Preliminary evidence of the presence of lymphatic vessels immunoreactive for D2-40 and Prox-1 in human pterygium.

Human pterygium is a benign fibrovascular outgrowth of the corneo-conjunctival junction, characterized by tissue remodeling, cellular proliferation, angiogenesis and inflammation. No data are available concerning the presence of lymphatic vessels in this pathological condition. The aim of this study was to evaluate by immunohistochemistry, using antibodies against D2-40, Prox-1 and Ki-67, the presence and the proliferative activity of lymphatic vessels in human pterygium. An increased lymphatic microvessel density was observed in the human pterygium compared to the normal conjunctiva. Moreover

Prognostic indicators for squamous cell carcinoma of the oral cavity: a clinicopathologic correlation.

Fifty-three patients with T1 squamous cell cancer of the floor of mouth and ventral surface of the tongue with a known clinical outcome were retrospectively analyzed and arbitrarily divided into "aggressive" and "nonaggressive" groups based on their clinical behavior. Various host and tumor factors were then evaluated in an attempt to determine whether the tumor behavior could have been predicted. The paraffin-embedded tumor specimens were evaluated for tumor differentiation, tumor thickness and tumor invasion, microvessel density, and p53 expression. In addition, a composite morphologic gradi

Mesenchymal stem/stromal cells overexpressing leukemia inhibitory factor (LIF) promote arteriogenesis and functional recovery in a mouse model of critical hindlimb ischemia.

BACKGROUND: Critical limb ischemia (CLI) is the most severe manifestation of peripheral arterial disease (PAD), for which there are still no effective therapeutic options. Mesenchymal stem/stromal cells (MSCs) can secrete bioactive molecules capable of promoting tissue regeneration and neovascularization. The therapeutic potential of MSCs can be enhanced by genetic modification to overexpress molecules of interest. Here, we produced an umbilical cord-derived MSC line overexpressing leukemia inhibitory factor (LIF), MSC_LIF, and evaluated its pro-angiogenic/arteriogenic potential. METHODS: MSC

Muscone abrogates breast cancer progression through tumor angiogenic suppression via VEGF/PI3K/Akt/MAPK signaling pathways.

BACKGROUND: Angiogenesis strongly reflects poor breast cancer outcome and an important contributor to breast cancer (BC) metastasis; therefore, anti-angiogenic intervention is a potential tool for cancer treatment. However, currently used antibodies against vascular endothelial growth factor A (VEGFA) or inhibitors that target the VEGFA receptor are not effective due to weak penetration and low efficiency. Herein, we assessed the anti-BC angiogenic role of muscone, a natural bioactive musk constituent, and explored possible anti-cancer mechanisms of this compound. METHODS: CCK-8, EdU, scratch

Early elevations in arterial pressure: a contributor to rapid depressive symptom emergence in female Zucker rats with metabolic disease?

One of the growing challenges to public health and clinical outcomes is the emergence of cognitive impairments, particularly depressive symptom severity, because of chronic elevations in metabolic disease and cerebrovascular disease risk. To more clearly delineate these relationships and to assess the potential for sexual dimorphism, we used lean (LZR) and obese Zucker rats (OZR) of increasing age to determine relationships between internal carotid artery (ICA) hemodynamics, cerebral vasculopathies, and the emergence of depressive symptoms. Male OZR exhibited progressive elevations in perfusio

The Effect of Short-Term Exposure to Cadmium on the Expression of Vascular Endothelial Barrier Antigen in the Developing Rat Forebrain and Cerebellum: A Computerized Quantitative Immunofluorescent Study.

Clinical and laboratory studies have shown that environmental exposure to cadmium produces damage to several organs, including bones, lungs, and kidneys. The involvement of cadmium in central nervous system (CNS) disorders has also been widely reported, but the precise pathophysiological mechanism is not yet fully understood. Children who were exposed to cadmium during pregnancy are known to suffer from developmental delays, learning difficulties, attention deficit hyperactivity disorder (ADHD), and other cognitive and neurobehavioral deficits. Results from numerous studies suggest that dysfun

Complex genetic and histopathological study of 15 patient-derived xenografts of aggressive lymphomas.

