Metagenomics and chemotherapy-induced nausea: A roadmap for future research.

Uncontrolled chemotherapy-induced nausea and vomiting can reduce patients' quality of life and may result in premature discontinuation of chemotherapy. Although nausea and vomiting are commonly grouped together, research has shown that antiemetics are clinically effective against chemotherapy-induced vomiting (CIV) but less so against chemotherapy-induced nausea (CIN). Nausea remains a problem for up to 68% of patients who are prescribed guideline-consistent antiemetics. Despite the high prevalence of CIN, relatively little is known regarding its etiology independent of CIV. This review summar

Overview of the molecular determinants contributing to the expression of Psoriasis and Psoriatic Arthritis phenotypes.

Psoriasis and psoriatic arthritis are multifactorial chronic disorders whose etiopathogenesis essentially derives from the alteration of several signalling pathways and the co-occurrence of genetic, epigenetic and non-genetic susceptibility factors that altogether affect the functional and structural property of the skin. Although shared and differential susceptibility genes and molecular pathways are known to contribute to the onset of pathological phenotypes, further research is needed to dissect the molecular causes of psoriatic disease and its progression towards Psoriatic Arthritis. This

Engineered Salmonella allows real-time heterologous gene expression monitoring within infected zebrafish embryos.

Microbial host-pathogen interactions have been traditionally well studied at genetic and physiological levels, but cell-resolution analyses have been particularly scarce. This has been especially remarkable for intracellular parasites for two major reasons: first, the inherent loss of bacteria traceability once infects its hosts; second and more important, the limited availability of genetic tools that allow a tight regulated expression of bacterial virulence genes once inside the host tissues. Here we present novel data supporting the use of zebrafish embryos to monitor Salmonella enterica se

Strain-level genomic variation of Streptococcus mutans and early childhood caries in preschool children from Northern Arizona and Hawaii.

BACKGROUND: Early childhood caries (ECC) is the most common chronic disease of childhood, with especially high prevalence in Arizona and Hawaii where several racial and ethnic groups experience disproportionate burden. Although ECC is a polymicrobial disease, Streptococcus mutans plays a central role in its development, and evidence suggests that strain-level genetic variation influences its cariogenic potential. Understanding whether specific S. mutans lineages are associated with higher ECC risk and whether these lineages are more common in disproportionately affected groups is an important

Comparative transcriptomics and scarless genome editing uncover a periplasmic c-type cytochrome important for extracellular electron uptake in Thioclava electrotropha ElOx9T.

UNLABELLED: Extracellular electron uptake (EEU) is a form of extracellular electron transfer that enables microorganisms to use solid-phase electron donors for respiration and biosynthesis, with important implications for biogeochemical cycling and biotechnology. Thioclava electrotropha is a metabolically versatile marine Alphaproteobacterium capable of autotrophic sulfur oxidation, heterotrophic growth, in addition to cathodic electron uptake despite lacking homologs to known extracellular electron transfer proteins from mineral-reducing or mineral-oxidizing microbes. Here, we explore the gen

Within-Host Genotypic and Phenotypic Diversity of Contemporaneous Carbapenem-Resistant Klebsiella pneumoniae from Blood Cultures of Patients with Bacteremia.

It is unknown whether bacterial bloodstream infections (BSIs) are commonly caused by single organisms or mixed microbial populations. We hypothesized that contemporaneous carbapenem-resistant Klebsiella pneumoniae (CRKP) strains from blood cultures of individual patients are genetically and phenotypically distinct. We determined short-read whole-genome sequences of 10 sequence type 258 (ST258) CRKP strains from blood cultures in each of 6 patients (Illumina HiSeq). Strains clustered by patient by core genome and pan-genome phylogeny. In 5 patients, there was within-host strain diversity by gen

Mobilizable Plasmids for Tunable Gene Expression in Francisella novicida.

Francisella tularensis is the causative agent of the life-threatening disease tularemia. However, the molecular tools to study Francisella are limited. Especially, expression plasmids are sparse and difficult to use, as they are unstable and prone to spontaneous loss. Most Francisella expression plasmids lack inducible promoters making it difficult to control gene expression levels. In addition, available expression plasmids are mainly designed for F. tularensis, however, genetic differences including restriction-modification systems impede the use of these plasmids in F. novicida, which is of

Gingival Enlargement Induced by Orthodontic Therapy: A Systematic Review.

OBJECTIVE: The etiology of orthodontic-induced gingival enlargement (OIGE) or orthodontic-induced gingival overgrowth (OIGO) is multifactorial and is related to issues such as poor oral hygiene and the hormonal changes that are common during puberty. It is known that brackets facilitate the accumulation of dysbiotic microbial biofilm and that mechanical stress on the periodontium is a possible factor in the development of OIGE. Therefore, the objective of this systematic review was to analyze the principal factors involved in the pathogenesis of OIGE. METHODS: Independent reviewers performed

Integrated multi-omics analysis identifies microbial and metabolic signatures and drivers of CNS autoimmunity.

Multiple sclerosis (MS) is an autoimmune disease of the central nervous system (CNS), driven by genetic and environmental determinants. The gut microbiome of people with MS (pwMS) is distinct and influences disease through immunomodulatory metabolite production. Circulating metabolites are altered in pwMS, but identifying microbial-metabolic drivers remains challenging. We previously showed that colonization by the gut commensal Limosilactobacillus reuteri exacerbates disease in the experimental autoimmune encephalomyelitis (EAE) model of MS in a tryptophan-dependent manner. Here, we integrate

Genome-wide discovery of epistatic loci affecting antibiotic resistance in Neisseria gonorrhoeae using evolutionary couplings.

