Actionable genotypes beyond the coding sequence and their association with lifespan in the UK Biobank.

BACKGROUND: The identification and reporting of actionable genotypes in the American College of Medical Genetics and Genomics (ACMG)-recommended secondary findings (SF) genes represent a key preventive strategy for hereditary diseases. Robust evidence quantifying their association with lifespan remains limited, particularly for non-coding regulatory variants, due to functional interpretation challenges. METHOD: Using data from the UK Biobank, we investigated associations between genetic variants and all-cause mortality in 490 086 participants with whole-genome sequencing. We systematically in…

Microplastic Pollution and Risk of Cancer: A Prospective Study Based on UK Biobank.

IntroductionVarious industrial pollutants have been implicated in tumorigenesis. However, the cancer risk associated with microplastics, a pervasive environmental pollutant, remains poorly understood. This study aimed to evaluate the association between basin-level environmental microplastic contamination and cancer incidence.MethodsIn this secondary analysis of a prospective UK Biobank cohort, 378,985 participants were linked to basin-level river microplastic measurements using residential coordinates. The 2019 measurements were treated as an ecological proxy for regional environmental microp…

Coffee and Parkinson's disease: associations by CYP1A2 genotype and sex in UK Biobank.

Coffee consumption has been associated with Parkinson's disease (PD), but findings remain inconsistent. We examined whether this association differs by CYP1A2 genotype and sex in 435,551 UK Biobank participants free of PD at baseline. During a median follow-up of 15.7 years, 3319 incident PD cases were identified. In overall analyses, coffee intake was not associated with PD risk. However, coffee intake significantly interacted with CYP1A2 genotype. Among AA carriers, coffee intake below 5 cups/day was associated with lower PD risk (HR 0.83, 95% CI 0.74-0.93), and spline analyses indicated a n…

Supine Hypertension as a Predictor of Atrial Fibrillation: A UK Biobank Cohort Study.

BACKGROUND: Atrial fibrillation (AF) is a common arrhythmia strongly associated with blood pressure (BP), yet most current assessments focus only on seated BP measurements. Supine BP may offer additional predictive value by capturing positional or autonomic variations often missed in standard evaluations. OBJECTIVES: This study aimed to investigate the association between supine BP and incident AF. METHODS: We analyzed 24,901 UK Biobank participants without prior cardiovascular disease or antihypertensive treatment, categorizing them into 4 groups based on the presence of seated and/or supin…

The Alpha-1 Antitrypsin Z-allele associates with sarcopenia in the UK Biobank.

BACKGROUND: Sarcopenia is common in chronic obstructive pulmonary disease (COPD) and contributes to adverse outcomes. Alpha-1 antitrypsin deficiency (AATD) is the most common genetic cause of COPD, but its impact on skeletal muscle remains to be determined. METHODS: We performed a retrospective analysis of 487,207 participants in the UK Biobank with available SERPINA1 genotyping. Associations between AAT genotypes and measures of skeletal muscle mass were determined using multivariable linear and logistic regression models. Models adjusted for age, sex, smoking status, genetic principal compo…

Delirium and adverse clinical outcomes: a matched cohort study in the UK Biobank.

BACKGROUND: Delirium complicates up to one in four hospitalisations among older adults, but its clinical sequelae beyond cognitive impairment, functional decline, and mortality are not well characterised. Whether delirium signals broader multisystem vulnerability leading to later adverse outcomes remains uncertain. We aimed to examine the association of in-hospital delirium with the occurrence of a range of adverse outcomes during later hospitalisations in the UK. METHODS: We performed a matched cohort study among hospitalised UK Biobank participants recruited in England, Scotland, and Wales …

eGDR and gastrointestinal cancer mortality in the UK Biobank: a prospective cohort study.

BACKGROUND: This study examined the associations of estimated glucose disposal rate (eGDR), a surrogate of insulin sensitivity, and the insulin resistance indices TG/HDL-C and TyG with gastrointestinal (GI) cancer mortality in the UK Biobank. METHODS: We included 369,447 participants without cancer at baseline and recorded 4,305 GI cancer-related deaths over a mean follow-up of 13.5 years. Fine-Gray models assessed associations of eGDR, TG/HDL-C, and TyG with GI cancer mortality. RESULTS: Higher eGDR was associated with lower pooled GI cancer mortality (sHR, 0.67; 95% CI, 0.61-0.74; P < 0.00…

Association of multimorbidity and disease clusters with neuroimaging and cognitive outcomes in UK Biobank.

