Changes in articular cartilage during the aging process are a stage of natural changes in the human body. Old age is the major risk factor for osteoarthritis but the disease does not have to be an inevitable consequence of aging. Chondrocytes are particularly prone to developing age-related changes. Changes in articular cartilage that take place in the course of aging include the acquisition of the senescence-associated secretory phenotype by chondrocytes, a decrease in the sensitivity of chondrocytes to growth factors, a destructive effect of chronic production of reactive oxygen species and

Mesothelial cell (MC) senescence contributes to malignancy and tissue fibrosis. The role of telomere erosion in MC senescence remains controversial, with evidence for both telomere-dependent and telomere-independent mechanisms reported. Single telomere length analysis revealed considerable telomere length heterogeneity in freshly isolated human peritoneal MCs, reflecting a heterogeneous proliferative history and providing high-resolution evidence for telomere-dependent senescence. By contrast the attenuated replicative lifespan, lack of telomere erosion and induction of p16 expression in in vi

Aging is a natural process by which every single living organism approaches its twilight of existence in a natural way. However, aging is also linked to the pathogenesis of a number of complex diseases. This is the case for osteoarthritis (OA), where age is considered to be a major risk factor of this important and increasingly common joint disorder. Half of the world's population, aged 65 and older, suffers from OA. Although the relationship between the development of OA and aging has not yet been completely understood, it is thought that age-related changes correlate with other risk factors.

Understanding the malleable determinants of cellular aging is critical to understanding human longevity. Telomeres may provide a pathway for exploring this question. Telomeres are the protective caps at the ends of chromosomes. The length of telomeres offers insight into mitotic cell and possibly organismal longevity. Telomere length has now been linked to chronic stress exposure and depression. This raises the question of mechanism: How might cellular aging be modulated by psychological functioning? We consider two psychological processes or states that are in opposition to one another-threat

Most human cells can only replicate a limited number of times in cultures before they lose the ability to divide, a phenomenon known as replicative senescence, which seems to play a role in aging at the organismal level. Recent studies have shown that culture in low magnesium (Mg) accelerates the senescence of human endothelial cells and fibroblasts. Given the numerous critical roles of Mg, it seems likely that Mg inadequacy would interfere with cellular metabolism, which could affect the senescence process. Since i) several pieces of evidence link low Mg to aging and age-related diseases and

Telomeres are specialized DNA-protein structures at the ends of the linear chromosomes. In mammalian cells, they are composed of multifold hexameric TTAGGG repeats and a number of associated proteins. The double-stranded telomeric DNA ends in a 3' single stranded overhang of 150 to 300 base pair (bp) which is believed to be required for a higher order structure (reviewed in (Blackburn, 2001)). One important model is that the telomeres form loop structures, the T-loops, and by invasion of the 3' overhang into the duplex region of the double stranded part protect the DNA against degradation and

Chaperone function plays a key role in sequestering damaged proteins and in repairing proteotoxic damage. Chaperones are induced by environmental stress and are called as stress or heat shock proteins. Here, we summarize the current knowledge about protein damage in aged organisms, about changes in proteolytic degradation, chaperone expression and function in the aging process, as well as the involvement of chaperones in longevity and cellular senescence. The role of chaperones in aging diseases, such as in Alzheimer's disease, Parkinson's disease, Huntington's disease and in other neurodegene…

PURPOSE: Neuronal senescence contributes to several age-related neurodegenerative diseases occurring with aging. Meanwhile, aging of our bodies is accompanied by the systemic dysregulation of type I collagen, the major component in the extracellular matrix. This study investigates whether type I collagen plays a protective role in neuronal senescence.
METHODS: Cellular senescence was assessed by evaluating senescence-associated β-galactosidase staining, cell area, senescence-associated proteins, and senescence-associated secretory phenotypes (SASPs); ferroptosis was evaluated using C11-BODIPY…

Ischemic stroke is a major cause of disability and mortality worldwide, but its underlying pathogenic mechanisms remain elusive. Although cellular senescence has been implicated in a wide range of age-related diseases, its specific contribution through endothelial cells to ischemic stroke pathogenesis remains unclear. To address this gap, the present study adopted an integrative strategy, combining single-cell RNA sequencing (scRNA-seq) and bulk RNA sequencing (RNA-seq), to systematically characterize the senescent endothelial landscape in ischemic stroke. We identified a distinct senescence-a…

