Measuring human mast cell-mediated cytotoxicity against human tumour cells: three-colour flow cytometry using monoclonal antibody target staining with annexin V / propidium iodide co-labelling of death.

BACKGROUND: Chromium release assay is the standard method for evaluation of cell-mediated cytotoxicity, including that of mast cells. Although this is a reproducible method, it has more drawbacks than even radioactivity. In addition to the shortcoming of measuring only necrotic killing, some non-radioactive methods have not been widely used either. The numerous limitations of these methods led researchers to develop other techniques. This study describes a new flow cytometric approach that measures human mast cell-mediated cytotoxicity by marking target cells with monoclonal antibody alongside

Chemical chaperones protect from effects of apoptosis-inducing mutation in carbonic anhydrase IV identified in retinitis pigmentosa 17.

Carbonic anhydrase (CA) IV is a glycosylphosphotidylinositol-anchored enzyme highly expressed on the plasma face of microcapillaries and especially strongly expressed in the choriocapillaris of the human eye. In collaboration with scientists at the University of Cape Town (Rondebosch, South Africa), we recently showed that the R14W mutation in the signal sequence of CA IV, which they identified in patients with the retinitis pigmentosa (RP) 17 form of autosomal dominant RP, results in accumulation of unfolded protein in the endoplasmic reticulum (ER), leading to ER stress, the unfolded protein

A tape-based intestinal chip co-culture model for assessing anti-inflammatory activity of chitosan oligosaccharides.

Intestinal inflammation is a central pathological process in gastrointestinal diseases, yet conventional in vitro models poorly reproduce epithelial-immune interactions, and accessible platforms for rapid anti-inflammatory screening remain lacking. Here, a low-cost, high-throughput tape-based intestinal chip co-culturing Caco-2 epithelial cells and RAW264.7 macrophages under dynamic flow was applied to approximate the epithelial-macrophage interface of the intestinal wall and to evaluate the anti-inflammatory activity of chitosan oligosaccharides (COS). Three COS fractions (COS1, COS2, COS3; 3

Long-term culture of patient-derived cardiac organoids recapitulated Duchenne muscular dystrophy cardiomyopathy and disease progression.

Duchenne Muscular Dystrophy (DMD) is an X-linked neuromuscular disease which to date is incurable. The major cause of death is dilated cardiomyopathy however, its pathogenesis is unclear as existing cellular and animal models do not fully recapitulate the human disease phenotypes. In this study, we generated cardiac organoids from patient-derived induced pluripotent stem cells (DMD-COs) and isogenic-corrected controls (DMD-Iso-COs) and studied if DMD-related cardiomyopathy and disease progression occur in the organoids upon long-term culture (up to 93 days). Histological analysis showed that D

Examining the NEUROG2 lineage and associated gene expression in human cortical organoids.

Proneural genes are conserved drivers of neurogenesis across the animal kingdom. How their functions have adapted to guide human-specific neurodevelopmental features is poorly understood. Here, we mined transcriptomic data from human fetal cortices and generated from human embryonic stem cell-derived cortical organoids (COs) to show that NEUROG1 and NEUROG2 are most highly expressed in basal neural progenitor cells, with pseudotime trajectory analyses indicating that NEUROG1-derived lineages predominate early and NEUROG2 lineages later. Using ChIP-qPCR, gene silencing and overexpression studie

Impaired T3 uptake and action in MCT8-deficient cerebral organoids underlie Allan-Herndon-Dudley syndrome.

Patients with mutations in the thyroid hormone (TH) cell transporter monocarboxylate transporter 8 (MCT8) gene develop severe neuropsychomotor retardation known as Allan-Herndon-Dudley syndrome (AHDS). It is assumed that this is caused by a reduction in TH signaling in the developing brain during both intrauterine and postnatal developmental stages, and treatment remains understandably challenging. Given species differences in brain TH transporters and the limitations of studies in mice, we generated cerebral organoids (COs) using human induced pluripotent stem cells (iPSCs) from MCT8-deficien

Chitooligosaccharides Improves Severe Acute Pancreatitis by Reducing Intestinal Mucosal Injury and Regulating Intestinal Microbiota.

