Hypoxanthine phosphoribosyltransferase deficiency: association of reduced catalytic activity with reduced levels of immunologically detectable enzyme protein.

In the present study hemolysates from fourteen patients with a genetically determined deficiency of hypoxanthine phosphoribosyltransferase (EC 2.4.2.8; IMP:pyrophosphate phosphoribosyltransferase) activity were examined immunologically for the presence of material that crossreacts with the normal enzyme. A quantitative assay for crossreacting material in enzyme-deficient hemolysates was based on the inhibition of the immunoprecipitation of the normal enzyme. As little as 3% of normal crossreacting material could be detected. One patient in this series was found to have a normal amount of cross

The enzymic conversion of protoporphyrinogen IX to protoporphyrin IX. Protoporphyrinogen oxidase activity in mitochondrial extracts of Saccharomyces cerevisiae.

The oxidation of protoporphyrinogen IX to protoporphyrin IX in yeast cells is enzyme-dependent. The enzyme, protoporphyrinogen oxidase, associated with purified mitochondria isolated from Saccharomyces cerevisiae was solubilized by sonic treatment in the presence of detergent and partially purified. The molecular weight of the enzyme was 180,000 plus or minus 18,000. The purified preparation could be stored at -20 degrees in the presence of 20% glycerol for several months without loss of activity. Enzyme activity was destroyed by heating above 40 degrees and by proteolytic digestion and irreve

The course of ketosis and the activity of key enzymes of ketogenesis and ketone-body utilization during development of the postnatal rat.

1. The highest blood concentrations of ketone bodies were found at 5 days of age, after which time the concentration fell to reach the adult value by 30 days of age. 2. Both mitochondrial and cytoplasmic hydroxymethylglutaryl-CoA synthase activities were detected, with highest activities being found in the mitochondria at all stages of development. Activity of the mitochondrial enzyme increases rapidly immediately after birth, showing a maximum at 15 days of age, thereafter falling to adult values. The cytoplasmic enzyme, on the other hand, increased steadily in activity after birth to reach a

Effect of dietary carrageenan supplementation on growth performance, digestive enzyme and antioxidant enzyme activity, immune gene expression, and capacity against S. agalactiae of Nile tilapia (Oreochromis niloticus).

The current study aimed to ascertain the growth, antioxidant capacity, intestinal architecture, digestive enzyme activity, and disease resistance of S. agalactiae of Nile tilapia (Oreochromis niloticus) fed a diet enriched with carrageenan. Using an alkali method, the carrageenan was extracted from the seaweed Kappaphycus alvarezii. Its antiviral and antibacterial properties against Tilapia Lake virus and aquatic pathogens like Aeromonas veronii, Edwardsiella tarda, Streptococcus agalactiae, and Streptococcus iniae were assessed. A total of 450 healthy juvenile Nile tilapia (4.43 ± 0.71 g) wer

Tweaking Enzymatic Electric Field Can Evolve CYP450-like Activity in Catalase and CYP-P411 Enzymes.

Unlike CYP450 (cysteine-ligated), the axial ligations in Catalase (tyrosine-ligated) and CYP411-BM3 (serine-ligated) are different, which is generally attributed to the different catalytic efficiencies of these enzymes. Studies show that an interplay between push-pull effects due to different ligations modulates the reactivity of different enzymes. In the present study, we employ extensive MD simulation and a hybrid QM/MM approach to demonstrate that differences in the axial ligation in conjunction with protein residues generate a preorganized local electric field (LEF) that, in turn, controls

Plant GH9 enzymes and their β-1,4-glycan substrates: From in vitro EGase activity to in vivo functions.

Cellulases are an increasingly important class of industrial enzymes owing to their use in a wide range of applications and sustainable products. In addition to bacteria and fungi, plants also evolved a variety of cellulases. Plant endo-β-1,4-glucanases (EGases) belong to glycoside hydrolase family 9 (GH9) enzymes, which catalyze the cleavage of internal 1,4-glycosidic bonds in plant cell wall polysaccharides. Plant EGases are divided into three types (GH9A, GH9B, GH9C), which have been implicated in cellulose biosynthesis as well as polysaccharide catabolism to remodel and disassemble cell wa

Thermostable Carbohydrate-Active Enzymes for High-Temperature Biomass Conversion: From Molecular Design to Process Integration.

High-temperature processing of carbohydrate-rich biomass, including starch liquefaction and lignocellulose saccharification, can improve sugar yields, reduce contamination risks, and facilitate integration with thermochemical pretreatments. However, the operational stability of native carbohydrate-active enzymes (CAZymes) at 60-90 °C remains a major bottleneck for efficient and economically viable biorefineries, and existing reviews rarely integrate multiscale molecular mechanisms with process-relevant engineering strategies. This review systematically examines the multiscale basis of thermost

Conformation-gated dual enzyme activity in a hemoglobin-based gadolinium single-atom catalyst for adaptive biosensing.

Single-atom catalysts (SACs) are known for their exceptional catalytic efficiency but suffer from structural rigidity, limiting their adaptability in biological environments. Here, we present a hemoglobin-based gadolinium single-atom catalyst (Hb-Gd SAC), in which Gd atoms are site-specifically anchored adjacent to the native heme-Fe center via biomimetic coordination. Leveraging the intrinsic flexibility of the protein scaffold, the system exhibits pH- and conformation-gated switching between two distinct enzymatic modes: peroxidase-like (POD-like) activity under acidic conditions and laccase

Dandelion-like Pd/Cu nanozyme with multi-enzyme activities: A colorimetric sensor array for PFAS discrimination.

Per- and polyfluoroalkyl substances (PFAS) were a class of hazardous contaminants known for their high environmental persistence and toxicity. Due to their strong polarity, wide variety, and broad distribution, a rapid, accurate, and on-site detection method was urgently needed for future screening and emergency response. Herein, a dandelion-like Pd/Cu nanozyme with triple enzyme-like activities (oxidase-like, peroxidase-like, and laccase-like) was synthesized by one-step hydrothermal method. Different PFAS species inhibited these three enzyme-like activities to varying extents. By quantifying

Visualizing structural dynamics during nuclease activation across evolutionarily diverse IscB and CRISPR-Cas9 enzymes.

Cas9 nucleases of CRISPR-Cas adaptive immune systems are programmable RNA-guided DNA endonucleases that evolved from the transposon-associated IscB enzymes. Comparative structural studies have revealed substantial architectural elaboration during this evolutionary transition, including the replacement of the larger ωRNA scaffold of IscB with the REC domains of Cas9, capable of recognizing longer guide-target heteroduplexes. However, these evolutionary insights remain restricted to static structural features, and the dynamic properties that underlie nuclease activation have not been systematica
