Dendritic Cells as Immunometabolic Regulatory Nodes in Diabetes: Molecular Mechanisms and Therapeutic Reprogramming.

Diabetes mellitus comprises a group of heterogeneous metabolic disorders characterized by persistent hyperglycemia, progressive β-cell dysfunction, and multi-organ complications. Although type 1 diabetes mellitus (T1DM) and type 2 diabetes mellitus (T2DM) have distinct pathogenic origins, both involve immune dysregulation, tissue stress, release of danger signals, and chronic inflammation. Dendritic cells (DCs), as antigen-presenting cells, integrate innate immune sensing, antigen presentation, cytokine production, T-cell regulation, and peripheral immune tolerance, placing them at a critical

Can SARS-CoV-2 Virus Use Multiple Receptors to Enter Host Cells?

The occurrence of the novel severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), responsible for coronavirus disease 2019 (COVD-19), represents a catastrophic threat to global health. Protruding from the viral surface is a densely glycosylated spike (S) protein, which engages angiotensin-converting enzyme 2 (ACE2) to mediate host cell entry. However, studies have reported viral susceptibility in intra- and extrapulmonary immune and non-immune cells lacking ACE2, suggesting that the S protein may exploit additional receptors for infection. Studies have demonstrated interactions between

Leukemia-targeting NK cell nanoengagers effectively promote robust NK activation and potent anti-acute myeloid leukemia response.

Natural killer (NK) cells are key components of innate immunity, playing a pivotal role in tumor recognition and eradication, and numerous NK cell-based immunotherapeutic approaches have been extensively investigated for cancer treatment. Here, we develop leukemia-targeting NK cell nanoengagers, termed AaLS/aCD16Nb/aCD13Nb, by simultaneously displaying NK cell-engaging nanobodies (aCD16Nb) and acute myeloid leukemia (AML)-targeting nanobodies (aCD13Nb) on lumazine synthase (AaLS) protein nanoparticles. The AaLS/aCD16Nb/aCD13Nb nanoengagers effectively bind to both NK cells and AML cells, there

Pathogenic TET2 Variants and Autoinflammatory Manifestations in Myeloid Hematologic Malignancies: Case Report and Literature Review.

Emerging evidence suggests that somatic mutations in genes associated with innate immunity can trigger adult-onset autoinflammatory diseases. Notably, loss-of-function variants in the TET2 gene have been linked to both hematological malignancies and immune-mediated disorders. A case is presented of a 73-year-old woman with chronic myelomonocytic leukemia who developed severe pericardial effusion secondary to inflammatory serositis, associated with a pathogenic TET2 variant. Despite initial treatment with corticosteroids and diuretics, her condition worsened, which led to the need to implement

A genome-wide map of the genetic network in monocytes that regulates type I interferon induction by the cGAS-STING pathway.

Type I interferons (IFNs) are induced by pattern recognition receptors (PRRs) of the innate immune system to protect against foreign pathogens and malignant transformation, and their production must be tightly regulated to balance antiviral defense with tissue homeostasis. To define the genetic network that governs IFN regulation, we conducted genome-wide CRISPR screens for genes that positively or negatively regulated IFNB1 gene expression induced by the PRR cGAS and its downstream effector STING, in both unprimed THP-1 cells and cells primed with IFN-α to mimic an ongoing inflammatory respon

Harnessing chitosan-based biomaterials for advanced cancer immunotherapy: from nanocarriers to tumor microenvironment modulation.

Cancer immunotherapy has transformed oncology, however, its clinical efficacy remains limited by immunosuppressive tumor microenvironments (TMEs), poor therapeutic delivery, systematic toxicity, treatment resistance. Chitosan, a biocompatible and biodegradable polysaccharide, has emerged as a versatile biomaterial capable of addressing these challenges through both intrinsics immunomodulatory activity and advanced drug-delivery functions. This review summarizs recent advances in chitosan-based biomaterials for cancer immunotherapy, highlighting their ability to activate innate and adaptive imm

[Research progress on mechanisms of macrophages in innate immunity against invasive fungal infections].

Invasive fungal infections (IFIs) are a group of diseases caused by fungal pathogens, primarily Candida, Aspergillus, and Cryptococcus, which invade the body, proliferate in deep tissues, organs, or the bloodstream, and lead to localized or systemic severe infections. These infections impose significantclinical and economic burdens due to their high mortality rates, the high cost and limited availability of antifungal drugs, and the frequent adverse effects. Invasive fungal pathogens invade the host through hyphae, spores, and secreted adhesive proteins, primarily triggering disease via signal

Neutrophil-targeted nanomedicine delivery systems: therapeutic applications and future perspectives in sepsis management.

