When the fungus is susceptible, but the patient is not: reading antifungal susceptibility from bench to bedside.

Antifungal susceptibility testing is increasingly available and is central to the management of invasive, refractory, recurrent, and breakthrough fungal infections. However, minimum inhibitory concentrations (MICs) are not self-interpreting results. They measure in vitro antifungal activity under defined laboratory conditions, but clinical outcome is shaped by species identification, method, breakpoint or epidemiological cutoff values, availability, pharmacokinetic and pharmacodynamic exposure, infection site, fungal burden, source control, biofilm formation, and host immunity. Non-classic fun

Phytohormones in fungi: inter-kingdom modulators or fungal self-controlling elements?

SUMMARYLeveraging data from innovative experimental approaches, omics technologies, and bioinformatics, we offer new insights into how fungi communicate with and perceive their environment to achieve ecological success. By integrating comparative data from both the fungal and plant kingdoms, we critically reassessed the evolutionary, biochemical, and functional landscape of phytohormones in fungi, challenging the conventional notion that these molecules serve exclusively as plant regulators or as means of communication with them. Our analysis demonstrates that fungi not only synthesize a diver

Soil redox potential and plant species identity likely affect root-associated fungi in salt marshes.

Root-associated fungi (RAF) are important for plant performance because they form beneficial, neutral, or pathogenic relationships with their hosts, thus regulating nutrient uptake, stress tolerance, and overall plant fitness. At the transition from marine to terrestrial ecosystems, salt marshes are shaped by strong environmental gradients and provide vital ecosystem services such as climate regulation and coastal protection, yet RAF remain poorly understood in these systems. Therefore, we examined the community structure and abundance along this ecotone of RAF in general, and arbuscular mycor

Invasive candidiasis in the intensive care unit: diagnostic stewardship, source control, and precision antifungal therapy.

INTRODUCTION: Invasive candidiasis remains one of the most challenging infections in the intensive care unit (ICU) because diagnosis is complicated by nonspecific clinical manifestations, frequent colonization, altered host immunity, complex anatomical sources and unstable pharmacokinetics. Distinguishing true invasive disease from colonization is fundamental to optimizing patient outcomes while avoiding unnecessary antifungal exposure. AREAS COVERED: This narrative review examines invasive candidiasis in critically ill adults from an ICU perspective, based on a structured PubMed/MEDLINE, Emb

Region-associated microbial signatures persist during inoculated fermentation of Nebbiolo grapes.

Wine fermentation is driven by dynamic microbial communities that influence fermentation kinetics and wine composition. During fermentation, both the dominant Saccharomyces cerevisiae and diverse non-Saccharomyces yeasts, along with bacteria, actively contribute to metabolite and aroma formation, potentially reflecting vineyard environmental and geographic characteristics. Despite growing interest in the spatial structuring of grape-associated microbiota, knowledge remains limited regarding the persistence of these microbial signatures during inoculated fermentations. This study characterized

The distinctive pharmacokinetic profile of rezafungin, a long-acting echinocandin developed in the era of modern pharmacometrics.

Echinocandin drugs are the current first-line therapy for fungal infections caused by Candida spp. Most patients require once-daily intravenous (IV) administration in a hospital or outpatient setting for treatment, which may negatively impact their quality of life and stress healthcare resources. Similar to other echinocandins, the novel FDA-, EMA-, and Medical and Healthcare Products Regulatory Agency-approved echinocandin, rezafungin (CD101), exhibited strong antifungal activity against several fungal pathogens and a low drug-drug interaction liability, which are important for medically comp

The antimicrobial activity of ETD151 defensin is dictated by the presence of glycosphingolipids in the targeted organisms.

Fungal infections represent a significant global health concern, with a growing prevalence of antifungal drug resistance. Targeting glucosylceramides (GlcCer), which are functionally important glycosphingolipids (GSL) present in fungal membranes, represents a promising strategy for the development of antifungal drugs. GlcCer are associated with the antifungal activity of certain plant and insect defensins. The 44-residue ETD151 peptide, optimized from butterfly defensins, is active against several fungal pathogens. ETD151 has been shown to induce a multifaceted mechanism of action (MOA) in Bot

The Triad of Pathogenesis in Allergic Bronchopulmonary Aspergillosis: Interactions among Aspergillus fumigatus, Epithelium, and Immunity.

