Traditional air dispersion modeling usually relies on deterministic frameworks that use multiple conservative assumptions as inputs. For example, atmospheric pollutant concentrations are often overestimated by using maximum emission rates for point sources, an approach recommended by regulatory agencies that may not reflect typical operating conditions, especially for sources with variable emissions. To better understand how emission variability affects modeled pollutant concentrations, this study presents a novel probabilistic modeling framework designed to estimate pollutant concentrations f

Shared-parameter joint modeling is a useful technique for properly associating longitudinal and time-to-event data. When the interest is in the survival outcome, the conditional logarithm of the hazard function for an event is conventionally presumed to be linearly related over time to a set of explanatory covariates, among other terms. However, this hypothesis is quite restrictive and may yield misleading results. Our objective here is to easily check such a modeling assumption for any continuous fixed covariate. For this purpose, we examine the appropriateness of a nonparametric test criteri

There is an extensive literature on methods for meta-analysis of diagnostic test accuracy, but it mainly focuses on a single test. A multinomial generalised linear mixed model was recently proposed for the joint meta-analysis of studies comparing two tests on the same participants in a paired tests design with a gold standard. In this setting, we propose a novel model for joint meta-analysis of studies comparing two diagnostic tests which assumes independent multinomial distributions for the counts of each combination of test results in diseased and non-diseased patients, conditional on the la

In mid-2022, New York City (NYC) became the epicenter of the US mpox outbreak. We provided real-time mpox case forecasts to the NYC Department of Health and Mental Hygiene to aid in outbreak response. Forecasting methodologies evolved as the epidemic progressed. Initially, lacking knowledge of at-risk population size, we used exponential growth models to forecast cases. Once exponential growth slowed, we used a Susceptible-Exposed-Infectious-Recovered (SEIR) model. Retrospectively, we explored if forecasts could have been improved using an SEIR model in place of our early exponential growth mo

Understanding the temporal relationship between key events in an individual's infection history is crucial for disease control. Delay data between events, such as infection and symptom onset times, is doubly censored because the exact time at which these key events occur is generally unknown. Current mathematical models for delay distributions are derived from heuristic justifications. Here, we derive a new model for delay distributions, specifically for incubation periods, motivated by bacterial-growth dynamics that lead to the Burr family of distributions being a valid modelling choice. We a

Traditional Hardy-Weinberg equilibrium (HWE) tests (the χ2 test and the exact test) have long been used as a metric for evaluating genotype quality, as technical artifacts leading to incorrect genotype calls often can be identified as deviations from HWE. However, in data sets composed of individuals from diverse ancestries, HWE can be violated even without genotyping error, complicating the use of HWE testing to assess genotype data quality. In this manuscript, we present the Robust Unified Test for HWE (RUTH) to test for HWE while accounting for population structure and genotype uncertainty,

Dengue fever (DF) is one of the most common and rapidly spreading mosquito-borne viral diseases in tropical and subtropical regions. In recent years, this imported disease has posed a serious threat to public health in China, especially in the Pearl River Delta (PRD). Although the severity of DF outbreaks in the PRD is generally associated with known risk factors, fine scale assessments of areas at high risk for DF outbreaks are limited. We built five ecological niche models to identify such areas including a variety of climatic, environmental, and socioeconomic variables, as well as, in some

Despite the availability of vaccines, influenza remains a major public health challenge. A key reason is the virus capacity for immune escape: ongoing evolution allows the continual circulation of seasonal influenza, while novel influenza viruses invade the human population to cause a pandemic every few decades. Current vaccines have to be updated continually to keep up to date with this antigenic change, but emerging 'universal' vaccines-targeting more conserved components of the influenza virus-offer the potential to act across all influenza A strains and subtypes. Influenza vaccination prog

Accurate modeling of the high strain-rate response of healthy human knee cartilage is critical to investigating the mechanism(s) of knee osteoarthritis and other cartilage disorders. Osteoarthritis has been suggested to originate from regional shifts in joint loading during walking and other high strain-rate physical activities. Tibial plateau cartilage under compression rates analogous to walking exhibits a non-linear and location-dependent mechanical response. A constitutive model of cartilage that efficiently predicts the non-linear and non-uniform high strain-rate mechanics of tibial plate

A new cut-off criterion has been proposed for the selection of uninformative variables prior to chemometric partial least squares (PLS) modelling. After variable elimination, PLS regressions were made and assessed comparing the results with those obtained by PLS models based on the full spectral range. To assess the prediction capabilities, uninformative variable elimination (UVE)-PLS and PLS were applied to diffuse reflectance near-infrared spectra of heroin samples. The application of the proposed new cut-off criterion, based on the t-Students distribution, provided similar predictive capabi

The use of nordihydroguaiaretic acid (NDGA)-polymerized collagen fibers as a novel local drug delivery system is introduced. The drug loading of these biocompatible fibers is illustrated with the anti-inflammatory agents dexamethasone and dexamethasone 21-phosphate. Capillary zone electrophoresis was used to measure the amount of drug released from the fibers into phosphate buffered saline with time. From these measurements and the use of a mathematical model, we were able to determine the diffusion coefficients for dexamethasone (D = 1.86 x 10(-14) m2/s) and dexamethasone 21-phosphate (D = 2.

