The arotinoid Ro 15-0778 (temarotene) is a third generation retinoid without a polar end-group. Established, palpable and measurable rat mammary tumours, chemically induced by oral administration of 12 mg/animal 7,12-dimethylbenz[a]anthracene, were treated with Ro 15-0778 as a feed-admix in daily doses of 100 mg/kg for 6 weeks and 200 and 400 mg/kg for 9 weeks. Sprague-Dawley rats received subcutaneous injections of bis-2-oxopropyl)-nitrosamine (BOPN) once weekly for life. The animals developed mainly papillomas and squamous cell carcinomas of the nasal cavity, follicle cell carcinomas of the thyroid gland and papillomas and transitional cell carcinomas of renal pelvis, ureter and urinary bladder. The tumour incidence was 100% in all the 3 treated groups. Survival time, tumour latency and multiplicity showed dose dependency.

A cell line derived from a human hepatoblastoma, HepG2, was examined for its ability to activate cyclophosphamide (CY) to a genotoxic form. Metabolism of CY to genotoxic product(s) was determined by the induction of sister-chromatid exchanges (SCE). The inhibitory effects of substituted nitro- and sulphobenzofurazans on DNA, RNA and protein synthesis were compared in a new malignant fibrosarcoma cell line at 37 degrees C and 41 degrees C. Studies were conducted to explore the effects of glucocorticoid hormones on the regulation of the metabolism of retinol-binding protein (RBP) by H4II EC4 rat hepatoma cells in culture. Metastatic cancer has a poor prognosis. Novel pharmacologic targets need to be identified. The receptor for advanced glycation endproducts (RAGE) is a pattern recognition receptor constitutively expressed in the lungs.
