Cell Death in Neurodegenerative Diseases: Molecular Mechanisms and Therapeutic Targets.

Neurodegenerative diseases (NDs) are marked by selective neuronal vulnerability and progressive failure of neural circuits. Increasing evidence indicates that neuronal loss is not driven by a single terminal event, but emerges from interacting regulated cell death (RCD) programs. These programs are closely coupled to mitochondrial injury, proteostatic collapse, lysosomal stress, metabolic imbalance, glial state transitions, and chronic neuroinflammation. Yet, how distinct death pathways are organized across cell types, disease stages, and disease-specific microenvironments remains unresolved.

Mechanistic insights into sulfate-driven performance adaptation and membrane fouling in a UASB-SBR-AXMBR system: metabolic network reconstruction and microbial community succession.

Sulfate-rich wastewater poses considerable challenges to the operational stability of biological treatment systems. This study investigated the long-term (294 days) response of a combined UASB-SBR-AXMBR process to stepwise increases in sulfate concentration from 100 to 2000 mg/L. The system maintained stable carbon and nitrogen removal performance under sulfate stress, with COD removal exceeding 90.3% and total nitrogen removal stabilizing at 85.5% via a partial nitritation-anammox (PN/A) pathway. However, high sulfate loading significantly intensified membrane fouling, with the primary drivin

Decoding the cold: integrated secretome profiling reveals extracellular adaptation mechanisms in Glaciozyma antarctica.

Glaciozyma antarctica is an Antarctic psychrophilic yeast providing an excellent eukaryotic model for dissecting molecular mechanisms of cold adaptation. Yet, its extracellular protein repertoire, the interface between the cell and extreme surroundings, remains unexplored. Here, we present the first integrated bioinformatics-proteomics secretome map of G. antarctica, linking computational prediction with experimental validation under varying temperature stress, yielding 9 high-confidence experimentally supported secreted proteins. Three complementary pipelines (Phobius, SignalP/TMHMM, and WoLF

Strain-Adaptive Micro-Wrinkled Carbon Films Enable Flexible High-Fidelity Intelligent Health Monitoring.

Wrinkled microstructures provide an effective strategy for constructing stable and low-noise soft bioelectronic interfaces, yet their large-area, continuous, and controllable fabrication remains challenging. Here, we report a large-area continuous ultrathin micro-wrinkled film that rapidly forms wrinkle microstructures through interfacial stress instability at a rate of 70 µm·s-1. The film exhibits excellent geometric deformability and mechanical compliance, enabling effective relaxation of local stress concentration at dynamic interfaces. Flexible strain sensors based on this microstructure a

The Neurospora crassa Pangenome: A Robust Framework for Population-Scale Analysis and Structural Variant Discovery.

Neurospora crassa is a widely distributed ascomycete with high genetic diversity, yet reliance on limited reference genomes has hindered a comprehensive understanding of its genetic landscape. To address this limitation, we integrated the functional annotation of the FGSC2225 genome with a comprehensive comparative genomic analysis of N. crassa strains. FGSC2225 gene and transposable element (TE) proportions mirrored those of FGSC2489, though TE levels were significantly higher than those in sister species Sordaria macrospora. Phylogenetic analysis resolved the N. crassa population into two pr

Combined inhibition of de novo pyrimidine synthesis and ATR promotes ATF4-mediated cell death in TP53-deficient ALL.

Mutations or deletions affecting the TP53 gene predict a dismal outcome in relapsed acute lymphoblastic leukemia (ALL). Loss of p53 function compromises the response to many anti-leukemic therapies, underscoring the need for agents that are effective in TP53-deficient cells. Leukemic blasts depend on de novo pyrimidine synthesis to sustain proliferation, and the mitochondrial enzyme dihydroorotate dehydrogenase (DHODH) is essential for this pathway. While DHODH inhibitors (DHODHis) show preclinical anti-leukemic activity, the efficacy in TP53-deficient contexts has not been tested. Using both

Targeting HMGB1 for Renal Ischemia and Reperfusion Injury: Mechanisms and Therapeutic Strategies.

Although the underlying mechanisms of renal IRI have been extensively studied, the corresponding effective treatments are still lacking. HMGB1, an important nuclear factor that is secreted outside cells when experiencing stress conditions, acts as a DAMP and exerts crucial effects on renal IRI. Many studies have suggested that the effect of HMGB1 on kidney damage is mediated mainly through the interaction of HMGB1 with pattern recognition receptors such as TLR4 and RAGE, which then results in the aggravation of local inflammatory response, increased infiltration of leukocytes, and finally rena

Hepatotoxic effect of DEP in zebrafish is via oxidative stress, genotoxicity, and modulation of molecular pathways.

