Characterization of the AP2/ERF gene family in Fritillaria thunbergii and functional evaluation of FtAP2/ERF-61 in conferring tolerance to drought and salt stress.

BACKGROUND: The APETALA 2/ethylene-responsive element binding factors (AP2/ERFs) are crucial modulators of plant development and stress responses. Fritillaria thunbergii is an important medicinal herb with considerable therapeutic efficacy. However, despite its importance, no systematic identification and functional characterization of the AP2/ERF gene family in F. thunbergii has been reported to date. RESULTS: This study identified a total of 71 AP2/ERF genes in F. thunbergii and classified them into 11 subgroups, with genes in the same subgroup exhibiting similar motifs and domains. Express

Integrated Transcriptomic and Metabolomic Profiling Reveals the Involvement of the miR397-5p-SbLAC14 Module in Condensed Tannin Accumulation in Developing Sorghum Seeds.

Sorghum seeds accumulate substantial amounts of condensed tannins (CTs), which are also referred to as proanthocyanidins (PAs), contributing to their characteristic astringent taste. Flavan-3-ol polymers, known as PAs, are sequestered within plant vacuoles and become catalytically activated via laccase enzymes. However, the biological roles and regulatory pathways of laccases in sorghum are still largely unclear. Here, integrated transcriptomic and metabolomic profiling of developing sorghum seeds identified 7942 differentially expressed genes between low- and high-CT lines, with Kyoto Encyclo

Functional Analysis of the Heat-Tolerance Candidate Gene BcNCED3 in Non-Heading Chinese Cabbage (Brassica campestris ssp. chinensis).

Non-heading Chinese cabbage (Brassica campestris ssp. chinensis) is an economically important leafy vegetable whose quality and yield are severely constrained by high temperature. Abscisic acid (ABA) is a key stress hormone, and 9-cis-epoxycarotenoid dioxygenase (NCED) is the rate-limiting enzyme of its biosynthesis; however, the function of NCED in the thermotolerance of this crop remains unclear. In this study, the heat-tolerance candidate gene BcNCED3 was functionally characterized in non-heading Chinese cabbage using bioinformatics, subcellular localization, expression profiling, exogenous

PodNet: Pod real-time instance segmentation in pre-harvest soybean fields.

Noninvasive analysis of pod phenotypic traits under field conditions is crucial for soybean breeding research. However, previous pod phenotyping studies focused on postharvest materials or were limited to indoor scenarios, failing to generalize to real-field environments. To address these issues, this paper employs an instance segmentation approach for the precise extraction of the pod area from multiplant RGB images in preharvest soybean fields. We first introduce a cost-effective workflow for constructing datasets of densely planted crop images with a uniform backdrop. Starting with video re

Nitric oxide-induced Na+/H+ exchange activity confers salt tolerance in pea (Pisum sativum L.) mesophyll cells.

Salinity stress severely hinders global agricultural productivity, and the issue will only increase under current climate scenarios. Due to complexity of salinity tolerance traits, crop breeding for salt tolerance remains a highly challenging task. The exogenous application of growth regulators, such as nitric oxide (NO), is considered a viable practical alternative to boost crop yield and quality under conditions of soil salinity. Numerous papers reported beneficial role of exogenous NO application on plant growth under salt stress, but very few explored the mechanistic basis of this process.

Has the Striga problem been solved? A field perspective critique of recent progress.

Three root-parasitic witchweed (Striga) species can cause up to total loss of grain and legume crops for millions of farmers in Africa. The damage is initiated before Striga emerges from the soil. Four decades of resistance breeding, especially maize, have at best conferred localized moderate resistance that has not greatly increased yields. Recent innovative basic research has led to major advances that are herein evaluated in biocontrol, formulating suicidal germination stimulants and gene-edited crops. A mycoherbicide was able to become a commercial success because the innovators rendered t

High-throughput sequencing and analysis of target genes uncover microRNA-mediated responses to low-temperature stress in Medicago falcata.

BACKGROUND: Low temperature severely restricts plant growth and development. Medicago falcata is an important germplasm resource for alfalfa breeding because of its strong adaptability and tolerance to abiotic stresses, particularly low-temperature stress. MicroRNAs (miRNAs) are key regulators of stress-responsive genes and modulate plant adaptation to stress by directing transcript cleavage or translational repression. However, the roles of miRNAs in low-temperature response mechanisms in M. falcata remain largely unclear. RESULTS: Here, we constructed miRNA libraries from M. falcata roots g

Integrating Multivariate Analysis and DNA Barcoding for Amaranth Germplasm Characterization and Promising Genotype Selection.

Amaranth (Amaranthus spp.) is a multifaceted genus of C4 plants with significant nutritional and agronomic potential, yet it remains underutilized in mainstream agriculture. Despite growing interest in Amaranth, most germplasm studies have used either phenotypic or molecular approaches alone, lacking integration. Multivariate methods have not been systematically applied to identify promising genotypes, and species-specific selection indices for grain Amaranth remain unexplored. To address these gaps, this study comprehensively characterized 84 Amaranth genotypes representing multiple species (

Transcriptome- and Metabolome-Based Regulation of Growth, Development, and Bioactive Compounds in Salvia miltiorrhiza (Lamiaceae) Seedlings by Different Phosphorus Levels.