Genome analysis should allow the discovery of interdependent loci that together cause antibiotic resistance. In practice, however, the vast number of possible epistatic interactions erodes statistical power. Here, we extend an approach that has been successfully used to identify epistatic residues in proteins to infer genomic loci that are strongly coupled. This approach reduces the number of tests required for an epistatic genome-wide association study of antibiotic resistance and increases the likelihood of identifying causal epistasis. We discovered 38 loci and 240 epistatic pairs that infl

Precision nutrition targeting the gut microbiota for weight management: mechanisms and applications.

The gut microbiota, a highly individualized microbial community, is increasingly recognized for its role in weight regulation and metabolic health. This narrative review examines the bidirectional associations between gut microbiota and weight management, focusing on mechanisms involving energy metabolism, immune modulation, inflammatory responses, and appetite control. We also explore the interplay between gut microbiota and host genetic/epigenetic factors in obesity development. The review discusses how precision nutrition interventions may modulate gut microbiota composition to support pers

Mechanisms of Inflammation in Post-Operative Recurrence of Crohn's Disease.

Despite advances in guideline-directed surveillance and initiation of biologic therapy after ileocecal resection for Crohn's disease, many patients still experience post-operative recurrence. There is a growing field of research on the mechanisms of inflammation that are specific to post-operative recurrence after surgical resection and anastomosis. Within the innate immune system, certain genetic risk factors and an imbalance in matrix metalloproteinases have been implicated; however, the roles of specific innate immune cells are still being studied. Within the adaptive immune system, increas

[Ocular rosacea : Clinical aspects, diagnostics, management and treatment].

Ocular rosacea is a chronic inflammatory disease that affects the surface of the eye and the eyelids. It often occurs in conjunction with rosacea in the facial area but can also occur independently. It is characterized by bilateral chronic posterior blepharitis and meibomitis, which can involve the entire surface of the eye, including the cornea, during the course of the disease. The diagnosis is largely based on clinical findings. The main symptoms include reddened, burning and itchy eyes, dryness, sensitivity to light and blurred vision. The exact etiology of ocular rosacea is not fully unde

Nanosystems for modulation of immune responses in periodontal therapy: a mini-review.

Periodontitis is one of the most common oral diseases. It is generally treated by non-surgical and/or surgical therapy with adjunctive approaches for prevention and control. The current understanding of the pathogenesis of periodontitis has unraveled the importance of the inflammatory and immune reactions to combat periodontitis whose etiology is an overlap of microbial, genetic, and environmental factors in a susceptible host. Based on this premise, many therapeutic modalities have been investigated or attempted to resolve this inflammatory disease. Amongst these, nanomedicine has been shown

CRISPR/Cas Systems as Diagnostic and Potential Therapeutic Tools for Enterohemorrhagic Escherichia coli.

Following its discovery as an adaptive immune system in prokaryotes, the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated proteins (Cas) system has been developed into a multifaceted genome editing tool. This review compiles findings aimed at implementation of this technology for selective elimination or attenuation of enterohemorrhagic Escherichia coli (EHEC). EHEC are important zoonotic foodborne pathogens that cause hemorrhagic colitis and can progress to the life-threatening hemolytic uremic syndrome (HUS). Advancements in the application of CRISPR metho

Use of proximity ligation shotgun metagenomics to investigate the dynamics of plasmids and bacteriophages in the gut microbiome following fecal microbiota transplantation.

Proximity ligation shotgun metagenomics facilitate the analysis of the relationships between mobile genetic elements, such as plasmids and bacteriophages, and their specific bacterial hosts. We applied this technique to investigate the changes in the fecal microbiome of patients receiving fecal microbiota transplantation (FMT) for recurrent Clostridioides difficile infections (rCDI). FMT was associated with successful engraftment of donor bacteria along with their associated bacteriophages. While fecal microbial diversity increased in all patients, the extent of specific bacterial taxa engraft

Can the COVID-19 pandemic advance neuroinfectious research?

Investments in SARS-CoV-2 research provide a unique opportunity to explore how microbes may contribute to neurological conditions, an area of investigation that has been chronically underfunded. As exemplified by HIV/AIDS funding, crisis-driven research can yield broader biomedical advances, including spillover effects that address unanticipated and unmet medical needs. Leveraging newly established SARS-CoV-2 funding opportunities to study immune crosstalk and genetic predispositions could reveal therapeutic pathways and biomarkers for individuals who are vulnerable to infection-related dement

The role of the gut and intestinal dysbiosis in the pathogenesis of spondyloarthritis.

Spondyloarthritis (SpA) encompasses chronic inflammatory diseases affecting both axial and peripheral joints. Emerging evidence highlights a pivotal role for the gut-joint axis in SpA pathogenesis, where intestinal dysbiosis and barrier dysfunction facilitate microbial translocation and trigger systemic immune activation. Clinical observations of subclinical gut inflammation, alongside findings from HLA-B27 transgenic rats and SKG mice, underscore the gut's role in initiating joint pathology. Genetic predispositions, particularly HLA-B27, further exacerbate these processes through mechanisms s