Diethyl phthalate (DEP) is a commonly used phthalate ester (PAE) plasticizer known for its excellent plasticity and flexibility. Environmental exposure to DEP may have adverse effects on human health. Currently, few studies have focused on the specific effects and mechanisms on aquatic organisms. This study investigated the effects of oxidative stress effects and genotoxicity of DEP on zebrafish liver, as well as molecular interactions with antioxidant enzymes both in vitro and in vivo. The results revealed that exposure to DEP led to changes in the activity of antioxidant enzymes, which may b

Hydrogen sulfide mitigates the immunotoxicity and neurotoxicity induced by cadmium in Tegillarca granosa.

The blood clam Tegilarca granosa is an economically important shellfish in Asia. The complex living environment often exposes the clams to cadmium (Cd) and hydrogen sulfide (H2S). H2S is well-known for its toxic effects, however, it is still unclear whether it can participate in regulating the heavy metal adaptation in clams. Here, we subjected T. granosa to chronic Cd stress (50 μg/L, 14 days) to explore their physiological and molecular response patterns under the addition of 0.2, 0.4, and 0.8 mM H2S. The results indicated that the Cd stress induced the immune-toxicity and neurotoxicity in t

Network Pharmacology Insights Into Statin-Induced Coenzyme Q10 Deficiency: Lipid Metabolic Crosstalk, TNF-MAPK Signaling, and Muscle Toxicity.

This in silico network-pharmacology investigation delineates the molecular interplay linking statin-induced Coenzyme Q10 (CoQ10) deficiency to statin-associated muscle symptoms (SAMS). GeneCards-derived targets related to SAMS, CoQ10 deficiency, and statins were integrated, and the intersecting gene set was analyzed through STRING-based protein-protein interaction mapping, followed by hub-gene prioritization using CytoHubba. Functional enrichment via ShinyGO revealed that the 145 common genes converge predominantly on inflammatory, metabolic, and mitochondrial pathways. Tumor necrosis factor (

aEEG and neurologic exam findings correlate with hypoxic-ischemic brain damage severity in a piglet survival model.

BACKGROUND: Newborn pigs offer theoretical advantages for studying newborn hypoxic-ischemic (HI) brain damage because of a development and structure similar to the human brain. However, the correlation between functional features and actual HI brain damage has not been reported. METHODS: Newborn pigs were examined daily for 3 days after a HI insult using amplitude-integrated EEG (aEEG), and a neurobehavioral score enriched with stress and social and object interaction-driven activity evaluation. Brain damage was then assessed using histologic, immunohistochemical, and proton magnetic resonanc

Mitochondria-Endoplasmic Reticulum Crosstalk in Pancreatic β-Cell Stress: A Driver of Diabetes.

Pancreatic β-cell failure is central to the pathogenesis of type 2 diabetes (T2D), developing when chronic metabolic stress overwhelms the adaptive capacity of these cells. Because insulin biosynthesis and glucose-stimulated secretion depend on tightly coordinated organelle function, crosstalk between the endoplasmic reticulum (ER) and mitochondria strongly influences β-cell fate. The two organelles are physically coupled at mitochondria-associated membranes (MAMs), contact sites that synchronize ER Ca2+ release with mitochondrial Ca2+ uptake and adenosine triphosphate (ATP) production, and th

A Cu/ZIF-8-based flexible sensor for real-time monitoring of protocatechuic acid in plants under environmental stress.

Understanding the dynamic regulation of endogenous metabolites in plants under environmental stress is essential for elucidating plant adaptive mechanisms and improving crop resilience. However, conventional analytical methods are typically destructive, time-consuming, and lack the capability for real-time monitoring, thereby limiting the investigation of in-vivo biochemical dynamics in plants. In particular, the in-situ, non-invasive detection of small-molecule regulators such as protocatechuic acid (PCA) remains a significant challenge. Herein, we report a wearable electrochemical sensing pl

Transcriptome Analysis Reveals the Mechanism by which Probiotic Alleviate the Response of Juvenile American Shad (Alosa sapidissima) to High Temperatures.

Probiotics show great potential in mitigating the impacts of temperature stress on fish. American shad (Alosa sapidissima), a species highly sensitive to high temperatures, served as an ideal model for studying fish adaptation to climate change. This study integrated biochemical and hepatic transcriptomic analyses to demonstrate that the probiotic Lactococcus lactis L103 alleviated thermal stress via a regulatory network. The findings revealed that probiotic-mediated upregulation of irf1b and nfil3-2 suppressed pro-inflammatory responses, as evidenced by significantly reduced AST and ALT activ