BACKGROUND: The relationship between multimorbidity, particularly disease clusters, with neuroimaging and cognitive outcomes that typically manifest prior to clinical diagnosis of dementia, remains understudied. This study investigated whether multimorbidity is associated with dementia-related neuroimaging and cognitive outcomes in the UK Biobank cohort. METHODS: This cross-sectional study used data from UK Biobank participants who attended imaging assessments between 2014-2023, and were free from neurological conditions, including dementia. Multimorbidity was defined as the coexistence of tw…

Associations of adherence to a healthy sleep pattern with the dementia risk in the UK biobank.

BACKGROUND: Existing evidence highlights associations between sleep behaviors and dementia risk; however, the impact of adhering to a healthy sleep pattern on dementia risk remains unclear. METHODS: Of 406,364 UK Biobank participants aged 40-64, we excluded those who had withdrawn, had incomplete sleep data, or had dementia at baseline, yielding a final sample of 333,014. Participants were enrolled between 2006 and 2010, with follow-up extending from recruitment to dementia diagnosis, death, loss to follow-up, or the censoring date (December 2022), whichever came first. Incident dementia was …

Association of Lipoprotein(a) and Interleukin-6 With Cardiovascular Risk: MESA and UK Biobank.

BACKGROUND: Elevated lipoprotein(a) [Lp(a)] is associated with atherosclerotic cardiovascular disease (ASCVD) risk, and vascular inflammation is one mechanism through which Lp(a) causes ASCVD. OBJECTIVES: The authors aimed to evaluate whether interleukin-6 (IL-6), a biomarker associated with inflammation and cardiovascular disease, helps risk-stratify individuals with elevated Lp(a). METHODS: Data from participants in the MESA (Multi-Ethnic Study of Atherosclerosis) (n = 6,514) and the UK Biobank (UKB) (n = 26,574) were used for this analysis. The associations between Lp(a) and IL-6 with cor…

Association of social isolation and loneliness with cancer prognosis: an analysis of the UK Biobank.

BACKGROUND: Evidence is scarce for the association between social isolation and loneliness and prognosis of any cancer or other common cancer types. METHODS: To address this gap, we performed a cohort study from 2006 to 2022 using data from the UK Biobank (UKB). Social isolation and loneliness were identified based on the questionnaire of the UKB. Cases of death were obtained from the death registry, where both primary and contributory causes were recorded using ICD-10 codes. The Cox proportional hazard models were used to estimate hazard ratios (HRs) and associated 95% confidence intervals (…

Metabolic Syndrome and the Risk of Ductal Carcinoma In Situ of the Breast in the UK Biobank.

BACKGROUND: Metabolic syndrome (MetS), defined by the presence of three of more metabolic dysregulations such as hyperlipidemia, hyperinsulinemia, central obesity, and hypertension, has been associated with increased risk of cardiovascular disease, diabetes, and various cancers, including invasive breast cancer (IBC). Whether MetS is a risk factor for ductal carcinoma in situ of the breast (DCIS), a nonobligate precursor of IBC, remains unknown. METHODS: A total of 198,748 women ages 40 to 69 years, DCIS- and IBC-free at enrolment in UK Biobank, were included in the current study. Multivariab…

Hippocampal volumes in UK Biobank are associated with APOE only in older adults.

INTRODUCTION: The hippocampus atrophies with age and is implicated in neurodegenerative disorders including Alzheimer's disease (AD). We examined the interplay between age and apolipoprotein E (APOE) genotype on total hippocampal volume. METHODS: Using neuroimaging data from 37,463 UK Biobank participants, we applied linear regression to quantify the association of age and APOE with hippocampal volume and identified the age when volumes of ε2/ε3, ε3/ε4, and ε4/ε4 carriers significantly deviated from ε3/ε3 using generalized additive modeling. RESULTS: Total hippocampal volume declined with ag…

Asthma exacerbations and eosinophilia in the UK Biobank: a genome-wide association study.

BACKGROUND: Asthma exacerbations reflect disease severity, affect morbidity and mortality, and may lead to declining lung function. Inflammatory endotypes (e.g. T2-high (eosinophilic)) may play a key role in asthma exacerbations. We aimed to assess whether genetic susceptibility underlies asthma exacerbation risk and additionally tested for an interaction between genetic variants and eosinophilia on exacerbation risk. METHODS: UK Biobank data were used to perform a genome-wide association study of individuals with asthma and at least one exacerbation compared to individuals with asthma and no…

Exploring the genetic correlation of cardiovascular diseases and mood disorders in the UK Biobank.

AIMS: Cardiovascular diseases (CVDs) are the leading cause of deaths globally. Mortality and incidence of CVDs are significantly higher in people with mood disorders. About 81.1% of CVD patients were reported with comorbidities in 2019, where the second most common comorbidity was due to major depressive disorder (MDD). This study, therefore, aimed to evaluate the genetic correlation between CVDs and mood disorders by using data from the UK Biobank towards understanding the influence of genetic factors on the comorbidity due to CVDs and mood disorders. METHODS: The UK Biobank database provide…
