Pseudomonas syringae activates ZAT18 to inhibit salicylic acid accumulation by repressing EDS1 transcription for bacterial infection.

Phytopathogens can manipulate plant hormone signaling to counteract immune responses; however, the underlying mechanism is mostly unclear. Here, we report that Pseudomonas syringae pv tomato (Pst) DC3000 induces expression of C2H2 zinc finger transcription factor ZAT18 in a jasmonic acid (JA)-signaling-dependent manner. Biochemical assays further confirmed that ZAT18 is a direct target of MYC2, which is a very important regulator in JA signaling. CRISPR/Cas9-generated zat18-cr mutants exhibited enhanced resistance to Pst DC3000, while overexpression of ZAT18 resulted in impaired disease resist

Bacterial Skin Infections in Livestock and Plant-Based Alternatives to Their Antibiotic Treatment.

Due to its large surface area, the skin is susceptible to various injuries, possibly accompanied by the entrance of infective agents into the body. Commensal organisms that constitute the skin microbiota play important roles in the orchestration of cutaneous homeostasis and immune competence. The opportunistic pathogen Staphylococcus aureus is present as part of the normal biota of the skin and mucous membranes in both humans and animals, but can cause disease when it invades the body either due to trauma or because of the impaired immune response of the host. Colonization of livestock skin by

Critical transitions in malaria transmission models are consistently generated by superinfection.

The history of modelling vector-borne infections essentially begins with the papers by Ross on malaria. His models assume that the dynamics of malaria can most simply be characterized by two equations that describe the prevalence of malaria in the human and mosquito hosts. This structure has formed the central core of models for malaria and most other vector-borne diseases for the past century, with additions acknowledging important aetiological details. We partially add to this tradition by describing a malaria model that provides for vital dynamics in the vector and the possibility of super-

Dietary grapes (Vitis vinifera) feeding attenuates ethanol-induced oxidative stress in blood and modulates immune functions in mice.

Ethanol metabolism is known to induce overwhelming production of reactive oxygen species (ROS) and also to cause associated immune dysfunction. Several interventional agents of plant origin, in particular fruits and vegetables have been used to counteract these alterations induced by ethanol. In this study, we investigated the efficacy of dietary feeding of skin and flesh of grapes (Vitis vinifera L.) on the alterations in immune and vascular functions in mice with liver abnormalities induced by chronic ethanol consumption. Results revealed that feeding of both grape skin and flesh (2.5 g/kg b

An oral vaccine based on U-Omp19 induces protection against B. abortus mucosal challenge by inducing an adaptive IL-17 immune response in mice.

As Brucella infections occur mainly through mucosal surfaces, the development of mucosal administered vaccines could be radical for the control of brucellosis. In this work we evaluated the potential of Brucella abortus 19 kDa outer membrane protein (U-Omp19) as an edible subunit vaccine against brucellosis. We investigated the protective immune response elicited against oral B. abortus infection after vaccination of mice with leaves from transgenic plants expressing U-Omp19; or with plant-made or E. coli-made purified U-Omp19. All tested U-Omp19 formulations induced protection against Brucell

Link between Viral Infections, Immune System, Inflammation and Diet.

The strong spread of COVID-19 and the significant number of deaths associated with it could be related to improper lifestyles, which lead to a low-grade inflammation (LGI) that not only increases the risk of chronic diseases, but also the risk of facing complications relating to infections and a greater susceptibility to infections themselves. Recently, scientific research has widely demonstrated that the microbiota plays a fundamental role in modulating metabolic responses in the immune system. There is, in fact, a two-way interaction between lifestyle, infection, and immunity. The immune res

Pathogenesis versus symbiosis: a dual perspective on plant-microbe interactions.

Plants function as integral components of soil ecosystems, embedded within diverse microbial consortia that engage in continuous chemical and informational exchange. This molecular dialog governs how plants distinguish harmful, neutral, and beneficial microbes, transforming the rhizosphere into a dynamic signaling interface. Microbial interactions operate along a chemical continuum in which metabolites, volatiles, peptides, and other small molecules act across aqueous, lipidic, and chemically heterogeneous microenvironments to shape recognition and response. The physicochemical organization of

Identification and Functional Analysis of Candidate Effectors in the Sorghum Aphid, Melanaphis sorghi.

The sorghum aphid (Melanaphis sorghi), a phloem-feeding insect pest, causes severe economic losses in sorghum-producing regions worldwide. During feeding, aphids secrete salivary proteins with potential effector functions that play pivotal roles in plant-insect interactions. However, relatively few secreted proteins and effectors have been characterized in M. sorghi. In this study, proteomic analysis of aphid-infested sorghum leaves identified 69 putative aphid-derived proteins. Bioinformatic analyses were subsequently used to predict signal peptides within these proteins. Among the five candi

Chemical and pharmacological interest of ziyuglycosides I-II: two triterpenoid saponins from Sanguisorba officinalis.