Severe acute pancreatitis (SAP), a widespread inflammatory condition impacting the abdomen with persistent organ dysfunction (> 48 h) and a high mortality rate, poses challenges due to its unclear pathogenesis and the absence of effective treatment options. Chitooligosaccharide (COS), a naturally occurring alkaline oligosaccharide, demonstrates robust anti-inflammatory, antioxidant, and regulating intestinal microbiota properties. However, the specific protective impact of COS on SAP remains to be fully elucidated. Maintaining intestinal microecological balance provides an effective method for

The Control of the Crossover Localization in Allium.

Meiotic crossovers/chiasmata are not randomly distributed and strictly controlled. The mechanisms behind crossover (CO) patterning remain largely unknown. In Allium cepa, as in the vast majority of plants and animals, COs predominantly occur in the distal 2/3 of the chromosome arm, while in Allium fistulosum they are strictly localized in the proximal region. We investigated the factors that may contribute to the pattern of COs in A. cepa, A. fistulosum and their F1 diploid (2n = 2x = 8C + 8F) and F1 triploid (2n = 3x = 16F + 8C) hybrids. The genome structure of F1 hybrids was confirmed using

Inhibition of Mitochondrial Bioenergetics by Esterase-Triggered COS/H2S Donors.

Hydrogen sulfide (H2S) is an important biological mediator, and synthetic H2S donating molecules provide an important class of investigative tools for H2S research. Here, we report esterase-activated H2S donors that function by first releasing carbonyl sulfide (COS), which is rapidly converted to H2S by the ubiquitous enzyme carbonic anhydrase (CA). We report the synthesis, self-immolative decomposition, and H2S release profiles of the developed scaffolds. In addition, the developed esterase-triggered COS/H2S donors exhibit higher levels of cytotoxicity than equivalent levels of Na2S or the co

Ca2+ uptake and release properties of a thapsigargin-insensitive nonmitochondrial Ca2+ store in A7r5 and 16HBE14o- cells.

In a previous study we overexpressed the thapsigargin (tg)-insensitive Pmr1 Ca(2+) pump of the Golgi apparatus of Caenorhabditis elegans in COS-1 cells and studied the properties of the Ca(2+) store into which it was integrated. Here we assessed the properties of an endogenous tg-insensitive nonmitochondrial Ca(2+) store in A7r5 and 16HBE14o- cells, which express a mammalian homologue of Pmr1. The tg-insensitive Ca(2+) store was considerably less leaky for Ca(2+) than the sarco(endo)plasmic-reticulum Ca(2+)-ATPase (SERCA)-containing Ca(2+) store. Moreover like for the worm Pmr1 Ca(2+) pump exp

Cross-scale genetic analysis prioritizes shared regulatory variation, cell-type contributions, co-expression programs, and exon-level patterns in atopic dermatitis and asthma.

Atopic dermatitis (AD) and asthma frequently co-occur, yet the regulatory mechanisms connecting cutaneous and airway inflammation remain unclear. We integrated European genome-wide association statistics for AD (22,474 cases; 774,187 controls) and asthma (56,167 cases; 352,255 controls) with expression and splicing quantitative trait loci (QTLs), immune single-cell expression QTLs, single-cell transcriptomes, and spatial transcriptomics. The AD dataset compared lesional skin with healthy controls, whereas the asthma dataset paired allergen and diluent challenges in four participants. Each was

The Combination of Methionine Adenosyltransferase 2A (MAT2A) Inhibitor AG-270 and Recombinant Methioninase Is Not Cancer-selective in a Co-culture Model of Colon Cancer Cells and Normal Fibroblasts.

BACKGROUND/AIM: Methionine addiction is a fundamental and general hallmark of cancer cells. Recombinant methioninase (rMETase) degrades extracellular methionine. rMETase, or other means of restricting methionine, in combination with numerous types of chemotherapy have shown synergistic cancer-selective efficacy. AG-270, a methionine adenosyltransferase 2A (MAT2A) inhibitor, blocks intracellular conversion of methionine to S-adenosylmethionine (SAM), the central reaction of the methionine cycle. The present study aimed to evaluate the synergistic and cancer-selective efficacy of the combination

Diffuse co-expression of TTF-1 and p40 (ΔNp63) defines a distinct biphenotypic non-small cell lung carcinoma: clinicopathologic and molecular characterization of 19 cases.