Phosphorus (P), as one of the essential bulk elements for plant growth and development, plays an important role in root growth, accumulation of secondary metabolites, and regulation of gene expression. In Salvia miltiorrhiza Bunge (S. miltiorrhiza), an important medicinal plant, the accumulation of its active components is closely related to the level of phosphorus supply, but the molecular regulatory mechanism of phosphorus treatment in the growth and secondary metabolism of S. miltiorrhiza is not clear. In this study, we investigated the effects of low phosphorus (P2), moderate phosphorus (P

Integrated transcriptomic and metabolomic analyses reveal that MsMYC2-A core transcription factor of the jasmine acid signaling pathway positively regulates NaHCO3-base tolerance in alfalfa (Medicago sativa L.).

Alfalfa is recognized as a high-quality forage crop exhibiting exceptional saline-alkaline tolerance. However, in China's Songnen Plain region, soda saline-alkaline soils dominated by NaHCO3 constitute a critical limiting factor for local ecosystems and alfalfa production. Consequently, enhancing alfalfa adaptation to saline-alkaline stress is considered essential for vegetation restoration in degraded lands and sustainable development of animal husbandry. This study employed physiological, transcriptomic, and metabolomic techniques to investigate the complex response mechanisms of alfalfa cul

Identification of drought tolerant Chickpea genotypes through multi trait stability index.

Drought is a major and constantly increasing abiotic stress factor, thus limiting chickpea production. Like other crops, Kabuli Chickpea genotypes are screened for drought stress through Multi-environment trials (METs). Although, METs analysis is generally executed taking into account only one trait, which provides less significant reliability for the recommendation of genotypes as compared to multi trait-based analysis. Multi trait-based analysis could be used to recommend genotypes across diverse environments. Hence, current research was conducted for selection of superior genotypes through

A single nucleotide polymorphism assay sheds light on the extent and distribution of genetic diversity, population structure and functional basis of key traits in cultivated north American cannabis.

BACKGROUND: The taxonomic classification of Cannabis genus has been delineated through three main types: sativa (tall and less branched plant with long and narrow leaves), indica (short and highly branched plant with broader leaves) and ruderalis (heirloom type with short stature, less branching and small thick leaves). While still under discussion, particularly whether the genus is polytypic or monotypic, this broad classification reflects putative geographical origins of each group and putative chemotype and pharmacologic effect. METHODS: Here we describe a thorough investigation of cannabi

A metabolomics characterisation of natural variation in the resistance of cassava to whitefly.

BACKGROUND: Cassava whitefly outbreaks were initially reported in East and Central Africa cassava (Manihot esculenta Crantz) growing regions in the 1990's and have now spread to other geographical locations, becoming a global pest severely affecting farmers and smallholder income. Whiteflies impact plant yield via feeding and vectoring cassava mosaic and brown streak viruses, making roots unsuitable for food or trading. Deployment of virus resistant varieties has had little impact on whitefly populations and therefore development of whitefly resistant varieties is also necessary as part of int

Oil palm natural diversity and the potential for yield improvement.

African oil palm has the highest productivity amongst cultivated oleaginous crops. Species can constitute a single crop capable to fulfill the growing global demand for vegetable oils, which is estimated to reach 240 million tons by 2050. Two types of vegetable oil are extracted from the palm fruit on commercial scale. The crude palm oil and kernel palm oil have different fatty acid profiles, which increases versatility of the crop in industrial applications. Plantations of the current varieties have economic life-span around 25-30 years and produce fruits around the year. Thus, predictable an

The soybean gene GmSP1L-8 improves salt tolerance through the enhancement of the ROS scavenging system.

Microtubule-associated proteins (MAPs) are key regulators of plant stress responses. Although salt stress-induced microtubule dynamics have been widely observed, how specific microtubule-associated factors coordinately regulate plant salt tolerance remains to be further elucidated. In this study, through genome-wide identification and expression profiling of the GmSP1L gene family in soybean (Glycine max), we screened and identified GmSP1L-8, a protein that is significantly induced by salt stress. Subcellular localization analysis revealed that GmSP1L-8 localizes to both the nucleus and cytopl

Shed-Microspore Culture in Ornamental Peppers for Doubled Haploid Plant Production.

The shed-microspore culture technique is an alternative sub-method combining anther and isolated microspore culture to induce microspore embryogenesis. Recently, its effective use in different types of peppers has drawn attention, because it has a higher embryo yield potential compared to anther culture and is more practical than isolated microspore culture. In this chapter, a stepwise protocol for shed-microspore culture of ornamental pepper is described. This protocol includes the steps of donor plant growth conditions, the choice of suitable flower buds based on DAPI staining of microspores
