A.actinomycetemcomitans-induced periodontal disease promotes systemic and local responses in rat periodontium.

AIM: To characterize the histologic and cellular response to A. actinomycetemcomitans (Aa) infection. MATERIAL & METHODS: Wistar rats infected with Aa were evaluated for antibody response, oral Aa colonization, loss of attachment, PMN recruitment, TNF-α in the junctional epithelium and connective tissue, osteoclasts and adaptive immune response in local lymph nodes at baseline and 4, 5 or 6 weeks after infection. Some groups were given antibacterial treatment at 4 weeks. RESULTS: An antibody response against Aa occurred within 4 weeks of infection, and 78% of inoculated rats had detectable A

The in vivo expression of the collagenolytic matrix metalloproteinases (MMP-2, -8, -13, and -14) and matrilysin (MMP-7) in adult and localized juvenile periodontitis.

Periodontal inflammation is characterized by irreversible degradation of periodontal ligament collagen fibers leading to loss of tooth attachment. Cultured gingival keratinocytes and fibroblasts express, in vitro, various matrix metalloproteinases (MMPs) which can degrade fibrillar collagens. We hypothesized that several MMPs are also synthesized in vivo by sulcular epithelium, and analyzed the collagenolytic MMPs (MMP-2, -8, -13, and -14) and matrilysin (MMP-7) in gingival tissue specimens and gingival crevicular fluid from adult and localized juvenile periodontitis patients by in situ hybrid

Specific primer for AP-PCR identification of Actinobacillus actinomycetemcomitans.

We used arbitrarily-primed polymerase chain reaction (AP-PCR) to design and construct a specific primer pair for the identification of Actinobacillus actinomycetemcomitans. We analyzed 25 DNA samples of A. actinomycetemcomitans isolated from patients with localized-juvenile periodontitis. From 90 AP-PCR primers screened, one amplification product was selected, cloned in pCR II vector, and sequenced. The sequence was used to design a single pair of specific primers. The sequence was compared with GenBank entries using BLAST and showed no significant matches. PCR amplification using the new prim

Subantimicrobial dose doxycycline efficacy as a matrix metalloproteinase inhibitor in chronic periodontitis patients is enhanced when combined with a non-steroidal anti-inflammatory drug.

BACKGROUND: Administration of subantimicrobial dose doxycycline (SDD) to chronic periodontitis (CP) patients has repeatedly been found to reduce mammalian collagenase and other matrix metalloproteinase (MMP) activity in gingival tissues and crevicular fluid, in association with clinical efficacy, without the emergence of antibiotic-resistant bacteria either orally or extra-orally. More recently, SDD adjunctive to repeated mechanical debridement resulted in dramatic clinical improvement in patients (>50% smokers) with generalized aggressive periodontitis. As an additional pharmacologic approach…

Extracellular matrix of ameloblastoma-derived negatively regulates osteogenic differentiation.

OBJECTIVE: To investigate the effects of key pathogenic genes involved in the development of jaw ameloblastoma (AB) and its associated extracellular matrix (ECM) on osteogenic differentiation in order to provide a theoretical foundation for future research into bone aggressiveness of AB. METHODS: The essential genes were identified by five AB patients for whole-exome sequencing and the microarray datasets GES38494 and GES132472. Moreover, the expression of key genes and their encoded proteins in AB tissues was explored. In addition, AB-derived the decellularized ECM (ABdECM) tissues were gene

The Medical, Clinical, and Radiographic Aspects of Multiple Idiopathic Tooth Resorption: A Systematic Review.

BACKGROUND: Many causes of resorption remain unclear and are thus identified as idiopathic. In such cases, management is difficult, especially when multiple teeth are involved. The aim of the present study was to assess the literature regarding the medical, clinical, and radiographic aspects of multiple idiopathic resorptions (MIR) and to examine the factors associated with the risk of extraction. METHODS: The title and protocol were registered a priori in PROSPERO (CRD42020191564), and the study followed the PRISMA methodology. Four electronic databases were searched to include reviews and c

Maxillary sporadic Burkitt's lymphoma associated with neuro-orbital involvement in an Indian male.

Burkitt's lymphoma (BL) is the fastest growing malignancy of the lymphoreticular system to affect humans and has a potential ability to double in size every day. A case of maxillary sporadic BL (sBL) associated with neuro-orbital involvement in an Indian male is presented. sBL initially presented as maxillary swelling with no obvious dental and periodontal changes. Histological specimen from incisional biopsy revealed a round cell malignant tumor and immunohistochemistry reactions favored nonHodgkin's lymphoma consistent with BL. Four weeks later, patient presented with orbital involvement as

Sequence diversity in the major fimbrial subunit gene (flp-1) of Actinobacillus actinomycetemcomitans.

Cells of the periodontal pathogen Actinobacillus actinomycetemcomitans exhibit tight adherence to surfaces such as glass, plastic and hydroxyapatite, a property that probably plays an important role in the ability of this bacterium to colonize teeth and other surfaces. Tight adherence is mediated by long fibrils of bundled pili (fimbriae) that form on the surface of the cell. The flp-1 gene encodes the major pilin protein component of A. actinomycetemcomitans fimbriae. In this study we compared flp-1 DNA sequences from 43 strains of A. actinomycetemcomitans isolated in Europe, Japan and the Un

Molecular basis of leukocyte adhesion molecules in early-onset periodontitis patients with decreased CD11/CD18 expression on leukocytes.