BACKGROUND: The diffuse co-expression of thyroid transcription factor-1 (TTF-1) and p40 defines a rare and diagnostically challenging subtype of non-small cell lung carcinoma (NSCLC). Due to its extreme rarity, the clinicopathologic and molecular characteristics of this entity remain poorly characterized. METHODS: We conducted a comprehensive analysis of the largest single-institution cohort to date (n = 19) of NSCLCs with diffuse TTF-1/p40 co-expression. All cases underwent detailed histopathologic review. Immunohistochemistry (IHC) utilized specific antibody clones (TTF-1: 8G7G3/1; p40). Mol

Oncolytic adenovirus co-expressing IL-12 and shVEGF potentiates immune checkpoint blockade for treatment of renal cell carcinoma: in vitro, organ-on-a-chip, and in vivo evaluation.

Renal cell carcinoma (RCC) has been known as highly malignant and resistant to standard therapy due to its immunosuppressive tumor microenvironment and aberrant VEGF-mediated angiogenesis. To overcome these therapeutic limitations, here we assessed the therapeutic efficacy of an oncolytic adenovirus (oAd) co-expressing Interleukin-12 (IL-12) and a short hairpin RNA against VEGF (shVEGF) in combination with immune checkpoint inhibitors (ICIs). The oAd/IL12/shVEGF induced RCC-specific cancer cell death in vitro and 3D bioprinted model, leading to greater anti-tumor effect compared with oAd-GMCSF

Daphnetin Attenuates Inflammatory and Apoptotic Responses Induced by High Glucose/High Free Fatty Acids and Lipopolysaccharide in an HK-2 Cell-Neutrophil Co-Culture System.

OBJECTIVES: The present study aimed to investigate the protective effect of daphnetin (dap) against high glucose and high free fatty acids (HGHF) and lipopolysaccharide(LPS)-induced injury in the aforementioned co-culture system under normal control conditions (normal glucose and no exogenous FFA supplementation), and to explore its potential molecular mechanisms. METHODS: Neutrophils were isolated from the peripheral blood of healthy individuals. Subsequently, HK-2 cells and a neutrophil suspension were co-cultured at 1x105 cells/mL to establish the experimental model. The non-cytotoxic conc

Methanol-Enhanced Low-Cell-Voltage Hydrogen Generation at Industrial-Grade Current Density by Triadic Active Sites of Pt1-Pdn-(Ni,Co)(OH)x.

Methanol (ME) is a liquid hydrogen carrier, ideal for on-site-on-demand H2 generation, avoiding its costly and risky distribution issues, but this "ME-to-H2" electric conversion suffers from high voltage (energy consumption) and competitive oxygen evolution reaction. Herein, we demonstrate that a synergistic cofunctional Pt1Pdn/(Ni,Co)(OH)x catalyst with Pt single atoms (Pt1) and Pd nanoclusters (Pdn) anchored on OH-vacancy(VOH)-rich (Ni,Co)(OH)x nanoparticles create synergistic triadic active sites, allowing for methanol-enhanced low-voltage H2 generation. For MOR, OH* is preferentially adsor

Analysis of Microarray and Single-Cell RNA-Seq Finds Gene Co-Expression and Tumor Environment Associated with Extracellular Matrix in Epithelial-Mesenchymal Transition in Prostate Cancer.

A complex and gradual process, the epithelial-mesenchymal transition (EMT) occurs both during embryonic development and tumor progression. Cells undergo a transition from an epithelial to a mesenchymal state throughout this process. More and more evidence points to EMT as a cause of increased metastatic spread of prostate cancer (PCa), along with stemness enhancement and therapy resistance. Here, we used bioinformatic methods to analyze gene expression microarray data, single-cell RNA sequencing, oncogenes, and tumor suppressor genes (TSGs) in order to reconstruct the network of differentially

Coupling of the galvanic cell and Z-scheme effects of zinc/gallium dual co-catalysts to enhance the photocatalytic conversion of carbon dioxide using water as an electron donor.

Artificial synthesis is an environment friendly photocatalytic strategy to converse carbon dioxide (CO2) into useful chemicals. However, water (H2O) splitting, producing (hydrogen) H2 strongly, is always a competitive reaction to CO2 conversion. Therefore, proper cocatalysts are generally needed to enhance CO2 conversion but suppress H2 production. In this work, zinc/gallium (Zn/Ga) dual co-catalysts consisting of Zn0 and amorphous ZnGa2O4 species were found to selectively produce carbon monoxide (CO) during the photocatalytic conversion of carbon dioxide (CO2) using water (H2O) as an electron
