Health benefits of a five-day at-home modified fasting program: a randomised controlled trial.

BACKGROUND: Fasting is one of the most cost-effective methods to improve cardiometabolic health. We tested a 5-day hypocaloric (~ 600 kcal/day) and ketogenic, modified fasting program (MFP) in a two-arm randomised controlled trial, where sixty-four healthy subjects were randomised to MFP or control group. METHODS: We randomly assigned 64 participants to a group receiving the MFP or to a group of participants who were told to continue with their usual eating behaviour and lifestyle (control group). The changes in blood pressure and body weight were considered as primary endpoints. Secondary ou

Magel2-null mice demonstrate contradictory outcomes in body weight, impulsivity, and motivation during distinct fasted states.

Prader- Willi Syndrome (PWS) is a complex neuroendocrine disorder that presents with hyperphagia, obesity, and social and cognitive impairment. The cognitive impairment is comparatively under-researched in PWS and in the Magel2-null mouse model. Cognitive function is commonly trained and tested in mice after the manipulation of metabolic state to induce motivation, yet physiologic function in the Magel2-null mouse model is most commonly assessed in the fed state. This project evaluates Magel2-null mouse physiology in ad libitum, acute fast, and chronic restriction settings to establish a basel

Food additive titanium dioxide (E171) alters gut microbial metabolic activity and butyrate production in the TIM-2 in vitro colon model.

AIMS: Food-grade titanium dioxide (E171) is widely used as a food additive, yet concerns persist regarding potential gastrointestinal effects, possibly mediated by interactions with the gut microbiome. This study aimed to investigate the physicochemical behavior of E171 under different digestive contexts and to assess its effects on gut microbial composition and metabolic activity. METHODS AND RESULTS: The dynamic in vitro colon model TIM-2 was used to expose human fecal microbiota to E171 under fasted (aqueous suspension; E171-aq) and fed (yogurt matrix; E171-yog) conditions. Hepatocyte-specific PPARγ Deletion Uncovers Role of an Antagonistic PPARγ-HNF4α Transcriptional Axis in Metabolic Dysfunction-Associated Steatotic Liver Disease Progression.

Systemic peroxisome proliferator-activated receptor gamma (PPARγ) agonist therapies have been investigated for clinical use in metabolic dysfunction-associated steatotic liver disease (MASLD). Previous studies, however, have shown that PPARγ activation in hepatocytes worsens MASLD. Overall, the precise molecular mechanisms regulated by PPARγ in hepatocytes during MASLD remain incompletely defined. Hepatocyte-specific PPARγ knockout mice were fed a clinically relevant fast-food diet for 2 or 5 months to characterize the role of PPARγ in MASLD progression. PPARγ expression and activity were incr

Physiological belching during impedance-pH monitoring: a meal with Coca-Cola provokes a prompt gastroesophageal reaction with increased prandial belching.

OBJECTIVES: To assess the belching-related esophageal events in healthy volunteers during and after fast-food meals. METHODS: Ten adult healthy volunteers had two lunches with a Big Mac® on different days, accompanied or not by Coca-Cola (500 mL, pH<3) in a random way. We assessed prandial and 2 h postprandial esophageal events using impedance-pH monitoring. RESULTS: During meals with Coca-Cola, most signed belching (68 %) were concomitant with prolonged esophageal acidification or acid reflux, and the belching number was superior [median: 9.5 (IQR 4.7-14) vs. 0 (0-3); p=0.002] and less freq

Intermittent fasting-induced autophagy normalization confers hepatic protection in metabolic dysfunction-associated fatty liver disease: Mechanistic insights and implications.

Metabolic dysfunction-associated fatty liver disease (MAFLD) is a chronic liver condition that can progress to steatohepatitis, cirrhosis, and even liver cancer. Macroautophagy (hereinafter referred to as autophagy) is a pro-survival mechanism that facilitates the lysosomal clearance of damaged organelles, abnormal proteins, and excess lipids. A growing body of evidence indicates that autophagy dysfunction and reduced autophagic flux play critical roles in the pathogenesis of MAFLD. Therefore, restoring autophagy in MAFLD may help reduce steatosis and prevent disease progression. Intermittent

Lipophilic Salts and Lipid-Based Formulations for Bridging the Food Effect Gap of Venetoclax.