For survival data regression, the Cox proportional hazards model is the most popular model, but in certain situations the Cox model is inappropriate. Various authors have proposed the proportional odds model as an alternative. Yang and Prentice recently presented a number of easily implemented estimators for the proportional odds model. Here we show how to extend the methods of Yang and Prentice to a family of survival models that includes the proportional hazards model and proportional odds model as special cases. The model is defined in terms of a Box-Cox transformation of the survival funct

BACKGROUND AND OBJECTIVE: The utility of predictive models depends on their external validity, that is, their ability to maintain accuracy when applied to patients and settings different from those on which the models were developed. We report a simulation study that compared the external validity of standard logistic regression (LR1), logistic regression with piecewise-linear and quadratic terms (LR2), classification trees, and neural networks (NNETs).
METHODS: We developed predictive models on data simulated from a specified population and on data from perturbed forms of the population not

A striking feature of lymphatic filariasis is the considerable heterogeneity in infection burden observed between hosts, which greatly complicates the analysis of the population dynamics of the disease. Here, we describe the first application of the moment closure equation approach to model the sources and the impact of this heterogeneity for macrofilarial population dynamics. The analysis is based on the closest laboratory equivalent of the life cycle and immunology of infection in humans--cats chronically infected with the filarial nematode Brugia pahangi. Two sets of long-term experiments a

This paper considers the joint modeling of longitudinal covariates and partly interval-censored time-to-event data. Longitudinal time-varying covariates play a crucial role in obtaining accurate clinically relevant predictions using a survival regression model. However, these covariates are often measured at limited time points and may be subject to measurement error. Further methodological challenges arise from the fact that, in many clinical studies, the event times of interest are interval-censored. A model that simultaneously accounts for all these factors is expected to improve the accura

Multistate models were developed to model survival data where several midpoints and endpoints are of interest; and they have been particular successful in modeling dynamics of cancer. As in any statistical model, identification of influential observations is an essential task, as they can significantly affect the validity of inferred parameters and conclusions drawn from the data. The local influence approach is a set of methods designed to detect the effect of small perturbations of the model or data on the inference, allowing for deeper data analysis. In this paper, we derive local influence

Prediction model selection and assessment are primary objectives of many statistical analyses. Covariance-based penalty estimators provide analytic estimates of the optimism associated with naive training error estimates for multiple classes of prediction models and error assessment rules. While the majority of work on covariance-based penalties has focused on prediction for uncensored data, little attention has been given to time-to-event data. In this article, we consider estimating the optimism for survival prediction models assessed via the Brier score. We first analytically derive an expr

In clinical trials, it is often of interest to know whether treatment works differently for some groups than others, known as heterogeneity of treatment effect. Such subgroup analysis is complicated to conduct because trials are typically not powered to find subgroups. Furthermore, it is difficult to identify characteristics of patients pertaining to such subgroups. In this article, we propose a semiparametric mixture model to identify subgroups with time-to-event outcomes. Specifically, we assume a proportional hazards model with subgroup-specific piecewise constant baseline hazards, where th

Independent censoring is a key assumption usually made when analyzing time-to-event data. However, this assumption is difficult to assess and can be problematic, particularly in studies with disproportionate loss to follow-up due to adverse events. This paper addresses the challenges associated with dependent censoring by introducing a likelihood-based approach for analyzing bivariate survival data under dependent censoring. A flexible Joe-Hu copula is used to formulate the interdependence within the quadruple times (two events and two censoring times). The marginal distribution of each event/

BACKGROUND: In Korea, Hand-foot-and-mouth disease (HFMD) is a recurring illness that presents significant public health challenges, primarily because of its unpredictable epidemic patterns. The accurate prediction of the spread of HFMD plays a vital role in the effective management of the disease.
METHODS: We have devised a dynamic model that accurately represents the transmission dynamics of HFMD. The model includes compartments for susceptible, exposed, inpatients, outpatients, recovered, and deceased individuals. By utilizing monthly inpatient and outpatient data, the ensemble Kalman filte
