Medical Applications of Porous Biomaterials: Features of Porosity and Tissue-Specific Implications for Biocompatibility.

Porosity is an important material feature commonly employed in implants and tissue scaffolds. The presence of material voids permits the infiltration of cells, mechanical compliance, and outward diffusion of pharmaceutical agents. Various studies have confirmed that porosity indeed promotes favorable tissue responses, including minimal fibrous encapsulation during the foreign body reaction (FBR). However, increased biofilm formation and calcification is also described to arise due to biomaterial porosity. Additionally, the relevance of host responses like the FBR, infection, calcification, and

Foreign body response to synthetic polymer biomaterials and the role of adaptive immunity.

Implanted biomaterials elicit a series of distinct immune and repair-like responses that are collectively known as the foreign body reaction (FBR). These include processes involving innate immune inflammatory cells and wound repair cells that contribute to the encapsulation of biomaterials with a dense collagenous and largely avascular capsule. Numerous studies have shown that the early phase is dominated by macrophages that fuse to form foreign body giant cells that are considered a hallmark of the FBR. With the advent of more precise cell characterization techniques, specific macrophage subs

Biomaterial and biocompatibility evaluation of tunicate nanocellulose for tissue engineering.

Extracellular matrix fibril components, such as collagen, are crucial for the structural properties of several tissues and organs. Tunicate-derived cellulose nanofibrils (TNC) combined with living cells could become the next gold standard for cartilage and soft-tissue repair, as TNC fibrils present similar dimensions to collagen, feasible industrial production, and chemically straightforward and cost-efficient extraction procedures. In this study, we characterized the physical properties of TNC derived from aquaculture production in Norwegian fjords and evaluated its biocompatibility regarding

Improvement of Interfacial Properties and Bioactivity of CF/PEEK Composites by Rapid Biomineralization of Hydroxyapatite.

The carbon fiber/hydroxyapatite (CF/HA) reinforcement with a three-dimensional flower-like structure was rapidly fabricated with the biomimetic mineralization method to strengthen the interfacial performance and bioactivity of carbon fiber/polyether ether ketone (CF/PEEK) composites. Silk fibroin (SF), which can construct a robust interphase between CF and PEEK, supported an organic template to regulate the growth of inorganic HA. The interfacial properties were markedly enhanced through mechanical interlocking and interface adhesion induced by increasing roughness and surface energy of CF. Be

Lysozyme Amyloid Fibril-Integrated PEG Injectable Hydrogel Adhesive with Improved Antiswelling and Antibacterial Capabilities.

Hydrogels with inherent antibacterial activities have been attracting increasing attention, particularly for biomedical applications. Biology provides a range of materials and mechanisms to meet diverse requirements for bacterial combating. Lysozyme after fibrillation (LZMF) has a much superior antibacterial ability than globular native lysozyme due to its decreased positive charges and increased hydrophobic β-sheet component. Here, we propose to design a poly(ethylene glycol) (PEG) cross-linked LZMF composite antibacterial hydrogel by utilizing the nucleophilic substitution reaction between L

Multiscale stiffness characterisation of both healthy and osteoporotic bone tissue using subject-specific data.

Severe bone fractures are often treated by appending internal fixations. In unhealthy or osteoporotic patients, post-implantation bone fractures can occur due to external impact (e.g. from a fall), day-to-day activities in highly-osteoporotic cases and mismatches in the stiffness of bone and the implant's biomaterial, since this causes stress concentrations. One approach to alleviating this problem is to use biomaterials that closely mimic the effective stiffness of real bone, thereby more seamlessly integrating the fixation. This requires to know the properties target (bone properties) and th

Could Curdlan/Whey Protein Isolate/Hydroxyapatite Biomaterials Be Considered as Promising Bone Scaffolds?-Fabrication, Characterization, and Evaluation of Cytocompatibility towards Osteoblast Cells In Vitro.