Decidualization, the transformation of endometrial stromal cells into specialized decidual cells, is essential for embryo implantation and pregnancy maintenance. This process involves immune cell infiltration, especially decidual natural killer (dNK) cells, which regulate immune responses and support tissue remodeling. Recent findings suggest that cellular senescence during decidualization is not just a byproduct but plays a functional role in enhancing uterine receptivity. However, excessive senescence leads to complications like recurrent pregnancy loss. dNK cells help maintain decidual home…

Senescent cells are cells that stop dividing but sustain viability. Cellular senescence is the hallmark of aging, but senescence also appears in cancer, triggered by cells stress, tumor suppression of gene activation, and oncogene activity. In lung cancer, senescent cancer cells secrete VEGF, which initiates a process of angiogenesis, enabling the cancer to grow and proliferate. Chemotherapy kills cancer cells, but some cancer cells become senescent. Hence, a senolytic drug, a drug that eliminates senescent cells, should significantly improve the efficacy of chemotherapy. In this paper, we dev…

Background: Bladder urothelial carcinoma (BLCA) is a prevalent malignant tumor known for its high recurrence rates and limited therapeutic efficacy. Cellular senescence has been extensively shown to inhibit tumorigenesis via cell cycle arrest. Consequently, the identification of senescence-associated biomarkers is essential for enhancing the diagnosis, prognosis, and immunotherapeutic outcomes of BLCA. Method: This study integrated the TCGA-BLCA and GSE13507 datasets to analyze senescence-related genes. ConsensusClusterPlus was employed for cluster analysis, while immune infiltration was asses…

Senescence is a degenerative process that occurs with ageing, and in the female reproductive system, senescence occurs earlier in the ovaries than in other tissues and organs, which implies a decrease in oocyte quality and exhaustion of the primordial follicular pool, leading to impaired ovarian function and an inability to maintain normal fertility. Unfortunately, the development of curative and effective treatments for ovarian senescence is still a considerable challenge. Currently, mesenchymal stem cells (MSCs)-based therapies for treating various refractory diseases, especially ovarian dys

Cellular senescence is a state of stable arrest and secretion linked to aging and disease. Here, we identify that senescent cells dispose of large fragments through cell-to-cell adhesion, which we term "senescent-cell adhesion fragments" (SCAFs). Found in many senescent states, including human and mouse cells, and mouse tissues, SCAFs lack nuclear material but contain organelles, including damaged mitochondria. Disrupting adherens junctions decreases SCAF formation but induces senescent-cell death, due to an inability to shed damaged mitochondria. Live imaging and proteomics show that SCAFs ul…

Cellular senescence is implicated as a driver of ovarian aging, but senescent cells in the human postmenopausal ovary remain poorly defined. Using spatially resolved p16INK4a protein expression, a canonical senescence marker, we identified and mapped senescent cells in postmenopausal ovaries. We integrated p16 immunohistochemistry, multiplexed immunofluorescence, spatial transcriptomics, and AI-guided digital pathology to map senescent microenvironments. p16-positive cells formed discrete stromal, vascular, and cyst-associated clusters that increased with age and were enriched for macrophages …

Cancer-associated fibroblasts (CAFs) are key components of the tumor microenvironment and contribute substantially to gastric cancer progression. However, the phenotypic and functional heterogeneity of CAFs in gastric cancer remains incompletely characterized. Here, we analyzed single-cell transcriptomic data from gastric cancer samples and revealed a distinct CAF subset with cellular senescence-associated features. Pathway enrichment analysis demonstrated marked activation of hypoxia-related signaling in senescent CAFs, whereas pseudotime analysis suggested that these cells may arise from oth…

BACKGROUND: Senescence, a state of permanent cell cycle arrest, is a complex cellular phenomenon closely affiliated with age-related diseases and pathological fibrosis. Cellular senescence is now recognized as a significant contributor to organ fibrosis, largely driven by transforming growth factor beta (TGF-β) signaling, such as in metabolic dysfunction-associated steatohepatitis (MASH), idiopathic pulmonary fibrosis (IPF), chronic kidney disease (CKD), and myocardial fibrosis, which can lead to heart failure, cystic fibrosis, and fibrosis in pancreatic tumors, to name a few. MASH is a progre…