Sepsis is a systemic organ dysfunction caused by an abnormal host infection response, and its high fatality rate is closely associated with uncontrolled inflammatory storms, immunological diseases, and multi-organ failure. As essential components of innate immunity, neutrophils play a dual role in sepsis. Initially, they protect tissues by phagocytosing pathogens and releasing antibacterial substances. As the disease progresses, however, they become over-activated and exacerbate tissue damage by triggering the release of cytokine and neutrophil extracellular traps (NETs). Nanomaterials, levera

Pentraxin family members of largemouth bass (Micropterus salmoides): Cloning, characterization and expression responses to LPS, Poly (I:C) and Nocardia seriolae.

Pentraxins (PTXs) are highly conserved soluble pattern recognition molecules, and play important roles in the innate immunity and autoimmune diseases. In present study, ten PTXs including CRP1, SAP, NPTX1-like, NPTX2a, NPTX2b, NPTXR-like, NPTXRb, PTX3a, PTX3-like and PTX4 were identified from largemouth bass (Micropterus salmoides). These PTXs exhibited high sequence identities with their counterparts of other fish species, and contained conserved motifs and structures of the PTX family, except of PTX3-like. Phylogenetic analysis revealed that these largemouth bass PTXs were closely clustered

Susceptibility of TSPAN5 polymorphisms to Kawasaki disease and immunoglobulin therapy resistance in southern Chinese children.

Kawasaki disease (KD) is a condition characterized by acute multi-system vasculitis and high fever in infants and children. Intravenous immunoglobulin (IVIG) is the established therapeutic approach of KD,foralleviating inflammation and mitigate the risk of arterial wall dilation and the development of coronary artery aneurysms (CAA). But almost 20% of the patients developed resistance to IVIG and displayed persistent fever after standard primary treatment. TSPAN5, belonging to the Tetraspanin family, has been demonstrated to modulate innate immunity in a range of human diseases. It accomplishe

Immunosuppressive Mechanisms in Brucellosis in Light of Chronic Bacterial Diseases.

Brucellosis is considered one of the major zoonoses worldwide, constituting a critical livestock and human health concern with a huge socio-economic burden. Brucella genus, its etiologic agent, is composed of intracellular bacteria that have evolved a prodigious ability to elude and shape host immunity to establish chronic infection. Brucella's intracellular lifestyle and pathogen-associated molecular patterns, such as its specific lipopolysaccharide (LPS), are key factors for hiding and hampering recognition by the immune system. Here, we will review the current knowledge of evading and immun

Trial watch: TLR3 agonists in cancer therapy.

UNLABELLED: Toll-like receptor 3 (TLR3) is a pattern recognition receptor that senses exogenous (viral) as well as endogenous (mammalian) double-stranded RNA in endosomes. On activation, TLR3 initiates a signal transduction pathway that culminates with the secretion of pro-inflammatory cytokines including type I interferon (IFN). The latter is essential not only for innate immune responses to infection but also for the initiation of antigen-specific immunity against viruses and malignant cells. These aspects of TLR3 biology have supported the development of various agonists for use as stand-al

Two novel proteins expressed by the venom glands of Apis mellifera and Nasonia vitripennis share an ancient C1q-like domain.

An in-depth proteomic study of previously unidentified two-dimensional polyacrylamide gel electrophoresis spots of honey bee (Apis mellifera, Hymenoptera) venom revealed a new protein with a C1q conserved domain (C1q-VP). BlastP searching revealed a strong identity with only two proteins from other insect species: the jewel wasp, Nasonia vitripennis (Hymenoptera), and the green pea aphid, Acyrthosiphon pisum (Hemiptera). In higher organisms, C1q is the first subcomponent of the classical complement pathway and constitutes a major link between innate and acquired immunity. Expression of C1q-VP

Dissociation of paramyxovirus interferon evasion activities: universal and virus-specific requirements for conserved V protein amino acids in MDA5 interference.