The number of bone fractures and cracks requiring surgical interventions increases every year; hence, there is a huge need to develop new potential bone scaffolds for bone regeneration. The goal of this study was to gain knowledge about the basic properties of novel curdlan/whey protein isolate/hydroxyapatite biomaterials in the context of their use in bone tissue engineering. The purpose of this research was also to determine whether the concentration of whey protein isolate in scaffolds has an influence on their properties. Thus, two biomaterials differing in the concentration of whey protei

Electrical field stimulated modulation of cell fate of pre-osteoblasts on PVDF/BT/MWCNT based electroactive biomaterials.

The present study reports the impact of the interplay between electroactive properties of the biomaterials and electrical stimulation (ES) toward the cell proliferation, migration and maturation of osteoprogenitors (preosteoblasts; MC3T3-E1) on the electroactive poly (vinylidene difluoride) (PVDF)-based composites. The barium titanate (BaTiO3; BT; 30 wt%) and multiwalled carbon nanotubes (MWCNT; 3 wt%) were introduced into the PVDF via melt mixing, which led to an enhancement of the dielectric permittivity, electrical conductivity, and surface roughness. We also present the design and developm

Treatment Effect of Platelet Gel on Reconstructing Bone Defects and Nonunions: A Review of In Vivo Human Studies.

In ideal circumstances, a fractured bone can heal properly by itself or with the aid of clinical interventions. However, around 5% to 10% of bone fractures fail to heal properly within the expected time even with the aid of clinical interventions, resulting in nonunions. Platelet gel is a blood-derived biomaterial used in regenerative medicine aiming to promote wound healing and regeneration of damaged tissues. The purpose of this paper is to review relevant articles in an attempt to explore the current consensus on the treatment effect of platelet gel on reconstructing bone defects and nonuni

Electroactive Biomaterials for Facilitating Bone Defect Repair under Pathological Conditions.

Bone degeneration associated with various diseases is increasing due to rapid aging, sedentary lifestyles, and unhealthy diets. Living bone tissue has bioelectric properties critical to bone remodeling, and bone degeneration under various pathological conditions results in significant changes to these bioelectric properties. There is growing interest in utilizing biomimetic electroactive biomaterials that recapitulate the natural electrophysiological microenvironment of healthy bone tissue to promote bone repair. This review first summarizes the etiology of degenerative bone conditions associa

Development of an Intervertebral Disc for Cervical Spondylosis Composed of Seeded Biomaterials.

Most of the current artificial disc prosthesis presented a restricted range of motion. Here we propose the design of a novel intervertebral disc composed of carbon fiber, hyaluronic methylcellulose hydrogel loaded with mesenchymal stem cells and polycaprolactone. The prosthesis was biomechanically evaluated under two static physiological conditions to study the mechanical influence of the material on the device. The results obtained in the simulations showed a not only a congruent behavior with preclinical condition, but also that the proposed materials met the desired biomechanical properties

Precisely Tuning the Pore-Wall Surface Composition of Bioceramic Scaffolds Facilitates Angiogenesis and Orbital Bone Defect Repair.

Orbital bone damage (OBD) may result in severe post-traumatic enophthalmos, craniomaxillofacial deformities, vision loss, and intracranial infections. However, it is still a challenge to fabricate advanced biomaterials that can match the individual anatomical structure and enhance OBD repair in situ. Herein, we aimed to develop a selective surface modification strategy on bioceramic scaffolds and evaluated the effects of inorganic or organic functional coating on angiogenesis and osteogenesis, ectopically and orthotopically in OBD models. It was shown that the low thermal bioactive glass (BG)

Behaviour at the peritoneal interface of next-generation prosthetic materials for hernia repair.

BACKGROUND: When using a prosthetic material in hernia repair, the behaviour of the mesh at the peritoneal interface is especially important for implant success. Biomaterials developed for their intraperitoneal placement are known as composites and are made up of two different-structure materials, one is responsible for good integration within host tissue and the other is responsible to make contact with the viscera. This study examines the behaviour at the peritoneal level of two composites, the fully degradable Phasix-ST® and the partially degradable Symbotex®. A polypropylene mesh (Optilene