Non-Hodgkin lymphomas (NHL) represent the most common hematologic malignancies. Patient-derived xenografts (PDXs) are used for various aspects of translational research including preclinical in vivo validation of experimental treatment approaches. While it was repeatedly demonstrated that PDXs keep majority of somatic mutations with the primary lymphoma samples, from which they were derived, the composition of PDX tumor microenvironment (TME) has not been extensively studied. We carried out a comparative genetic and histopathological study of 15 PDX models derived from patients with various ty

Weipixiao attenuate early angiogenesis in rats with gastric precancerous lesions.

BACKGROUND: Angiogenesis is a pathobiological hallmark of gastric cancer. However, rare studies focus on angiogenesis in gastric precancerous lesions (GPL). Weipixiao (WPX), a Chinese herbal preparation, is proved clinically effective in treating GPL. Here, we evaluated WPX's anti-angiogenic potential for GPL, and also investigated the possibility of its anti-angiogenic mechanisms. METHODS: HPLC analysis was applied to screen the major chemical components of WPX. After modeling N-methyl-N'-nitro-N-nitrosoguanidine (MNNG)-induced GPL in male Sprague-Dawley rats, different doses of WPX were adm

TGF-β signaling promotes tumor vasculature by enhancing the pericyte-endothelium association.

BACKGROUND: The breast cancer microenvironment promotes tumor vascularization through the complex interactions involving tumor-associated fi … components. RESULTS: Our studies revealed that TAFs significantly increase the lumen size of blood microvessels. Inactivation of TGF-β signaling in tumor cells by dnTGFBR1 reduced the microvessel density and lumen sizes, decreasing tumor growth. In contrast, caTGFBR1-tumors exhibited greater vessel density and lumen sizes. Tumors with inactive dnTGFBR1 showed …

Naringin inhibits vascular endothelial cell apoptosis via endoplasmic reticulum stress‑ and mitochondrial‑mediated pathways and promotes intraosseous angiogenesis in ovariectomized rats.

In this study, to investigate the effects of naringin on vascular endothelial cell (VEC) function, proliferation, apoptosis, and angiogenesi … r density assessment showed that the naringin treatment groups had a significantly higher number of microvessels than the OVX group, and that microvascular density was positively correlated with BMD. In summary, naringin inhibits apoptosis in VECs by blocking the endoplasmic reticulum (ER) stress‑ and mitochondrial‑mediated pathways. Naringin also regulates en …

SPARC suppresses lymph node metastasis by regulating the expression of VEGFs in ovarian carcinoma.

Lymph node metastasis is one of the most valuable determinants for the prognosis of ovarian cancer. However, the molecular mechanisms underlying lymphangiogenesis in ovarian cancer is still poorly understood. Secreted protein acidic and rich in cysteine (SPARC), a Ca2+-binding matricellular glycoprotein that modulates cell adhesion, migration and differentiation, is thought to play a decisive role in tumor metastasis. Vascular endothelial growth factor (VEGF)-C and VEGF-D contributes to tumor-associated lymphatic vessel growth, enhancing the metastatic spread of tumor cells to lymph nodes. The

Scrib organizes cortical actomyosin clusters to maintain adherens junctions and angiogenic sprouting.

Spatiotemporal control of adherens junction fluidity and integrity is critical for angiogenesis, but underlying mechanisms are incompletely understood. To identify unappreciated regulators of endothelial adherens junctions, we performed VE-cadherin proximity ligation mass spectrometry, revealing significant interaction with the multifunctional scaffold Scrib. Utilizing a 3D angiogenesis-on-chip model, we find Scrib-depleted microvessels generate reduced intact sprouts and increased single-cell detachments. This defect was characterized by adherens junction instability and decreased actomyosin …

RBC deformability and clinical relevance in transfusion recipients.

Red blood cell (RBC) deformability-the ability of RBCs to change shape-is crucial for their passage through microvessels and effective oxygen delivery. Deformability depends on the cell's surface area-to-volume ratio, internal viscosity, and membrane elasticity. Deformability is assessed using single-cell methods like micropipette aspiration and bulk flow techniques such as ektacytometry. RBC deformability decreases during cold storage due to biochemical and structural changes known as storage lesions. Additional processes like cryopreservation and irradiation further impair the deformability …
