Integration of hepatic lipidomics and transcriptomics reveals dysregulation of lipid metabolism in a golden hamster model of visceral leishmaniasis.

Visceral leishmaniasis (VL), the most severe form of leishmaniasis, remains a significant public health concern that cannot be overlooked in underdeveloped regions. Studies suggest that lipids play a crucial role in the survival of Leishmania parasites in mammalian hosts. However, a comprehensive understanding of the characteristics and underlying mechanisms of lipid metabolism in VL hosts is lacking. In this study, we conducted lipidomic and transcriptomic analyses of liver tissues from VL golden hamsters at 12 weeks post-infection (WPI) and performed integrated analysis. Simultaneously, qPCR

Nutrition and ulcerative colitis.

The role of diet in the aetiology and pathogenesis of ulcerative colitis (UC) remains uncertain. Impaired utilization by colonocytes of butyrate, a product of bacterial fermentation of dietary carbohydrates escaping digestion, may be important. Sulphur-fermenting bacteria may be involved in this impaired utilization. Oxidative stress probably mediates tissue injury but is probably not of causative importance. Patients with UC are prone to malnutrition and its detrimental effects. However, there is no role for total parenteral nutrition and bowel rest as primary therapy for UC. The maintenance

An in vitro mechanistic study of cholinergic-associated mechanisms relevant to MASLD in HepG2 cells.

BACKGROUND/AIM: Polyunsaturated fatty acids (PUFAs) and the hepatic cholinergic axis are critical regulators of lipid metabolism and inflammatory signaling; however, their time-dependent interplay in hepatocytes remains incompletely understood. Clarifying how n-6 and n-3 PUFAs interact with cholinergic pathways may provide mechanistic insight into metabolic dysfunction-associated steatotic liver disease (MASLD). MATERIALS AND METHODS: HepG2 cells were used to examine the combined effects of butyrylcholinesterase (BChE) overexpression and PUFA exposure. Cells were treated with linoleic acid (L

Ursolic acid improves growth performance, intestinal health, and antioxidant status in broilers by regulating lipid metabolism and the KEAP1-NRF2 pathway.

In response to the urgent need for alternatives to antibiotics in poultry production, this study evaluated the effects of ursolic acid (UA), a plant-derived triterpenoid, on growth performance, metabolism, intestinal function, and antioxidant capacity in broilers. A total of 320 Cobb broilers were randomly divided into four groups: a control group and three groups supplemented with UA at 0, 50, 200, or 400 mg/kg of diet, with 8 replicates of 10 birds each, over a 42-day period. Body weight and feed intake were measured on days 21 and 42. On day 42, samples were collected for enzyme-linked immu

Organic chromium derived from the chelation of Ganoderma lucidum polysaccharide and chromium (III) alleviates metabolic syndromes and intestinal microbiota dysbiosis induced by high-fat and high-fructose diet.

Organic chromium is of great interest and has become an important chromium supplement resource in recent years because of its low toxicity and easy absorption. In our previous study, we synthesized a novel organic chromium [GLP-Cr] through the chelation of Ganoderma lucidum polysaccharide and chromium (III). The purpose of this study was to investigate the beneficial effects of GLP-Cr on the improvement of metabolic syndromes (MetS) in mice fed with a high-fat and high-fructose diet (HFHFD) and its mechanism of action. The results indicated that oral administration of GLP-Cr inhibited the exce

Comparative serum metabolomics between SCID mice and BALB/c mice with or without Schistosoma japonicum infection: Clues to the abnormal growth and development of schistosome in SCID mice.

The small blood flukes of genus Schistosoma, which cause one of the most prevalent and serious parasitic zoonosis schistosomiasis, are dependent on immune-related factors of their mammalian host to facilitate their growth and development, and the formation of granulomatous pathology caused by eggs deposited in host's liver and intestinal wall. Schistosome development is hampered in the mice lacking just T cells, and is even more heavily retarded in the severe combined immunodeficient (SCID) mice lacking both T and B lymphocytes. Nevertheless, it's still not clear about the underlying regulator

A multi-omics investigation reveals the hepatic response to salinity stress in grass carp (Ctenopharyngodon idella).

