Liver cirrhosis is a critical stage in the progression of various chronic liver diseases, often leading to severe complications such as ascites, hepatic encephalopathy, and a high mortality rate, and it thus poses a serious threat to patient life. The activation of hepatic stellate cells is a central driver of disease progression. Cellular autophagy, a lysosome-mediated degradation process, plays a key role in maintaining cellular function and dynamic homeostasis. Research has shown that autophagy is closely associated with proteins like LC3, Beclin-1, P62, and mTOR, and is regulated through s…

INTRODUCTION: Podocytopathies are a uniquely renal disease syndrome, in which direct or indirect podocyte injury leads to proteinuria or nephrotic syndrome. Of the many factors that contribute to podocytopathies, the abnormal regulation of autophagy, such insufficient or excessive autophagy levels, have been proposed to play a significant role in the occurrence and development of podocytopathies. However, there still has been a lack of systematic and comparative research to elucidate exact role of autophagy in podocytopathies and its current research status. This study aims to utilize bibliome…

Acute lung injury (ALI) poses a significant clinical challenge due to its high morbidity and mortality rates. Current treatment options are limited in their efficacy, necessitating the exploration of novel therapeutic targets. The mammalian target of rapamycin (mTOR), a crucial regulator of various cellular processes, has been implicated in the pathogenesis of ALI. Autophagy, a tightly regulated cellular degradation process controlled by mTOR, plays a pivotal role in the pathogenesis of ALI and cellular homeostasis. Mounting evidence also suggests that the mTOR pathway and autophagy play cruci…

Osteoporosis (OP) is a serious public health issue, and fragility fractures resulting from OP are a major cause of death in the elderly. Osteoblast (OB)-mediated insufficient bone formation and osteoclast (OCs)mediated abnormalities in bone destruction can result in OP. Autophagy is the process by which cells degrade and recycle their own proteins and organelles. The differentiation and activity of OBs, OCs, and other bone cells depend on autophagy activity. The regulation of autophagy has the potential to influence the metabolic processes of these cells, which may contribute to the treatment …

Emerging research has confirmed the crucial role of autophagy, an endogenous repair mechanism, in various blast injuries. However, its role in explosive ocular injury (EOI) under microgravity (MG) and normal gravity (NG) environments remains poorly understood. Therefore, this study aimed to investigate the changes in retinal lesions and retinal autophagy over time following EOI under both NG and MG environments. This study employed the hind-limb unloading model in Sprague-Dawley (SD) rats to simulate MG conditions and used self-made device with compressed gas to induce EOI. SD rats were random…

BACKGROUND: Osteoporosis (OP) is a globally prevalent metabolic bone disorder, particularly affecting aging populations, and is characterized by an imbalance between bone resorption and formation. The pathogenesis of OP involves a complex interplay of genetic predisposition, hormonal changes, and chronic inflammatory pathways. Emerging evidence now indicates that pyroptosis, an inflammasome-mediated form of programmed cell death, and autophagy, a conserved intracellular catabolic process, are critical regulators of osteocyte viability and bone metabolism homeostasis.
METHODS: A systematic lit…

PURPOSE: Patients with meningiomas mostly present good outcomes and optimal prognosis, but different grades of tumors have very different symptoms and recurrence rates. Therefore, effective diagnosis is crucial for early intervention and controlling tumor development. Circadian cycle and autophagy have both been proven to be related to neoplasm formation and pathogenesis; however, there is limited exploration and discussion on the relationships between the circadian cycle and autophagy in patients with meningiomas. This study was undertaken to elucidate the relationship between two autophagy m…

Impaired autophagy and increased cellular senescence in the aging epididymis may lead to cellular stress, inflammation, and epithelial dysfunction. This reduction in autophagy can be identified using light and electron microscopy, as well as molecular biology techniques. Autophagy, which may be selective or non-selective, is tightly regulated by genes such as LC3 and LAMP-2. Decreased autophagic activity in the epididymis results in reduced sperm quality and male fertility. These findings highlight autophagy as a potential therapeutic target in age-related male reproductive decline.

Pituitary adenomas (PAs) are tumors originating from the anterior lobe of the pituitary gland. Although most are benign, some may exhibit invasive behavior. While surgical treatment is effective in controlling most PAs, it carries inherent risks of complications, some of which may be severe. Consequently, managing PAs remains a significant clinical challenge. Autophagy, a cellular recycling process, plays a pivotal role in tumor biology and is frequently activated in response to stimuli such as nutrient deprivation, hypoxia, or mitochondrial dysfunction. Autophagy's role in tumors is complex, …

Macroautophagy/autophagy-mediated anoikis resistance is crucial for tumor metastasis. As a key autophagy-related protein, ATG4B has been demonstrated to be a prospective anti-tumor target. However, the existing ATG4B inhibitors are still far from clinical application, especially for tumor metastasis. In this study, we identified a novel circRNA, circSPECC1, that interacted with ATG4B. CircSPECC1 facilitated liquid-liquid phase separation of ATG4B, which boosted the ubiquitination and degradation of ATG4B in gastric cancer (GC) cells. Thus, pharmacological addition of circSPECC1 may serve as an…

This review systematically examines the influence of oxidative stress on the reproductive function of male livestock, with a particular focus on the modulation of autophagy. Spermatogenesis, a highly precise biological process, is vulnerable to a range of internal and external factors, among which oxidative stress notably disrupts autophagic processes within the testes. This disruption results in diminished sperm quality, impaired testosterone synthesis, and compromised integrity of the blood-testis barrier. Furthermore, this review elucidates the molecular mechanisms by which oxidative stress…

Autophagy is an essential degradative process that governs the renewal of organelle and maintains the homeostasis of cellular microenvironment. Its dysregulation has been demonstrated to be an indicator for neuroinflammation. To elucidate the interrelationship between neuroinflammation and autophagy, optical probes are ideal tools as they offer a number of advantages such as high spatiotemporal resolution and non-invasive sensing, which help to visualize the physiological and pathological functions of interested analytes. However, single autophagy parameter-response probes may generate false-p…

The developmental activation of the corpus luteum (CL) structurally and functionally is critical for the temporally regulated establishment, maintenance, and termination of pregnancy in rats. In this study, we have investigated the possible involvement of autophagy in the regulation of the CL during pregnancy in rats. The expression ratio of microtubule-associated protein light chain 3 (LC3)-II/-I, a widely used indicator of autophagic activity, in the CL remained relatively stable until day 15 of pregnancy. Subsequently, it progressively increased until day 21, and then declined until day 3 p…

Atg9, the only transmembrane protein among many autophagy-related proteins, was first identified in the year 2000 in yeast. Two homologs of Atg9, ATG9A and ATG9B, have been found in mammals. While ATG9B shows a tissue-specific expression pattern, such as in the placenta and pituitary gland, ATG9A is ubiquitously expressed. Additionally, ATG9A deficiency leads to severe defects not only at the molecular and cellular levels but also at the organismal level, suggesting key and fundamental roles for ATG9A. The subcellular localization of ATG9A on small vesicles and its functional relevance to auto…

Proper protein degradation is required for cellular protein homeostasis and organ function. Particularly, in post-mitotic cells, such as cardiomyocytes, unbalanced proteolysis due to inflammatory stimuli and oxidative stress contributes to organ dysfunction. To ensure appropriate protein turnover, eukaryotic cells exert two main degradation systems, the ubiquitin-proteasome-system and the autophagy-lysosome-pathway. It has been shown that proteasome activity affects the development of cardiac dysfunction differently, depending on the type of heart failure. Studies analyzing the inducible subty…