We analyzed the cell-cell adherence related to CD11/CD18 and CD18 mRNA in individuals with decreased CD11/CD18 expression on their neutrophil surface. Epstein Barr virus-transformed B cell lines were developed from one localized juvenile periodontitis (LJP) patient with decreased CD11/CD18 in the peripheral blood neutrophils and without systemic diseases; two siblings with generalized prepubertal periodontitis (GPP) caused by leukocyte adhesion deficiency (LAD); another LJP patient; one localized prepubertal periodontitis (LPP) patient; and two healthy subjects. Rothia aeria Causing Prosthetic Valve Endocarditis in an Immunocompetent Patient.

Rothia aeria is a part of the normal human oral flora and respiratory tract. This Gram-positive, pleomorphic rod has been linked to periodontal infections, typically occurring within immunocompromised hosts. To our knowledge, we present the first case of prosthetic valve endocarditis (PVE) secondary to R. aeria in an immunocompetent host. A 76-year-old man with a history of aortic valve replacement presented with generalized weakness and dyspnea on exertion eight weeks after deep dental cleaning for periodontal disease. He improved and was discharged from the hospital. Subsequently, blood cult

A test of the Lamendin method of age estimation in South African canines.

Age estimation in unknown adult skeletons remains a considerable problem in forensic anthropology. In 1992, Lamendin et al. published a non-destructive method of age estimation on single rooted teeth. With this method, periodontosis and root transparency are judged against root height, and these are then used in regression formulae to estimate age. The aim of this study was to test the accuracy of the Lamendin method on a large sample of canines of South Africans, and if necessary to adapt the formulae for this population. A sample of known sex, age and population group was used. This included

Localisation of a gene for prepubertal periodontitis to chromosome 11q14 and identification of a cathepsin C gene mutation.

Prepubertal periodontitis (PPP) is a rare and rapidly progressive disease of young children that results in destruction of the periodontal support of the primary dentition. The condition may occur as part of a recognised syndrome or may occur as an isolated finding. Both autosomal dominant and recessive forms of Mendelian transmission have been reported for PPP. We report a consanguineous Jordanian family with four members affected by PPP in two nuclear sibships. The parents of the affected subjects are first cousins. We have localised a gene of major effect for PPP in this kindred (Zmax=3.55

Kaposiform hemangioendothelioma in the mandibular gingiva: a rare case report with literature review.

Kaposiform hemangioendothelioma (KHE) is a locally aggressive vascular tumor in neonates and children. KHE can occur anywhere in the body, but occurrence in the oral cavity is exceptionally rare. The clinical features of KHE are similar to those of oral lesions, such as infantine hemangioma, pyogenic granuloma, and chronic periodontitis. However, unlike other oral lesions, KHE invades neighboring tissue planes and recurs frequently as malignancy. Moreover, some instances of KHE are accompanied by a life-threatening condition named Kasabach-Merritt phenomenon. Correct diagnosis of KHE is very d

C-1 esterase inhibitor dysfunction localised to the periodontal tissues: clues to the role of stress in the pathogenesis of chronic periodontitis?

BACKGROUND: C1-esterase inhibitor (C1eIn) is an important modulator of complement activation via the classical pathway. Deficiencies or dysfunction involving this inhibitor underlie the condition of angioneurotic oedema. AIM: The purpose of this report is to describe a female patient who presented at the age of 24 years with an apparently aggressive form of periodontitis and severe oedema, localised to the free gingival tissues. After 21 years of repeated surgical reduction of the gingiva, a diagnosis of C1eIn dysfunction was made. METHODS: Exhaustive serological investigations were performe

A versatile bacterial cell wall-based nanomedicine for combination treatment of oral squamous cell carcinoma.

Oral squamous cell carcinoma (OSCC), an aggressive malignancy characterized by high recurrence and metastasis rates, continues to pose a significant clinical challenge to human health. Bacterial immunotherapy has emerged as a promising strategy for cancer treatment by activating multimodal immune responses. Porphyromonas gingivalis (Pg), an oral pathogenic bacterium strongly associated with periodontitis, can produce abundant μ-oxo bisheme on its cell wall (CW) to avoid the oxidative damage induced by H2O2 during inflammatory processes. Owing to the presence of μ-oxo bisheme, the CW extracted

Impact of interventional periodontal therapy on the longevity of failing dental implants: A retrospective analysis.

BACKGROUND: Biological complications such as peri-implantitis present a significant challenge to the long-term stability of dental implants, often leading to failure if left untreated. While a history of periodontitis is a known risk factor, there is a lack of real-world data quantifying the "survival extension" provided by therapeutic interventions. This study aimed to evaluate the efficacy of surgical and non-surgical "rescue" therapies in extending the lifespan of dental implants that ultimately failed. MATERIAL AND METHODS: A retrospective cohort analysis was conducted using the multi-ins

Saliva from subjects harboring Actinobacillus actinomycetemcomitans kills Streptococcus mutans in vitro.

BACKGROUND: Previous research indicated that patients with localized aggressive periodontitis (LAgP) had minimal proximal decay. We speculated that differences in these two proximal dental diseases (LAgP and proximal decay) in LAgP could be due to the effect of saliva on the growth of key microorganisms related to these two infections. Carbon dioxide (CO(2)) is required for growth of Actinobacillus actinomycetemcomitans (Aa), the reputed cause of LAgP. Bicarbonate, a source of CO(2), buffers acid production by Streptococcus mutans (Sm), a key organism associated with caries. The purpose of thi…
