The use of low dose methotrexate in rheumatoid arthritis - are we entering a new era of therapeutic drug monitoring and pharmacogenomics?

Methotrexate (MTX) is one of the most commonly used medications in the treatment of rheumatoid arthritis (RA). It has proven efficacy as a sole agent as well as in combination with other disease modifying anti-rheumatic agents (DMARDs) including the newer biological agents. MTX is generally well tolerated although there are a number of potentially serious adverse effects. Of these, haematopoietic suppression, hepatotoxicity and pulmonary toxicity are the more severe and patients are therefore required to have appropriate monitoring while they remain on MTX. In the past, attempts at therapeutic

Distinct cellular and therapeutic effects of obatoclax in rituximab-sensitive and -resistant lymphomas.

Bcl-2 proteins represent a rheostat that controls cellular viability. Obatoclax, a BH3-mimetic, has been designed to specifically target and counteract anti-apoptotic Bcl-2 proteins. We evaluated the biological effects of obatoclax on the anti-tumour activity of rituximab and chemotherapy agents. Obatoclax induced cell death of rituximab/chemotherapy-sensitive (RSCL), -resistant cell lines (RRCL) and primary tumour-cells derived from patients with B-cell lymphomas (N=39). Obatoclax also enhanced the activity of rituximab and had synergistic activity when combined with chemotherapy agents. The

Machine learning approaches for predicting antibiotic resistance genes abundance changes during biological nitrogen removal process.

Wastewater treatment plants (WWTPs) serve as reservoirs for multiple antimicrobial agents (AAs), thereby promoting the risk of antibiotic resistance genes (ARGs) transmission in sewage and sludge during biological nitrogen removal (BNR) processes. And the fate of ARGs is challenging to be deciphered due to the influence of multiple factors and microbial interactions. This study employed four machine learning (ML) models for predicting changes in the abundance of ARGs and mobile genetic elements (MGEs) during BNR processes. Categorical Boosting (CatBoost) model achieved the highest prediction a

Autophagy in health and disease: From molecular mechanisms to therapeutic target.

Macroautophagy/autophagy is an evolutionally conserved catabolic process in which cytosolic contents, such as aggregated proteins, dysfunctional organelle, or invading pathogens, are sequestered by the double-membrane structure termed autophagosome and delivered to lysosome for degradation. Over the past two decades, autophagy has been extensively studied, from the molecular mechanisms, biological functions, implications in various human diseases, to development of autophagy-related therapeutics. This review will focus on the latest development of autophagy research, covering molecular mechani

Current role of short-term intensive insulin strategies in newly diagnosed type 2 diabetes.

Type 2 diabetes mellitus (T2DM) is a progressive disease characterized by worsening insulin resistance and a decline in β-cell function. Achieving good glycemic control becomes more challenging as β-cell function continues to deteriorate throughout the disease process. The traditional management paradigm emphasizes a stepwise approach, and insulin has generally been reserved as a final armament. However, mounting evidence indicates that short-term intensive insulin therapy used in the early stages of type 2 diabetes could improve β-cell function, resulting in better glucose control and more ex

Lipopeptide biodiversity in antifungal Bacillus strains isolated from Algeria.

Several Bacillus strains have been well studied for their ability to control soil-borne plant diseases. This property is linked to the production of several families of lipopeptides. Depending of their structure, these compounds show antifungal and/or plant systemic resistance inducing activities. In this work, the biodiversity of lipopeptides produced by different antifungal Bacillus strains isolated from seeds, rhizospheric, and non-rhizospheric soils in Algeria was analyzed. Sixteen active strains were characterized by PCR for their content in genes involved in lipopeptide biosynthesis and

Molecular mechanisms and clinical implications of Wnt pathway inhibition.

The dysregulation of the evolutionarily conserved Wnt signaling pathway is a fundamental factor in the pathogenesis of numerous diseases. This review consolidates current knowledge regarding the therapeutic targeting of Wnt signaling inhibitors. Endogenous inhibitors exert multi-layered regulatory control by mechanisms such as extracellular ligand sequestration, receptor antagonism, cytoplasmic β-catenin degradation, and nuclear transcriptional repression, thereby ensuring precise regulation of the pathway. Dysfunction in these regulatory mechanisms often results in aberrant Wnt activation and

The relative brightness of photoelectron microscopy images as a function of exposure to UV light has been determined from model systems representative of biological cell surface components. Quantitative data for amino acid homopolymers, yields. The photoelectron quantum yields, increase substantially over the initial values. For example, the quantum yields fo poly-L-tyrosine at 200 nm is initially about 5 X 10(-8) electron/incident photon. The quantum yield increases with 254 nm irradiation, leveling off at about 5 X 10(-4) electrons/incident photon after a dose of 3 X 10(21) quanta cm-2. Pre-
