Analysis of high temperature stress on the dynamics of antenna size and reducing side heterogeneity of Photosystem II in wheat leaves (Triticum aestivum).

This study demonstrates the effect of high temperature stress on the heterogeneous behavior of PSII in Wheat (Triticum aestivum) leaves. Photosystem II in green plant chloroplasts displays heterogeneity both in the composition of its light harvesting antenna i.e. on the basis of antenna size (α, β and γ centers) and in the ability to reduce the plastoquinone pool i.e. the reducing side of the reaction centers (Q(B)-reducing centers and Q(B)-non-reducing centers). Detached wheat leaves were subjected to high temperature stress of 35°C, 40°C and 45°C. The chlorophyll a (Chl a) fluorescence trans…

Genome-Wide Identification and Expression Analysis of TaDES1 Gene Family Responded to Biotic and Abiotic Stress in Wheat (Triticum aestivum L.).

L-Cysteine desulfhydrase (DES1), a key enzyme in eukaryotes, catalyzes the synthesis of hydrogen sulfide (H2S), which is a gaseous signaling molecule. However, the genes encoding DES1 enzymes in wheat, one of the world's most important crop species, have yet to be fully characterized. This study offers a comprehensive analysis of the wheat TaDES1 gene family and delves into its potential functions. We identified a total of 18 TaDES1 genes, located across 13 chromosomes, categorized them into four subfamilies, and comprehensively analyzed their physicochemical properties. Furthermore, three DES

High Resolution Genome Wide Association Studies Reveal Rich Genetic Architectures of Grain Zinc and Iron in Common Wheat (Triticum aestivum L.).

Biofortification is a sustainable strategy to alleviate micronutrient deficiency in humans. It is necessary to improve grain zinc (GZnC) and iron concentrations (GFeC) in wheat based on genetic knowledge. However, the precise dissection of the genetic architecture underlying GZnC and GFeC remains challenging. In this study, high-resolution genome-wide association studies were conducted for GZnC and GFeC by three different models using 166 wheat cultivars and 373,106 polymorphic markers from the wheat 660K and 90K single nucleotide polymorphism (SNP) arrays. Totally, 25 and 16 stable loci were

Molecular characterization of the celiac disease epitope domains in α-gliadin genes in Aegilops tauschii and hexaploid wheats (Triticum aestivum L.).

Nineteen novel full-ORF α-gliadin genes and 32 pseudogenes containing at least one stop codon were cloned and sequenced from three Aegilops tauschii accessions (T15, T43 and T26) and two bread wheat cultivars (Gaocheng 8901 and Zhongyou 9507). Analysis of three typical α-gliadin genes (Gli-At4, Gli-G1 and Gli-Z4) revealed some InDels and a considerable number of SNPs among them. Most of the pseudogenes were resulted from C to T change, leading to the generation of TAG or TAA in-frame stop codon. The putative proteins of both Gli-At3 and Gli-Z7 genes contained an extra cysteine residue in the u

An early auxin-responsive Aux/IAA gene from wheat (Triticum aestivum) is induced by epibrassinolide and differentially regulated by light and calcium.

The plant hormone auxin plays a central role in regulating many aspects of plant growth and development. This largely occurs as a consequence of changes in gene expression. The Aux/IAA genes are best characterized among the early auxin-responsive genes, which encode short-lived transcriptional repressors. In most plants examined, including Arabidopsis, soybean, and rice, the Aux/IAA genes constitute a large gene family. By screening the available databases, at least 15 expressed sequence tags (ESTs) have been identified from wheat (Triticum aestivum), which exhibit high sequence identity with …

Temporal transcriptional networks coordinate ABA-GA-JA crosstalk during cold-induced modulation of seed dormancy and germination in wheat (Triticum aestivum L.).

Using a combination of genome-wide transcriptome, targeted hormonome and pharmacological approaches, this study showed that temporally resolved transcriptional regulatory networks integrating hormonal crosstalk underlie cold-induced modulation of seed dormancy and germination in wheat. Gene homoeologues (GHs) differentially expressed between cold-treated and untreated dormant seeds imbibed at optimal temperature (OT, 22°C) as well as between the same seed samples imbibed in cold and at OT represent distinct biological processes (BPs), indicating the role of coordinated activation/repression sp

Foliar GR24 and γ-aminobutyric acid modulate redox signaling, root-system architecture, and mineral homeostasis in tellurite-stressed wheat (Triticum aestivum L.).

Tellurite (TeO₃2-) is a phytotoxic tellurium (Te) species that disrupts cellular redox homeostasis, mineral nutrition, and root development. However, the signaling events underlying tellurite toxicity and its mitigation in cereals remain poorly understood. This study examined whether foliar application of GR24, a synthetic strigolactone analog, and γ-aminobutyric acid (GABA), individually or jointly, modulates early root signaling and downstream physiological acclimation in wheat (Triticum aestivum L.) exposed to tellurite. A 2 × 4 factorial experiment was conducted using two tellurite conditi…

Taurine modulates dynamics of oxidative defense, secondary metabolism, and nutrient relation to mitigate boron and chromium toxicity in Triticum aestivum L. plants.

The present study was undertaken to appraise the efficacy of exogenous taurine in alleviating boron (B) and chromium (Cr) toxicity. Taurine protects cell membranes from lipid peroxidation due to its function as a ROS scavenger. However, there exists no report in the literature on the role of taurine in plants under abiotic stresses. The present investigation indicated the involvement of exogenous taurine in mediating plant defense responses under B and Cr toxicity. Wheat plants manifested a significant drop in growth, chlorophyll molecules, SPAD values, relative water content, nitrate reductas

QTL Mapping Using a High-Density Genetic Map to Identify Candidate Genes Associated With Metribuzin Tolerance in Hexaploid Wheat (Triticum aestivum L.).

Tolerance to metribuzin, a broad-spectrum herbicide, is an important trait for weed control in wheat breeding. However, the genetics of metribuzin tolerance in relation to the underlying quantitative trait loci (QTL) and genes is limited. This study developed F8 recombinant inbred lines (RILs) from a cross between a highly resistant genotype (Chuan Mai 25) and highly susceptible genotype (Ritchie), which were used for QTL mapping of metribuzin tolerance. Genotyping was done using a diversity arrays technology sequencing (DArTseq) platform, and phenotyping was done in controlled environments. H

Preliminary mapping of wheat (Triticum aestivum L.) tolerance genes to the English grain aphid (Sitobion avenae Fabricius) by genome-wide association study.

Six Sitobion avenae-tolerant wheat accessions, mapped 110 associated SNPs and six candidate genes were identified, providing valuable genetic resources for breeding wheat with tolerance to S. avenae. Wheat tolerance to the English grain aphid (Sitobion avenae) is rarely incorporated into integrated pest management strategies for wheat fields. The scarcity of tolerant accession and insufficient mapping of tolerance-related gene are key limiting factors. To address these gaps, 640 wheat accessions were evaluated for S. avenae tolerance, combined with genome-wide association study (GWAS) and qPCR