BACKGROUND: Aspergillus fumigatus is a common airborne fungal pathogen responsible for a range of pulmonary diseases, particularly in individuals with pre-existing respiratory conditions. Among these, allergic bronchopulmonary aspergillosis (ABPA) represents the most severe allergic manifestation. It arises from persistent airway colonization by A. fumigatus and repeated immune activation, often leading to uncontrolled respiratory symptoms and progressive lung damage. Despite its clinical relevance, the pathogenesis of ABPA remains poorly understood. SUMMARY: This review examines the pathogen

Plant defensin MtDef4-derived antifungal peptide with multiple modes of action and potential as a bio-inspired fungicide.

Chemical fungicides have been instrumental in protecting crops from fungal diseases. However, increasing fungal resistance to many of the single-site chemical fungicides calls for the development of new antifungal agents with novel modes of action (MoA). The sequence-divergent cysteine-rich antifungal defensins with multisite MoA are promising starting templates for design of novel peptide-based fungicides. Here, we experimentally tested such a set of 17-amino-acid peptides containing the γ-core motif of the antifungal plant defensin MtDef4. These designed peptides exhibited antifungal propert

Design, Characterization, and Antimicrobial Evaluation of Copper Nanoparticles Utilizing Tamarixinin a Ellagitannin from Galls of Tamarix aphylla.

The application of plant extracts or plant-derived compounds in the green synthesis of metal nanoparticles (NPs) was researched. Determining the exact metabolite implicated in the formation of NPs would necessitate comprehensive investigations. Copper nanoparticles (CuNPs) are gaining a lot of attention because of their unique properties and effectiveness against a wide range of bacteria and fungi, as well as their potential for usage in catalytic, optical, electrical, and microelectronics applications. In the course of this study, we aimed to formulate CuNPs utilizing pure tamarixinin A (TA)

AICAR transformylase/IMP cyclohydrolase (ATIC) is essential for de novo purine biosynthesis and infection by Cryptococcus neoformans.

The fungal pathogen Cryptococcus neoformans is a leading cause of meningoencephalitis in the immunocompromised. As current antifungal treatments are toxic to the host, costly, limited in their efficacy, and associated with drug resistance, there is an urgent need to identify vulnerabilities in fungal physiology to accelerate antifungal discovery efforts. Rational drug design was pioneered in de novo purine biosynthesis as the end products of the pathway, ATP and GTP, are essential for replication, transcription, and energy metabolism, and the same rationale applies when considering the pathway

Identification and Phenotypic Characterization of Hsp90 Phosphorylation Sites That Modulate Virulence Traits in the Major Human Fungal Pathogen Candida albicans.

The highly conserved, ubiquitous molecular chaperone Hsp90 is a key regulator of cellular proteostasis and environmental stress responses. In human pathogenic fungi, which kill more than 1.6 million patients each year worldwide, Hsp90 governs cellular morphogenesis, drug resistance, and virulence. Yet, our understanding of the regulatory mechanisms governing fungal Hsp90 function remains sparse. Post-translational modifications are powerful components of nature's toolbox to regulate protein abundance and function. Phosphorylation in particular is critical in many cellular signaling pathways an

Design and testing of selective inactivators against an antifungal enzyme target.

Systemic infections from fungal organisms are becoming increasingly difficult to treat as drug resistance continues to emerge. To substantially expand the antifungal drug landscape new compounds must be identified and developed with novel modes of action against previously untested drug targets. Most drugs block the activity of their targets through reversible, noncovalent interactions. However, a significant number of drugs form irreversible, covalent bonds with their selected targets. While more challenging to develop, these irreversible inactivators offer some significant advantages as nove

Moonlighting proteins are variably exposed at the cell surfaces of Candida glabrata, Candida parapsilosis and Candida tropicalis under certain growth conditions.

