Altered mitochondrial respiration in a chromosomal mutant of Neurospora crassa.

A mutant of Neurospora crassa (cni-1) has been isolated that has two pathways of mitochondrial respiration. One pathway is sensitive to cyanide and antimycin A, the other is sensitive only to salicyl hydroxamic acid. Respiration can proceed through either pathway and both pathways together in this mutant account for greater than 90% of all mitochondrial respiration. The cni-1 mutation segregates as a nuclear gene in crosses to other strains of Neurospora. Absorption spectra of isolated mitochondria from cni-1 show typical b- and c-type cytochromes but the absorption peaks corresponding to cyto

Genetic diversity of nine necrophagous Sarcophagidae (Diptera) species in Portugal.

Flesh flies (Diptera: Sarcophagidae) are among the most important insect groups used in forensic investigations, particularly for post-mortem interval estimation and assessing possible corpse relocation. Their practical use, however, is limited by the high morphological similarity of closely related species and by the limited representation of regional populations in DNA reference libraries. This study assessed two mitochondrial markers, cytochrome c oxidase subunit I (COI) and NADH dehydrogenase subunit 4 (ND4), for the identification and population-level characterisation of nine necrophagous

Lamellodiscus louiseuzeti n. sp. (Monopisthocotyla: Diplectanidae: Lamellodiscinae): a parasite of Chaetodon hoefleri (Teleostei, Chaetodontidae) with distinct attachment organ morphology.

During a study on representatives of Diplectanidae (Monopisthocotyla) infecting Chaetodon hoefleri, a marine teleost from Senegal, we collected individuals belonging to a species of Lamellodiscinae, which showed a unique arrangement of the haptoral lamellodiscs, so far unreported in the literature, with the first lamella of the ventral lamellodisc open anteriorly, unlike the dorsal lamellodisc, which is closed. Due to this arrangement, we describe the species Lamellodiscus louiseuzeti n. sp.. Apart from the morphology of the lamellodiscs, the new species differs from its congeners by having a

Forensic implications of early postmortem changes in skeletal, cardiac, and smooth muscles following acute colchicine intoxication in rats.

Acute colchicine (CHC) poisoning can be fatal due to its narrow therapeutic index, involvement of multiple organ systems, and the lack of a specific antidote. This study investigated early postmortem biochemical, histopathological, and immunohistochemical changes in skeletal, smooth, and cardiac muscles within 24 h of death following acute CHC intoxication in rats. Sixty male Sprague Dawley rats were divided into control and CHC-intoxicated groups (n = 30 each). Controls received saline and were euthanized humanely; CHC rats received a single oral lethal dose (4.9 mg/kg b.wt) and died naturall

Commensal oerstediid nemerteans from deep-sea styelid ascidians.

We report a new commensal relationship between oerstediid nemerteans and deep-sea ascidians. A total of six nemertean individuals were collected from the peribranchial cavities of several Styelidae ascidians attached to small rocks at bathyal depths (1,400-1,409 m) off northeastern Japan in the northwestern Pacific Ocean. Phylogenetic analyses based on combined mitochondrial cytochrome c oxidase subunit I (COI), 16S rRNA, histone H3, 18S rRNA, and 28S rRNA gene sequences placed this species within the 'Clade Gononemertes' of the hoplonemertean family Oerstediidae. Previous records of commensal

Necrophagous fly occurrence and primary colonizing species in forensic entomology cases from the Yeongnam Region, South Korea.

Forensic entomology is widely used in death investigations to estimate the minimum postmortem interval (mPMI). In South Korea, however, casework-based ecological data remain limited, particularly for indoor scenes. We established a collaborative framework with investigating officers in the Yeongnam region (Gyeongsangbuk-do, Daegu, and Busan) and compiled forensic entomology cases from 2019 to 2022 to summarize the occurrence patterns of necrophagous flies. Scene information and specimens were collected using a standardized field record form, and species were identified by morphological examina

Optic neuropathy arising from the synergy between YARS2 and mitochondrial COX1 mutations.

Leber hereditary optic neuropathy (LHON) is a paradigm for mitochondrial retinopathy. Here, we investigate the mechanism underlying the interaction between nuclear modifier and mtDNA mutation(s) that manifests optic neuropathy in vivo to develop an effective therapeutic approach for this disease, using mouse models bearing LHON-linked Yars2G186V or COIV421A mutation alone and double mutations. Yars2G186V alters mitochondrial translation and assembly and activities of complex I, III, and IV, while COIV421A reduces complex IV activity. However, a single Yars2G186V or COIV421A mutation causes mil

New Contracaecum species (Nematoda, Anisakidae) parasitizing Ardea cocoi Linnaeus (Aves, Ardeidae) in a mixed infection: morphological and molecular studies in Argentina.

