Tissue engineering of cardiac valve prostheses I: development and histological characterization of an acellular porcine scaffold.

BACKGROUND AND AIMS OF THE STUDY: Several deficiencies in current heart valve prostheses make them problematic for use in younger patients. Tissue valve substitutes are non-viable with a life expectancy of only 10-15 years, while mechanical valves require long-term anti-coagulation therapy. A solution to these problems would be to develop a tissue-engineered heart valve containing autologous cells, enabling the valve to maintain its biochemical and mechanical properties, yet grow with the patient. The study aim was to optimize a protocol to produce a porcine acellular matrix scaffold for use i

Soft Tissue Anchoring Performance, Biomechanical Properties, and Tissue Reaction of a New Hernia Mesh Engineered to Address Hernia Occurrence and Recurrence.

One opportunity to reduce hernia occurrence and recurrence rates (currently estimated to be 30% at 10 years postoperatively) is by enhancing the ability of hernia meshes to anchor into tissue to prevent mesh migration, mesh contraction, and mesh tearing away from tissue. To address this, we developed a novel moderate-weight, macroporous, polypropylene mesh (termed the T-line mesh) with mesh extensions to optimize anchoring. We examined the physical properties, biomechanical performance, and biocompatibility of this novel mesh versus a predicate mesh anchored with #0-suture. The physical proper

Mechanisms underlying biomaterial-regulated macrophage polarization in bone tissue engineering: From interface sensing to immune microenvironment remodeling.

Traditional biomaterials commonly trigger a foreign body reaction after implantation, leading to fibrous encapsulation and poor osseointegration. Advances in osteoimmunology have enabled contemporary tissue engineering to redefine biomaterials as active modulators of the osteoimmune microenvironment rather than as passive scaffolds. At the core of this reconceptualization is the targeted control of macrophage polarization to establish an immune environment conducive to bone repair. The plasticity of macrophages reflects the dynamic balance between the M1 pro-inflammatory and M2 pro-regenerativ…

Development of alendronate-loaded 3D-printed PCL/LDH scaffolds for controlled drug delivery in bone tissue engineering.

Bone tissue engineering has made significant advances in recent years; however, the development of scaffolds that not only support tissue regeneration but also effectively deliver therapeutic agents remains a major challenge. The present study investigates the fabrication and evaluation of 3D-printed polycaprolactone (PCL)/layered double hydroxide (LDH) hybrid scaffolds incorporating alendronate (ALN) for bone tissue engineering applications. First, LDH particles were synthesized via a co-precipitation method and loaded with ALN. Subsequently, scaffolds with varying PCL-to-LDH ratios (2 : 1, 3…

Tocotrienol-Incorporated Gelatin Hydrogel Crosslinked with Genipin for Future Bone Tissue Engineering Applications: Physiochemical Characterization and Biocompatibility.

Oral administration of tocotrienol has poor systemic distribution due to poor selectivity by the α-tocopherol transfer protein at the liver. Local injection of tocotrienols with appropriate drug delivery systems is significant to ensure that the drug is delivered directly to the site of injury or fracture. This paper presents a tocotrienol-loaded gelatin hydrogel crosslinked with genipin for bone regeneration. This innovative method improves the incorporation and sustained delivery of tocotrienol while overcoming its incompatibility with hydrophilic biomaterials. It establishes a novel platfor

A 3D Tissue-Engineering Model of Craniosynostosis to Study the Microenvironmental Signals Leading to Premature Suture Ossification.

Craniosynostosis is a congenital bone developmental condition characterized by the premature ossification of calvarial sutures, leading to restricted skull expansion and potential neurological complications. Although little is known about the signaling that governs this accelerated fusion, our research group has previously identified a stiffness-dependent upregulation of osteogenic genes in cells derived from fused sutures, highlighting the role of mechanotransduction in disease progression. Building on these findings, the present study describes the development of a unique patient-derived thr

In vitro and i n vivo assessment of biomineralized amorphous silica fiber scaffolds for bone tissue engineering.

Scaffolds that simultaneously provide bioactivity, osteoinduction, and structural stability are essential yet remain elusive in bone tissue engineering. Here, we developed degradable amorphous silica fiber scaffolds that actively support biomineral nucleation and growth while maintaining long-term stability. The scaffolds degraded linearly over 40 weeks, and biomineralization enhanced compressive modulus by more than 100%. In vitro, biomineralized scaffolds exhibited excellent biocompatibility, robust mesenchymal stem cell (MSC) proliferation, and accelerated osteogenic differentiation, with s…

Biomaterials-based approaches to mandibular tissue engineering: where we were, where we are, where we are going.

The mandible is the largest craniofacial bone and plays a crucial role in speech, mastication, swallowing, and facial aesthetics. The form or function of the mandible can be altered by defects as a result of tumors, trauma, infection, and congenital conditions. This paper covers the evolution of biomaterials-based approaches to the reconstruction of critical size mandibular defects. Historically the gold standard for critical size mandibular defect repair has been autologous fibula grafts. The emergence of the field of tissue engineering has led to the current research on biomaterial scaffolds…