BACKGROUND: Adaptability to different environmental conditions is an essential characteristic of pathogenic microorganisms as it facilitates their invasion of host organisms. The most external component of pathogenic yeast-like fungi from the Candida genus is the multilayered cell wall. This structure is composed mainly of complex polysaccharides and proteins that can undergo dynamic changes to adapt to the environmental conditions of colonized niches. RESULTS: We utilized cell surface shaving with trypsin and a shotgun proteomic approach to reveal the surface-exposed proteins of three import

Isavuconazole for Treatment of Experimental Fungal Endophthalmitis Caused by Aspergillus fumigatus.

Fungal endophthalmitis remains a significant cause of vision impairment and blindness. Moreover, the prognosis is poor, in part due to delay in diagnosis and to limited availability of effective antifungal agents with good ocular penetration. Thus, it is imperative to evaluate the therapeutic efficacy in fungal endophthalmitis of newer antifungal agents. In this study, we assessed the efficacy of isavuconazole in treating Aspergillus fumigatus endophthalmitis in an exogenous mouse model of the disease. Briefly, endophthalmitis was induced by intravitreal (IVT) injection of A. fumigatus spores

Mannoprotein MP84 mediates the adhesion of Cryptococcus neoformans to epithelial lung cells.

The capsule is the most important virulence factor of the fungal pathogen Cryptococcus neoformans. This structure consists of highly hydrated polysaccharides, including glucuronoxylomannan (GXM), and galactoxylomannan (GalXM). It is also composed of mannoproteins (MPs) which corresponds to less than 1% of the capsular weight. Despite MPs being the minority and least studied components, four of these molecules with molecular masses of 115, 98, 88, and 84 kDa were identified and characterized as C. neoformans immunoreactive antigens involved in the pathogenesis, and are potential cryptococcosis

Soil bacterial and fungal community successions under the stress of chlorpyrifos application and molecular characterization of chlorpyrifos-degrading isolates using ERIC-PCR.

Chlorpyrifos is a widely used insecticide in recent years, and it will produce adverse effects on soil when applied on crops or mixed with soil. In this study, nested polymerase chain reaction (PCR) and denaturing gradient gel electrophoresis (DGGE) were combined to explore the bacterial and fungal community successions in soil treated with 5 and 20 mg/kg of chlorpyrifos. Furthermore, isolates capable of efficiently decomposing chlorpyrifos were molecular-typed using enterobacterial repetitive intergenic consensus-PCR (ERIC-PCR). Under the experimental conditions, degradation of chlorpyrifos i

Detection of antibodies against Paracoccidioides brasiliensis melanin in in vitro and in vivo studies during infection.

Several cell wall constituents, including melanins or melanin-like compounds, have been implicated in the pathogenesis of a wide variety of microbial diseases caused by diverse species of pathogenic bacteria, fungi, and helminthes. Among these microorganisms, the dimorphic fungal pathogen Paracoccidioides brasiliensis produces melanin in its conidial and yeast forms. In the present study, melanin particles from P. brasiliensis were injected into BALB/c mice in order to produce monoclonal antibodies (MAbs). We identified five immunoglobulin G1 (IgG1) κ-chain and four IgM melanin-binding MAbs. T

Engineering Penicillium expansum antifungal proteins unveils new clues about their mode of action.

Fungal antifungal proteins (AFPs) are promising biofungicides. PeAfpA and PeAfpB from Penicillium expansum show different activity profiles and potency, with PeAfpA being more active. Based on the PeAfpB solved structure, we had previously designed PeAfpB-PeAfpA chimeras that showed different properties. From these, we engineer here two additional variants, chPeAFPV6 and chPeAFPV7, that revealed novel aspects of the AFP structure, antifungal determinants and mechanism. chPeAFPV6, with a single E11K mutation in the loop L1 that is part of the γ-core motif, increased PeAfpB antifungal activity t
