Dextran, a renewable hydrophilic polysaccharide, is nontoxic, highly stable but intrinsically biodegradable. The α-1, 6 glycosidic bonds in dextran are attacked by dextranase (E.C. 3.2.1.11) which is an inducible enzyme. Dextranase finds many applications such as, in sugar industry, in the production of human plasma substitutes, and for the treatment and prevention of dental plaque. OBJECTIVE: To analyze adenovirus-mediated endosome lysis of T cells, we developed a novel approach based on pHrodo dextran (pH-sensitive fluorescent dye).
METHODS: After incubating Jurkat cells (T cell leukemia) with serotype 5 adenovirus (Ad5) and pHrodo dextran, we determined the optimal incubation time and concentration of pHrodo dextran. To assess viral lysis of the endosome, we monitored the ratio changes of mean fluorescence intensity in different time points by laser scanning confocal microscopy.

Experimental studies have suggested that oxidized cellulose and the combined use of gelatin sponge and dextran 70 may prevent intraperitoneal adhesion formation. Peritoneal adhesions were produced in 50 rats by a standardized caecal trauma. In 10 rats the injured caecum was covered by oxidized cellulose, in 10 by gelatin sponge, and in 10 by a mixture of gelatin sponge and dextran 70. Twenty rats served as controls. We describe the case of a 24-year-old healthy volunteer who underwent a dextran-induced anaphylactic/anaphylactoid reaction (DIAR) type III after administration of 10 ml 6% Dextran60 (0.6 g) during a preliminary examination. A reliable test for premalignant lesions in the development of colonic cancer in chronic ulcerative colitis has been needed. Thus, we designed this cytochemical study, using a model of experimental colitis and colonic tumors induced in Wistar male rats by the feeding of dextran sulfate sodium. Acute and chronic experimental ulcerative colitis models were produced in mice by providing them with drinking water containing synthetic dextran sulfate sodium. Mice that developed acute colitis showed signs of diarrhea, gross rectal bleeding, and weight loss within 6-10 days after ingesting 3%-10% dextran sulfate sodium. In adult albino rabbits, an opening wound was made in the central area of the cornea in one eye and the anterior chamber was made flat for 48 hours. Fluorescein-labeled dextran with a molecular weight of 70,000 was injected intravenously, and the uveal tissues were examined by fluorescence microscopy.
