Ultrastructure of macrophages and dendritic cells in osteopetrosis (op) mutant mice lacking macrophage colony-stimulating factor (M-CSF/CSF-1) activity.

The ultrastructural features of macrophages and dendritic cells of mice homozygous for osteopetrosis (op/op) mutation were studied. The mutant mice are characterized by defective differentiation of osteoclasts, monocytes, and tissue macrophages due to the lack of functional macrophage colony stimulating factor (M-CSF/CSF-1) activity. In op/op mice, tissue macrophages were reduced in number and smaller than in normal littermates. Macrophages in op/op mice showed various degrees of phagocytosis but the development of intracytoplasmic organelles and microvillous projections was poor. After admini

Evidence that granulocyte macrophage-colony-stimulating factor regulates the distribution and differentiated state of dendritic cells/Langerhans cells in human lung and lung cancers.

GM-CSF has been shown to be important for the survival and function of cells of dendritic cell/Langerhans cell (LC) lineage in vitro. Since these cells have been demonstrated to infiltrate human lung and some lung carcinomas, we hypothesized that the production of GM-CSF in the lung could be important in their recruitment and differentiation. Using both immunohistochemistry and in situ hybridization, we have shown that: (a) GM-CSF was produced by normal bronchiolar epithelium, the only site were CD1a+ LC are observed in the normal lung, whereas neither GM-CSF production nor LC were identified

Hyperimmune human IgG or recombinant human granulocyte-macrophage colony-stimulating factor as adjunctive therapy for group B streptococcal sepsis in newborn rats.

Group B streptococcus (GBS) continues to cause considerable morbidity and death in newborn infants despite the use of antibiotics. We investigated the use of adjunctive therapies to be used with antibiotics in the treatment of neonatal sepsis, using a neonatal rat model of established GBS disease. After the development of GBS bacteremia, a human IgG preparation hyperimmune for GBS, administered with penicillin, decreased the mortality rate compared with the use of penicillin alone (14% vs 57%; p = 0.02). Similarly, recombinant human granulocyte-macrophage colony-stimulating factor, administere

[Granulocyte-macrophage colony-stimulating factor. Treatment of patients with highly malignant lymphomas and acute lymphoblastic leukemias after autologous bone marrow transplantation].

We have evaluated the effect of intravenous infusions of granulocyte-macrophage colony-stimulating factor (GM-CSF) in ten patients undergoing autologous bone marrow transplantation after total body irradiation (11 Gy) and cyclophosphamide (120 mg/kg) conditioning for malignant lymphomas or acute lymphoblastic leukaemias. Mild side effects related to GM-CSF were seen in six patients, of whom one chose to discontinue treatment. Compared to a historic control group consisting of six patients treated with an identical conditioning regimen, a tendency was seen towards a more rapid neutrophil regene

Therapeutic administration of recombinant human granulocyte colony-stimulating factor accelerates hemopoietic regeneration and enhances survival in a murine model of radiation-induced myelosuppression.

The primary cause of death after radiation exposure is infection resulting from myelosuppression. Because granulocytes play a critical role in host defense against infection and because granulocyte proliferation and differentiation are enhanced by granulocyte colony-stimulating factor (G-CSF), this agent was evaluated for the ability to accelerate hemopoietic regeneration and to enhance survival in irradiated mice. C3H/HeN mice were irradiated and G-CSF (2.5 micrograms/day, s.c.) or saline was administered on days 3-12, 1-12 or 0-12 post-irradiation. Bone marrow, splenic and peripheral blood c

Escalated therapy for refractory urothelial tumors: methotrexate-vinblastine-doxorubicin-cisplatin plus unglycosylated recombinant human granulocyte-macrophage colony-stimulating factor.

Thirty-two assessable patients with metastatic urothelial tumors refractory to standard chemotherapy with methotrexate, vinblastine, doxorubicin, and cisplatin (MVAC) were treated with escalated doses of MVAC plus unglycosylated recombinant human granulocyte-macrophage colony-stimulating factor (rhGM-CSF). Results of this phase I trial revealed that escalated MVAC (30 mg of methotrexate/m2, 4 mg of vinblastine/m2, 60 mg of doxorubicin/m2, and 100 mg of cisplatin/m2) can be tolerated by heavily pretreated patients. The side effects of rhGM-CSF are dose- and schedule-dependent. The maximum toler

Effect of recombinant human granulocyte-macrophage colony-stimulating factor (rhuGM-CSF) on normal peripheral B lymphocytes and B lymphoblastoid cell lines.

The role of the granulocyte-macrophage colony-stimulating factor (GM-CSF) for the proliferation and differentiation of normal and leukemic myeloid cells has been extensively investigated. We examined whether rhuGM-CSF has any functional effect on normal purified B cells and cell lines from patients with B cell non-Hodgkin's lymphomas (NHL). Normal B cells were prepared by combining E-rosetting followed by two non-adherence procedures. Further B cell enrichment was achieved by complement-mediated lysis with a panel of antibodies directed against various T cell antigens. Alternatively, we incuba

Substance P enhances tissue factor release from granulocyte-macrophage colony-stimulating factor-dependent macrophages via the p22phox/β-arrestin 2/Rho A signaling pathway.

