A rapid, inexpensive, and accurate high-performance liquid chromatographic method for the simultaneous determination of pentisomide and its major mono-N-dealkylated metabolite has been developed. After a simple and inexpensive solvent extraction procedure, the unchanged drug, its metabolite, and the internal standard are separated using a C18 reversed-phase column with a 5-microns particle size. The eluent is monitored with ultraviolet detection at 260 nm. Endogenous substances or a variety of drugs do not interfere with the assay. The mean recoveries of pentisomide and its metabolite are 92.6

A gas chromatography method using steam as the carrier gas was established for the determination of salicylic acid in plasma. A 0.5 mL plasma sample was mixed with 0.5 mL of 6N HCl and then extracted with 2 mL of dichloromethane. The dichloromethane was evaporated to dryness and the residue was redissolved in 0.5 mL of water and then analysed by gas chromatography using steam as the carrying gas. The result was calculated by external standardization.

A rapid and sensitive method for the routine quantitative determination of melatonin in pineal and plasma is described. The assay used reversed-phase high-performance liquid chromatography (RP-HPLC) separation combined with either amperometric (system A) or coulometric (system B) detection. Fluorides and monofluorophosphates present in drugs, toothpastes and mineral waters are converted into an organic compound by trimethylchlorosilane at acidic pH. The trimethylfluorosilane formed is determined with isopentane as internal standard by gas liquid chromatography with flame ionization detection. Recently available instruments dedicated to the clinical assay of hydrogen in breath were tested and the results compared with those from a conventional thermal conductivity gas chromatograph. We found the Quintron Model 12 Microlyzer and the GMI Exhaled Hydrogen Monitor to give much faster and more sensitive results than the Gow-Mac 550 conventional gas chromatograph, and their readout is digital rather than graphical. Chloroform and ether commonly are used as solvents to extract metabolic organic acids for analysis by gas-liquid chromatography in the identification of anaerobic bacteria. Because these solvents are potentially hazardous to personnel, modified extraction procedures involving the use of a safer solvent, methyl tert-butyl ether were developed which remained both simple to perform and effective for organism identification.

A new method has been developed for the determination of acetylsalicylic acid, salicylic acid, salicyluric acid and gentisic acid in plasma, urine and tissue homogenates by simple extraction with ethyl acetate, evaporation and redissolution and measuring by high-performance liquid chromatography. Linearity, reproducibility and recovery were determined. Experiments were carried out to investigate the decomposition of acetylsalicylic acid in plasma with fluoride at different temperatures. The method has been used for pharmacokinetic experiments and an example is given.
