolic effects depend on the duration of exposure remains unclear, particularly in relation to adipose thermogenesis under high-fat diet (HFD) conditions.
METHODS: C57BL/6 mice fed an HFD were treated with a broad-spectrum antibiotic cocktail for either short-term (2 weeks) or long-term (10 weeks). Body weight, adiposity, metabolic parameters, and energy expenditure were assessed. Thermogenic capacity was evaluated by histology and gene expression analysis in brown an

UNLABELLED: Zinc and ring finger protein 3 (ZNRF3) functions as a transmembrane E3 ubiquitin ligase. Although genome-wide association studies have implicated ZNRF3 in body fat distribution, the evidence from human genetics and animal models has been lacking. Here, through whole-exome sequencing of 1,944 obese individuals and 2,161 healthy lean control individuals, we identified a rare loss-of-function mutation in human ZNRF3 (p.V228L) observed exclusively in 8 obese individuals. These carriers exhibit increased hip circumference and subcutaneous fat area and lower fasting glucose levels compar

This study examined the relationship between human brown adipose tissue (BAT) activity and thermogenesis in response to topical menthol application, which activates the transient receptor potential melastatin-8 in individuals with various BAT activity. Thirteen healthy male participants were separated into high (n = 6) and low (n = 7) BAT groups based on BAT activity (SUVmax). They were placed in a supine position at 27°C for 120 min with 4% l-menthol or a control solution applied to their abdomen and lower limbs. Expired gas was measured to calculate energy expenditure (EE) and substrate oxid

Although hepatocyte-released adenosine triphosphate (ATP) plays important roles in maintaining metabolic homeostasis, how its hydrolyzation outside hepatocytes impacts on glucolipid metabolism remains unclear. The authors aim to identify the enzyme(s) that hydrolyzes hepatocyte-secreted ATP to regulate metabolic homeostasis. All known ATP-hydrolyzing enzymes are expressed with the highest expression of ectonucleoside triphosphate diphosphohydrolase 5 (ENTPD5) in hepatocytes. ENTPD5 expression is reduced in steatotic mouse and human livers. Hepatic ENTPD5 overexpression ameliorates the deregula

OBJECTIVE: G-protein-signaling modulator 1 (GPSM1) has been proved the potential role in brain tissues, however, whether GPSM1 in hypothalamic nuclei, especially in POMC neurons is essential for the proper regulation of whole-body energy balance remains unknown. The aim of our current study was to explore the role of GPSM1 in POMC neurons in metabolic homeostasis.
METHODS: We generated POMC neuron specific GPSM1 deficiency mice and subjected them to a High Fat Diet to monitor metabolic phenotypes in vivo. By using various molecular, biochemical, immunofluorescent, immunohistochemical analyses

Stress elicits a variety of autonomic responses, including hyperthermia (stress fever) in humans and animals. In this present study, we investigated the circuit basis for thermogenesis and heat conservation during this response. We first demonstrated the glutamatergic identity of the dorsal hypothalamic area (DHAVglut2) neurons that innervate the raphe pallidus nucleus (RPa) to regulate core temperature (Tc) and mediate stress-induced hyperthermia. Then, using chemogenetic and optogenetic methods to manipulate this hypothalamomedullary circuit, we found that activation of DHAVglut2 neurons pot

We have tested the hypothesis that there is a role for the cerebral cortex in the control of non-shivering thermogenesis during fever induced by prostaglandin E1 (PGE1). While under urethan anesthesia, the firing rate of nerves innervating interscapular brown adipose tissue (IBAT), IBAT and colonic temperatures (TIBAT and Tc) and oxygen (O2) consumption were monitored during the fever from PGE1 injection (400 and 800 ng) in a lateral cerebral ventricle in controls and in functionally decorticated Sprague-Dawley rats. Rats were functionally decorticated by applying 3.3 M KCl solution on the fro

The role of brown adipose tissue in total energy balance and cold-induced thermogenesis was studied. Mice expressing mitochondrial uncoupling protein 1 (UCP-1) from the fat-specific aP2 gene promoter (heterozygous and homozygous aP2-Ucp transgenic mice) and their nontransgenic C57BL6/J littermates were used. The transgenic animals are resistant to obesity induced by a high-fat diet, presumably due to ectopic synthesis of UCP-1 in white fat. These animals exhibited atrophy of brown adipose tissue, as indicated by smaller size of brown fat and reduction of its total UCP-1 and DNA contents. Norep

Caffeine produces enhanced oxygen consumption, an effect that may reflect an action of caffeine on brown adipose thermogenesis. In Experiment 1, adult male rats were anesthetized with 1.2 g/kg urethane and treated (IP) with either 0.9% saline or 10, 20 or 40 mg/kg caffeine (n = 4 each group). Interscapular BAT (IBAT) and rectal temperatures were recorded every minute for 10 minutes prior to and 30 minutes following drug injection. Stable IBAT and rectal temperatures were observed prior to and after saline injection whereas rats treated with 20 and 40 mg/kg caffeine exhibited moderate increases

The gut microbiota is increasingly recognized as a key regulator of host metabolic adaptation to environmental challenges. However, whether breed-specific gut microbiota contributes to variation in heat tolerance among poultry breeds remains unclear. This study used Cyan-shank partridge chickens, a native Chinese breed with superior environmental adaptability, to investigate the role of the gut microbiota in heat tolerance and its underlying mechanisms. Cyan-shank partridge chickens exhibited a higher inflection point temperature of rectal temperature and greater tolerance to heat stress than