Lipid based formulations (LBF) have shown to overcome food dependent bioavailability for some poorly water-soluble drugs. However, the utility of LBFs can be limited by low dose loading due to a low drug solubility in LBF vehicles. This study investigated the solubility and drug loading increases in LBFs using lipophilic counterions to form lipophilic salts of venetoclax. Venetoclax docusate was formed from venetoclax free base and verified by 1H NMR. Formation of stable venetoclax-fatty acid associations with either oleic acid or decanoic acid were attempted, however, the molecular associatio

Social jetlag, eating behaviours and BMI among adolescents in the USA.

There is a lack of research on associations of social jetlag with eating behaviours and obesity among adolescents. We examined the associations of social jetlag with eating behaviours and BMI in adolescents before and after adjustment for potential confounders. Self-report data were collected from 3060 adolescents (48·1 % female, mean age 15·59 (sd 0·77) years) from the Fragile Families and Child Wellbeing Study. In regression models, social jetlag predicted odds of consumption of breakfast, fruits/vegetables, fast food and sweetened drinks and BMI percentile. Primary models adjusted for schoo

LEAP2 changes with body mass and food intake in humans and mice.

Acyl-ghrelin administration increases food intake, body weight, and blood glucose. In contrast, mice lacking ghrelin or ghrelin receptors (GHSRs) exhibit life-threatening hypoglycemia during starvation-like conditions, but do not consistently exhibit overt metabolic phenotypes when given ad libitum food access. These results, and findings of ghrelin resistance in obese states, imply nutritional state dependence of ghrelin's metabolic actions. Here, we hypothesized that liver-enriched antimicrobial peptide-2 (LEAP2), a recently characterized endogenous GHSR antagonist, blunts ghrelin action dur

Untangling narcolepsy and diabetes: Pathomechanisms with eyes on therapeutic options.

Narcolepsy (NA) is a primary sleep disorder characterized by loss of hypocretinergic/orexinergic neurons. NA is associated with an increased risk for metabolic disorders such as diabetes mellitus (DM). Proposed mechanisms for this association are alterations in food intake, disruption of energy balance, glucose tolerance, and insulin sensitivity, as well as inflammation and genetic factors. Orexin deficiency, is associated with increased food intake and reduced basal metabolic rate (BMR) both leading to obesity. The anti-apoptotic effect of orexin on pancreatic beta-cells, increase in peripher

Oral insulin improves metabolic parameters in high fat diet fed rats.

INTRODUCTION/AIM: The gut has shown to have a pivotal role on the pathophysiology of metabolic disease. Food stimulation of distal intestinal segments promotes enterohormones secretion influencing insulin metabolism. In diabetic rats, oral insulin has potential to change intestinal epithelium behavior. This macromolecule promotes positive effects on laboratorial metabolic parameters and decreases diabetic intestinal hypertrophy. This study aims to test if oral insulin can influence metabolic parameters and intestinal weight in obese non-diabetic rats. METHODS: Twelve weeks old Wistar rats wer

PYY3-36 injection in mice produces an acute anorexigenic effect followed by a delayed orexigenic effect not observed with other anorexigenic gut hormones.

Peptide YY (PYY) is secreted postprandially from the endocrine L cells of the gastrointestinal tract. PYY(3-36), the major circulating form of the peptide, is thought to reduce food intake in humans and rodents via high-affinity binding to the autoinhibitory neuropeptide Y (NPY) receptor within the arcuate nucleus. We studied the effect of early light-phase injection of PYY(3-36) on food intake in mice fasted for 0, 6, 12, 18, 24, and 30 h and show that PYY(3-36) produces an acute anorexigenic effect regardless of the duration of fasting. We also show evidence of a delayed orexigenic effect in

Preclinical studies on human insulin. I. Acute and subacute toxicity studies.

Human insulin (prepared from porcine insulin) and porcine insulin were tested for acute toxicity by subcutaneous administration to unfasted mice and rats and fasted mice. The animals were observed for signs of reaction and post mortem examination was performed. LD50 values were calculated when possible. The LD50 values in the unfasted animals were several times higher than in the fasted mice, but similar for the two insulin preparations. The deaths and the signs observed were most probably caused by hypoglycaemia. No dose-related macroscopic organ changes were found. In a 28 day toxicity study