This study provides the description of a new species of Anisakidae, Contracaecum cocoi sp. nov., as well as the record of Contracaecum jorgei, both species parasitizing the Cocoi heron Ardea cocoi (Ardeidae) in a locality from the Buenos Aires Province, Argentina. An integrative taxonomic approach was used, involving phylogenetic analyses and the examination of diagnostic morphological features in the studied specimens. Among other features, the new species can be morphologically distinguished by the papillae arrangement on the male tail: while C. jorgei exhibits a simple morphotype A, C. coco

Metagenomic Exploration of the Bacteriome Reveals Natural Wolbachia Infections in Yellow Fever Mosquito Aedes aegypti and Asian Tiger Mosquito Aedes albopictus.

Dengue and associated complications are spreading to non-endemic regions of Pakistan. Vector control, the foremost and widely adopted strategy for managing dengue has been implemented through various measures in Pakistan. Biological control through the use of Wolbachia, a bacterium naturally present in various insect genera, including Aedes, has demonstrated promising results globally. In this study we collected Aedes species and investigated its microbiomes with a particular focus on identifying the endosymbiont Wolbachia. Mosquitoes were collected via Gravitraps in the Peshawar region of Pak

Arterial tracing-based technique for rapid purification of pulmonary arterial smooth muscle cells from rat lungs.

Primary pulmonary arterial smooth muscle cells (PASMCs) of pre-capillary vessels are used to study the molecular mechanism of hypoxic pulmonary vasoconstriction (HPV). Cytochrome c oxidase subunit IV isoform 2 (COX4i2) is highly expressed in these PASMCs and an essential player in HPV, as its knockout abolishes the response. However, the exact molecular mechanism is not fully understood; therefore, fast and reliable methods to isolate PASMC from small pulmonary vessels that retain their phenotype during culture are essential. In the current study, we aimed to develop a fast and user-friendly m

Cardiac Tyrosine 97 Phosphorylation of Cytochrome c Regulates Respiration and Apoptosis.

It was previously reported that tyrosine 97 (Y97) of cytochrome c is phosphorylated in cow heart tissue under physiological conditions. Y97 phosphorylation was shown to partially inhibit respiration in vitro in the reaction with purified cytochrome c oxidase. Here, we use phosphomimetic Y97E Cytc to further characterize the functional effects of this modification both in vitro and in cell culture models. In vitro, phosphomimetic Y97E Cytc showed lower activity in the reaction with purified cow heart cytochrome c oxidase (COX), decreased caspase-3 activity, and reduced rate of reduction. Additi

CORM-3 mitigates hypoxia/reoxygenation-induced injury in neonatal rat cardiomyocytes by regulating mitochondrial-mediated apoptosis and complex IV activity.

BACKGROUND: Myocardial ischemia-reperfusion injury (MIRI) is a major contributor to myocardial infarction and leads to significant myocardial dysfunction. Mitochondria, crucial for cellular energy production, are particularly susceptible to damage during ischemia/reperfusion (I/R) events. Carbon monoxide-releasing molecule-3 (CORM-3), a water-soluble compound that releases carbon monoxide (CO), has demonstrated multiple protective effects against I/R injury. Mitochondria are recognized as selective targets for CO's protective actions in cells. PURPOSE: This study aimed to explore whether CORM

HIF1A transcriptional regulation of COX4I2 impacts angiogenesis in pheochromocytoma.

BACKGROUND: Pheochromocytoma (PCC) is a rare neuroendocrine tumor. Angiogenesis is primary contributing factor for tumorigenesis. Cytochrome c oxidase 4I2 (COX4I2) has been confirmed to take part in the progression of cancer. Hypoxia-inducible factor 1A (HIF1A) is the main regulatory factor for the steady-state response of hypoxia, involved in metabolism and angiogenesis. In this study, we intended to explore the functions of COX4I2 in PCC and the effect mechanism between HIF1A and COX4I2. MATERIALS AND METHODS: The RNA-sequencing and immunohistochemistry tested COX4I2 expression in highly va

Nuclear Control of Mitochondrial Homeostasis and Venetoclax Efficacy in AML via COX4I1.

Cell signaling pathways are enriched for biological processes crucial for cellular communication, response to external stimuli, and metabolism. Here, a cell signaling-focused CRISPR screen identified cytochrome c oxidase subunit 4 isoform 1 (COX4I1) as a novel vulnerability in acute myeloid leukemia (AML). Depletion of COX4I1 hindered leukemia cell proliferation and impacted in vivo AML progression. Mechanistically, loss of COX4I1 induced mitochondrial stress and ferroptosis, disrupting mitochondrial ultrastructure and oxidative phosphorylation. CRISPR gene tiling scans, coupled with mitochond