The V protein of the paramyxovirus subfamily Paramyxovirinae is an important virulence factor that can interfere with host innate immunity by inactivating the cytosolic pathogen recognition receptor MDA5. This interference is a result of a protein-protein interaction between the highly conserved carboxyl-terminal domain of the V protein and the helicase domain of MDA5. The V protein C-terminal domain (CTD) is an evolutionarily conserved 49- to 68-amino-acid region that coordinates two zinc atoms per protein chain. Site-directed mutagenesis of conserved residues in the V protein CTD has reveale

Constitutive expression of a bacterial pattern recognition receptor, CD14, in human salivary glands and secretion as a soluble form in saliva.

Источник пометил публикацию как отозванную

Saliva contains a number of proteins and glycoproteins that protect oral tissues, but little is known about the role of human saliva in innate immunity. Here we showed that human major salivary gland cells constitutively expressed a bacterial pattern recognition receptor, CD14, by immunohistochemistry. Human salivary gland cells in culture express CD14 mRNA and a 55-kDa CD14 protein in, but not on the cells, and secrete a soluble form with the same molecular mass. Human whole saliva contains a 55-kDa CD14, and the concentration of parotid saliva was 10-fold higher than whole saliva, which is c

African swine fever virus MGF_110-3L and MGF_110-4L signal TLR2-mediated lethal inflammation.

African swine fever virus (ASFV) is a lethal pathogen that triggers uncontrolled cytokine storms and severe immunopathology. However, the viral factors responsible for systemic inflammation remain unclear. Here, we show that the ASFV-encoded proteins MGF_110-3L and MGF_110-4L are secreted via the conventional ER-Golgi pathway. Single-cell RNA sequencing of porcine PBMCs revealed that MGF_110-3L preferentially activates inflammatory responses in monocytic cells. Mechanistically, both proteins bind Toll-like receptor 2 (TLR2) and signal through TLR2/TLR1 and TLR2/TLR6 heterocomplexes, with the c

The pattern recognition receptors Nod1 and Nod2 account for neutrophil recruitment to the lungs of mice infected with Legionella pneumophila.

The intracellular bacterium Legionella pneumophila induces a severe form of pneumonia called Legionnaires diseases, which is characterized by a strong neutrophil (NE) infiltrate to the lungs of infected individuals. Although the participation of pattern recognition receptors, such as Toll-like receptors, was recently demonstrated, there is no information on the role of nod-like receptors (NLRs) for bacterial recognition in vivo and for NE recruitment to the lungs. Here, we employed a murine model of Legionnaires disease to evaluate host and bacterial factors involved in NE recruitment to the m

Interaction of Bovine Peripheral Blood Polymorphonuclear Cells and Leptospira Species; Innate Responses in the Natural Bovine Reservoir Host.

Cattle are the reservoir hosts of Leptospira borgpetersenii serovar Hardjo, and can also be reservoir hosts of other Leptospira species such as L. kirschneri, and Leptospira interrogans. As a reservoir host, cattle shed Leptospira, infecting other animals, including humans. Previous studies with human and murine neutrophils have shown activation of neutrophil extracellular trap or NET formation, and upregulation of inflammatory mediators by neutrophils in the presence of Leptospira. Humans, companion animals and most widely studied models of Leptospirosis are of acute infection, hallmarked by

Circadian Control of Inflammasome Pathways: Implications for Circadian Medicine.

The innate immune system senses "non-self" molecules derived from pathogens (PAMPs) as well as endogenous damage-associated molecular patterns (DAMPs) and promotes sterile inflammation that is necessary for injury resolution, tissue repair/regeneration, and homeostasis. The NOD-, LRR- and pyrin domain containing protein 3 (NLRP3) is an innate immune signaling complex whose assembly and activation can be triggered by various signals ranging from microbial molecules to ATP or the abnormal accumulation of crystals, thus leading to IL-1β and IL-18 maturation and secretion. One-dimensional nanosonosensitizer boosted multiple branches of immune responses against MHC-deficient immune-evasive urologic tumor.

Cancer immunotherapies rely on CD8+ cytolytic T lymphocytes (CTLs) in recognition and eradication of tumor cells via antigens presented on major histocompatibility complex class I (MHC-I) molecules. However, we observe MHC-I deficiency in human and murine urologic tumors, posing daunting challenges for successful immunotherapy. We herein report an unprecedented nanosonosensitizer of one-dimensional bamboo-like multisegmented manganese dioxide@manganese-bismuth vanadate (BMMBV) to boost multiple branches of immune responses targeting MHC-I-deficient tumors.
