Primary aldosteronism and lower-extremity arterial disease: a two-sample Mendelian randomization study.

BACKGROUND AND AIMS: Primary aldosteronism (PA) is an adrenal disorder of autonomous aldosterone secretion which promotes arterial injury. We aimed to explore whether PA is causally associated with lower-extremity arterial disease (LEAD). METHODS: We included 39,713 patients with diabetes and 419,312 participants without diabetes from UK Biobank. We derived a polygenic risk score (PRS) for PA based on previous genome-wide association studies (GWAS). Outcomes included LEAD and LEAD related gangrene or amputation. We conducted a two-sample Mendelian randomization analysis for PA and outcomes to

Identification of novel common variants associated with chronic pain using conditional false discovery rate analysis with major depressive disorder and assessment of pleiotropic effects of LRFN5.

Chronic pain is a complex trait that is moderately heritable and genetically, as well as phenotypically, correlated with major depressive disorder (MDD). Use of the conditional false discovery rate (cFDR) approach, which leverages pleiotropy identified from existing GWAS outputs, has been successful in discovering novel associated variants in related phenotypes. Here, genome-wide association study outputs for both von Korff chronic pain grade and for MDD were used to identify variants meeting a cFDR threshold for each outcome phenotype separately, as well as a conjunctional cFDR (ccFDR) thresh

Blood and urine biomarkers and myocardial infarction: A 2-sample and multivariate combination of Mendelian randomization.

Myocardial infarction (MI) is one of the most serious cardiovascular diseases in the world. Nevertheless, the majority of diagnostic procedures conducted subsequent to the illness do not provide any means to prevent several risks associated with MI. Blood and urine tests are frequently employed in clinical examinations to detect cardiovascular diseases at an early stage. Mendelian randomization (MR) is commonly employed to explore disease-trait relationships and uncover therapeutic targets. Our goal was to explore the genetic links between 35 blood and urine biomarkers and MI. Blood and urine

Exploring the Role of B-Cell Biotherapy in Myasthenia Gravis from a Genetic Perspective: A Mendelian Randomization Study.

Myasthenia gravis (MG) is an autoimmune disease caused by dysfunction at the neuromuscular junction. Current studies on MG are increasingly focusing on the involvement of B cell immunophenotypes in the development of the disease. Nonetheless, the specific correlation between B cells and MG still needs to be fully clarified. From published GWAS studies, we gathered summary results on B cell immunophenotypes and MG. We performed a Two-sample Mendelian randomization (MR) analysis to investigate the potential causal relationship between B cells and MG. The primary estimation was done by using IVW

Evaluation for causal effects of socioeconomic traits on risk of female genital prolapse (FGP): a multivariable Mendelian randomization analysis.

BACKGROUND: Although observational studies have established some socioeconomic traits to be independent risk factors for pelvic organ prolapse (POP), they can not infer causality since they are easily biased by confounding factors and reverse causality. Moreover, it remains ambiguous which one or several of socioeconomic traits play predominant roles in the associations with POP risk. Mendelian randomization (MR) overcomes these biases and can even determine one or several socioeconomic traits predominantly accounting for the associations. OBJECTIVE: We conducted a multivariable Mendelian ran

There may be a differential mechanistic impact on colorectal cancer of lactose-containing foods between lactase persistent and lactase non-persistent populations.

It is generally suggested that milk and milk products reduce the risk of colorectal cancer (CRC). While there is some controversy over specific sites affected throughout the colon and the benefits of specific dairy foods (DFs), there is a general consensus that calcium intake is the main mechanism of the cancer-reducing effects. This opinion may be sidelined by several other mechanisms. There is also a potentially important compensatory mechanism in populations with adult genetic lactase deficiency. The microbiome changes occur through a process of adaptation to continued lactose consumption.

Genetic Causal Association Between Vitamin E and Depression: A Two-Sample Mendelian Randomization Study.

BACKGROUND AND PURPOSE: Evidence regarding the association between vitamin E and depression is inconsistent. To address this uncertainty, we employed a two-sample Mendelian randomization (MR) approach to evaluate the potential causal effect of vitamin E on depression risk. METHODS: Genetic associations for vitamin E and depression were obtained from large-scale, publicly available genome-wide association studies (GWAS). We applied five complementary MR methods: inverse-variance weighted (IVW), MR-Egger, weighted median, simple mode, and weighted mode, with IVW serving as the primary analysis.

Relationship between creatine kinase and gastric cancer: A two-sample Mendelian randomization study.

Observational studies have suggested associations between circulating creatine kinase (CK) levels and gastric cancer (GC), yet causality remains uncertain. To evaluate whether genetically predicted CK levels are causally related to GC risk, and vice versa, using a 2-sample Mendelian randomization (MR) design. A 2-sample MR design was employed, incorporating statistical methods such as inverse-variance weighting (IVW), Weighted Median, and MR-Egger regression. Candidate single-nucleotide polymorphisms associated with CK and GC were identified from genome-wide association studies datasets. Heter

Causal relationship between serum metalloproteinase 12 levels and aortic dissection and aortic aneurysm: a bidirectional Mendelian randomization study.

BACKGROUND: Elevated matrix metalloproteinase-12 (MMP-12) levels have been shown to be elevated in patients with aortic dissection (AD) and aortic aneurysm (AA). However, whether MMP-12 is associated with AD and AA has not been conclusively examined. The aim of this study was to clarify the role of MMP-12 in AD and AA formation and to verify the correlation between MMP-12 and AD and AA development at the gene level via Mendelian randomization (MR) analysis. METHODS: The data for analyzing MMP-12 gene mutations were obtained from the Integrative Epidemiology Unit (IEU) OpenGWAS database, which

Mendelian randomization analysis unveiling the genetic connection: How psychiatric disorders heighten cancer risk.

The potential links between psychiatric disorders and cancer risk have attracted significant attention in recent years. However, the causal relationships between these conditions remain unclear. To assess causality, a Mendelian randomization analysis was conducted using single nucleotide polymorphisms from large-scale European genome-wide association studies (GWAS). The inverse variance weighted (IVW) method was applied as the primary method for causal estimation, with additional sensitivity analyses performed to evaluate potential pleiotropic effects. IVW estimates revealed that bipolar disor