In the context of increasing freshwater scarcity and the effects of climate change on aquatic environments, developing salt-tolerant fish strains has become a critical strategy for sustainable aquaculture. The limited availability of suitable species for saline-water aquaculture poses a significant challenge, severely impacting the development of the fishery economy. This study investigated the effects of 24 h exposure to freshwater, and seawater at salinities of 4, 9 ppt and 12 ppt, on juvenile grass carp (Ctenopharyngodon idella). Following these exposures, liver tissues were collected to as

Uremic pruritus.

Uremic pruritus or chronic kidney disease-associated pruritus (CKD-aP) remains a frequent and compromising symptom in patients with advanced or end-stage renal disease, strongly reducing the patient's quality of life. More than 40% of patients undergoing hemodialysis suffer from chronic pruritus; half of them complain about generalized pruritus. The pathogenesis of CKD-aP remains obscure. Parathormone and histamine as well as calcium and magnesium salts have been suspected as pathogenetic factors. Newer hypotheses are focusing on opioid-receptor derangements and microinflammation as possible c

Transport mechanisms for long-chain polyunsaturated fatty acids in the human placenta.

To understand the placental role in the processes responsible for the preferential accumulation of maternal long-chain polyunsaturated fatty acids (LCPUFAs) in the fetus, we investigated fatty acid uptake and metabolism in the human placenta. A preference for LCPUFAs over nonessential fatty acids has been observed in isolated human placental membranes as well as in BeWo cells, a human placental choriocarcinoma cell line. A placental plasma membrane fatty acid binding protein (p-FABP(pm)) with a molecular mass of approximately 40 kDa was identified. The purified p-FABP(pm) preferentially bound

Metabolic pathways and chemotherapy resistance in acute myeloid leukemia (AML): Insights into Enoyl-CoA hydratase domain-containing protein 3 (ECHDC3) as a potential therapeutic target.

BACKGROUND: Acute myeloid leukemia (AML) is characterized by high relapse and mortality rates. Our previous investigation identified enoyl-CoA hydratase domain-containing protein 3 (ECHDC3) as being of prognostic significance in AML; however, the underlying pathways remain elusive. The intricate crosstalk among genetic abnormalities, metabolic pathways, and protein dysfunctions underpins the complexity contributing to its poor prognosis. METHODS: ECHDC3 was knocked down by siRNA and subjected to RNA sequencing, chromatin immunoprecipitation (ChIP), and metabolomic assessments. Cell culture me

Carbonic anhydrase 8 (CAR8) negatively regulates GLP-1 secretion from enteroendocrine cells in response to long-chain fatty acids.

Glucagon-like peptide-1 (GLP-1) is an incretin secreted from enteroendocrine preproglucagon (PPG)-expressing cells (traditionally known as L cells) in response to luminal nutrients that potentiates insulin secretion. Augmentation of endogenous GLP-1 secretion might well represent a novel therapeutic target for diabetes treatment in addition to the incretin-associated drugs currently in use. In this study, we found that PPG cells substantially express carbonic anhydrase 8 (CAR8), which has been reported to inhibit inositol 1,4,5-trisphosphate (IP3) binding to the IP3 receptor and subsequent Ca2

Integrated Multi-Omics Analysis Reveals Molecular Features Associated with Energy Metabolism Adaptations in Brooding Taihe Black-Bone Silky Fowls.

Broodiness is a natural behavior whereby female birds stop laying eggs to sit on and hatch them. This behavior is regulated through genetic, hormonal, and environmental factors. The Taihe Black-Boned Silky Fowl (TBSF), a Chinese traditional domestic breed, exhibits a strong brooding tendency; however, the molecular mechanisms underlying this trait remain unclear. In this study, we performed an integrated multi-omics analysis to characterize differences between two groups of TBSF hens: 8 individuals undergoing 30 days of active brooding (BR30) and 8 individuals in the normal laying egg stage (N

Qijiao Shengbai capsule ameliorates ulcerative colitis in mice by modulating gut microbiota and protecting the intestinal barrier, associated with suppression of the TLR4/MyD88/NF-κB pathway.

BACKGROUND: Qijiao Shengbai capsule (QJSB) is a classic traditional Chinese compound formulation composed of seven herbal ingredients, primarily used clinically to treat leukopenia. Individual herbs and their active components within QJSB demonstrate favorable efficacy against ulcerative colitis (UC). Nevertheless, no evidence exists of QJSB being a full-form prescription for treating UC. METHODS: This study identified the chemical composition of QJSB using UHPLC-ESI-Q-Exactive Plus Orbitrap-MS. At the same time, a mouse model of ulcerative colitis (UC) was established using DSS to examine th