Ziyuglycosides I-II are two glycosylated triterpenoids isolated from the edible medicinal plant Sanguisorba officinalis L. (great burnet). Ziyu-1 is a bidesmosidic saponin whereas its metabolite Ziyu-2 is a monodesmoside. They were initially isolated from the plant roots (Sanguisorbae radix) but they can be found in diverse medicinal plants and polyherbal preparations. This review retraces the origin of the products, discusses synthetic processes to obtain derivatives from the sapogenin precursor pomolic acid, and presents an overview of the pharmacological properties of Ziyu-1/2. The natural

Cucumber Mosaic Virus Coat Protein Sequesters Host CDPK7-Like Into Phase-Separated Condensates to Promote Viral Infection.

Plant viruses often manipulate host proteins to facilitate infection, but the molecular mechanisms underlying these interactions remain largely unclear. Here, we provide evidence that the cucumber mosaic virus (CMV) coat protein (CP) interacts with the host kinase CDPK7-like in Nicotiana benthamiana. In an overexpression system, this interaction facilitates liquid-liquid phase separation (LLPS), forming condensate-like structures that could influence host immune-related signalling. Strikingly, plants overexpressing the CDPK7-like gene exhibited significantly enhanced resistance against diverse

Discovery of highly effective antiviral agents based on flavonoid-benzothiazole against TMV.

A series of flavonol derivatives containing benzothiazole were designed and synthesized. The structures of all the compounds were characterized by NMR and HRMS. The results of the activity assay showed that some of the target compounds possessed outstanding in vivo antiviral activity against the tobacco mosaic virus (TMV). Among them, the median effective concentration (EC50) of L20 was 90.5 and 202.2 μg/mL for curative and protective activity against TMV, respectively, which was better than that of ningnanmycin (NNM: 252.0 and 204.2 μg/mL). The results of microcalorimetric thermophoresis (MST

Overview of Viral Pneumonia Associated With Influenza Virus, Respiratory Syncytial Virus, and Coronavirus, and Therapeutics Based on Natural Products of Medicinal Plants.

Viral pneumonia has been a serious threat to global health, especially now we have dramatic challenges such as the COVID-19 pandemic. Approximately six million cases of community-acquired pneumonia occur every year, and over 20% of which need hospital admission. Influenza virus, respiratory virus, and coronavirus are the noteworthy causative agents to be investigated based on recent clinical research. Currently, anaphylactic reaction and inflammation induced by antiviral immunity can be incriminated as causative factors for clinicopathological symptoms of viral pneumonia. In this article, we i

Phytochemistry, Bioavailability, and Molecular Mechanisms Underlying Multitarget Anticancer Activity of Aloe vera.

Background/Objectives: Cancer, a multifactorial disease with uncontrolled cell growth, oxidative stress, inflammation, genomic instability, and molecular signaling pathways, is a global health concern, leading to the ~20 million newly diagnosed cases annually. Although conventional therapy has been shown to enhance the survival rates of cancer patients, its clinical efficacy is limited by certain side effects that occur as a result of treatment, thus necessitating the exploration of plant-derived bioactive compounds for their potential as safer and alternative supportive therapeutic agents. Multiple scenarios for sexual crosses in the fungal pathogen Zymoseptoria tritici on wheat residues: Potential consequences for virulence gene transmission.

Little is known about the impact of host immunity on sexual reproduction in fungal pathogens. In particular, it is unclear whether crossing requires both sexual partners to infect living plant tissues. We addressed this issue in a three-year experiment investigating different scenarios of Zymoseptoria tritici crosses according to the virulence ('vir') or avirulence ('avr') of the parents against a qualitative resistance gene. Co-inoculations ('vir × vir', 'avr × vir', 'avr × avr') and single inoculations were performed on a wheat cultivar carrying the Stb16q resistance gene (Cellule) and a sus

Identification and Characterization of NPR1 and PR1 Homologs in Cymbidium orchids in Response to Multiple Hormones, Salinity and Viral Stresses.

The plant nonexpressor of pathogenesis-related 1 (NPR1) and pathogenesis-associated 1 (PR1) genes play fundamental roles in plant immunity response, as well as abiotic-stress tolerance. Nevertheless, comprehensive identification and characterization of NPR1 and PR1 homologs has not been conducted to date in Cymbidium orchids, a valuable industrial crop cultivated as ornamental and medicinal plants worldwide. Herein, three NPR1-like (referred to as CsNPR1-1, CsNPR1-2, and CsNPR1-3) and two PR1-like (CsPR1-1 and CsPR1-2) genes were genome-widely identified from Cymbidium orchids. Sequence and ph…