Granulocyte-macrophage colony stimulating factor (GM-CSF) induces procoagulant activity of macrophages. Tissue factor (TF) is a membrane-bound glycoprotein and substance P (SP) is a pro-inflammatory neuropeptide involved in the formation of membrane blebs. This study investigated the role of SP in TF release by GM-CSF-dependent macrophages. SP significantly decreased TF levels in whole-cell lysates of GM-CSF-dependent macrophages. TF was detected in the culture supernatant by enzyme-linked immunosorbent assay after stimulation of macrophages by SP. Aprepitant (an SP/neurokinin 1 receptor antag

Normal distribution of tumor necrosis factor-alpha messenger ribonucleic acid and protein in the uteri, placentas, and embryos of osteopetrotic (op/op) mice lacking colony-stimulating factor-1.

In order to map mouse tumor necrosis factor-alpha (TNF) gene expression in detail and to determine whether transcription or translation of the TNF gene is regulated by uterine colony stimulating factor-1 (CSF-1), preimplantation embryos, oviducts, uteri, and uteroplacental units were studied in various strains of mice. These included homozygous osteopetrotic (op/op) female mice, which completely lack CSF-1, and heterozygous (+/op) females, which have normal levels of CSF-1. TNF mRNA was identified in all samples except preimplantation embryos by use of Northern blot hybridization or reverse tr

Upfront use of plerixafor and granulocyte-colony stimulating factor (GCSF) for stem cell mobilization in patients with multiple myeloma: efficacy and analysis of risk factors associated with poor stem cell collection efficiency.

Plerixafor (P), an agent that selectively and reversibly binds to the chemokine receptor CXCR4, has been approved in combination with G-CSF (P + G-CSF) for stem cell (SC) mobilization in patients with multiple myeloma (MM). The goal of this study was to determine the SC collection success rate of P + G-CSF using a clinically relevant outcome defined as the ability to collect at least 5 × 106 CD34+ cells/kg to allow safely two transplants, and identify risk factors impacting SC mobilization. One hundred and thirty-eight patients were mobilized with P + G-CSF upfront following induction. The SC

Immune checkpoint inhibitor-based chemotherapy for granulocyte-colony stimulating factor-producing advanced esophageal squamous cell carcinoma: a report of three cases.

INTRODUCTION: Granulocyte-colony stimulating factor (G-CSF)-producing esophageal squamous cell carcinoma (ESCC) is a rare and highly aggressive malignancy. Currently, no specific treatment approach has been established for G-CSF-producing tumors, including those in the lungs, esophagus, and other organs. Here, we present three cases of advanced G-CSF-producing ESCC treated with immune checkpoint inhibitor (ICI) combination chemotherapy. CASE PRESENTATION: Case 1: A 52-year-old man was diagnosed with G-CSF-producing ESCC with multiple lymph node and lung metastases. First-line chemotherapy wit

A Marked Improvement in Mild but Progressive Autoimmune Pulmonary Alveolar Proteinosis Treated with Inhaled Granulocyte-Macrophage Colony-Stimulating Factor Therapy: A Case Report.

A 46-year-old man was diagnosed with autoimmune pulmonary alveolar proteinosis (aPAP). Despite repeated segmental bronchoscopic lavage, his symptoms, pulmonary function, and radiological findings gradually worsened, and he declined whole-lung lavage. Although classified as disease severity score 2 (partial pressure of arterial oxygen 75.2 mmHg), inhaled granulocyte-macrophage colony-stimulating factor (GM-CSF) therapy (sargramostim) was initiated. After 6 months, a marked improvement was observed, and the alveolar-arterial oxygen difference decreased from 21.8 to 1.1 mmHg. Given the limited ev

Multimodal evaluation of mannose engineered poly lactic glycolic acid nanoparticles with granulocyte colony stimulating factor focused delivery to bone marrow for neutropenia.

Neutropenia, characterized by a critical reduction in neutrophils, demands targeted therapeutic strategies to enhance the delivery efficiency of granulocyte colony-stimulating factor (G-CSF) specifically to bone marrow macrophages. This study focused on engineering mannose-modified poly(D, L-lactide-co-glycolide) (PLGA) nanoparticles (NPs) to achieve ligand-directed delivery of G-CSF. Mannose anchoring was achieved via Ethylenediamine (EDA)-mediated chemical ligation using N-hydroxysulfosuccinimide (NHS) and dicyclocarbodiimide as coupling agents, resulting in Mn-EDA-PLGA NPs. Systemic Neutralization of Granulocyte-Macrophage Colony-Stimulating Factor Attenuates Stellate Ganglion Neuroinflammation and Cardiac Sympathetic Overactivation in Chronic Heart Failure in Rats.

Chronic heart failure (CHF) is characterized by heightened cardiac sympathetic activity and neuroinflammation within the stellate ganglia (SG), contributing to cardiac arrhythmogenesis. However, the upstream mediators of this neuroimmune interaction remain incompletely defined. This study investigated whether systemic neutralization of granulocyte-macrophage colony-stimulating factor (GM-CSF, an inflammatory cytokine) attenuates SG inflammation and cardiac sympathetic overactivation in a rat model of coronary artery ligation-induced CHF. Male Sprague-Dawley rats underwent myocardial infarction
