Gene therapy for spine fusion.

Spine fusion is a commonly performed yet often unsuccessful surgical procedure. As many as 40% of patients undergoing spine fusion may have a nonunion or failure to form a continuous bone bridge. This clinical challenge has focused much of the attention of osteoinductive bone growth factors toward spine applications. Clinical pilot and pivotal trials will show the feasibility of recombinant and purified bone growth factors to promote spine fusion in humans. Despite this, strategies of gene therapy for spine fusion and other bone healing applications are being pursued. This article reviews the

High-efficiency gene transfer and pharmacologic selection of genetically engineered human keratinocytes.

Low efficiencies of gene transfer to somatic cells have frustrated therapeutic gene delivery efforts in a wide array of tissues including the skin. Production of populations of keratinocytes in which all cells contain the desired therapeutic gene may be important in future genetic therapies. This may be the case in disorders such as epidermolysis bullosa and ichthyosis, where a failure to correct the vast majority of cells within tissue could perpetuate central disease features such as skin fragility and defective barrier function. We have refined retroviral gene transfer parameters to achieve

Transposition of a beta-lactamase locus from RP1 into Pseudomonas putida degradative plasmids.

The beta-lactamase gene from the RP1 plasmid transposes into at least two Pseudomonas putida degradative plasmids. Donor strains that carry RP1 (bla+ tet+ aphA+) and a degradative plasmid yield transconjugants that have only the bla+ marker of RP1. This occurs in up to 80% of all bla+ transconjugants. Segregation of the bla+ marker requires the presence of a degradative plasmid in the donor and is only observed in transconjugants that have received degradative markers. The bla+ tet aphA transconjugants show 100% linkage of bla+ to degradative markers in conjugation, transduction, and transforma

A Comparative Analysis of Transcriptome-Wide Differential Gene Expression and Alternative Polyadenylation in the Ovaries of Meat Ducks and Laying Ducks.

Significant differences in reproductive performance exist between meat-type ducks (e.g., Qiangying Duck, QD) and laying-type ducks (e.g., Shaoxing Duck, SD). The molecular mechanisms underlying these differences, particularly concerning ovarian development and function, remain incompletely understood. This study aimed to comprehensively characterize the ovarian transcriptomes of these two duck types, focusing on differential gene expression and post-transcriptional regulatory events. We performed an integrated full-length transcriptome analysis of ovarian tissues from these two breeds using Pa

Multiomics Approach Identifies Novel Genetic Determinants and Therapeutic Targets for Asthma.

Asthma constitutes a widespread chronic respiratory disease with multifactorial origins, in which definitive genetic components remain incompletely understood. This investigation synthesized transcriptomic data and Mendelian randomization (MR) to delineate causative genes contributing to asthma predisposition. Evaluation of the GSE43696 cohort disclosed 267 genes displaying expression alterations between healthy controls (n = 20) and asthmatic cases (n = 88). MR methods discerned seven causal genes, consisting of six protective determinants (GPRIN3, SHISA2, DUSP5, FCER1A, USP36, and LGALS2; OR

3D-printed solid-contact chloride sensor for sweat-based cystic fibrosis screening.

Cystic fibrosis (CF) is a genetic disorder marked by impaired chloride transport, with elevated sweat chloride concentration serving as the primary diagnostic biomarker. Here, we report a 3D-printed chloride-selective ion-selective electrode (3Dp-Cl--ISE) for sweat chloride analysis relevant to CF screening. This work introduces a solvent-free, 3D-printed solid-contact chloride ISE that integrates a photocurable non-polyvinyl chloride selective membrane with a carbon-cloth transducer. The sensor combines a stereolithography (SLA)-printed membrane with a porous, hydrophobic carbon cloth substra

Exogenous ABA regulates stress tolerance of Leymus chinensis via modulation of PYL‑mediated signaling and antioxidant enzymes.

As an important species for ecological restoration and high-quality forage in northern China, Leymus chinensis has drawn considerable research interest regarding its stress tolerance regulatory mechanisms. Exogenous abscisic acid (ABA), a key stress-signaling hormone, can effectively pre-activate the antioxidant defense system in L. chinensis. To elucidate the ABA response mechanism, this study combined physiological assessments-including antioxidant enzyme activities-with transcriptome sequencing of ABA-treated plants. The results demonstrated that ABA treatment enhanced the activities of per

Multi-Omics Integration Unravels the Genetic and Hormonal Regulatory Mechanisms Underlying Increased Main Stem Node Number in Soybean.

Soybean (Glycine max L.) yield is critically influenced by the number of nodes on the main stem (MSN), which serves as the primary site for pods and seeds. To elucidate the genetic mechanisms underlying MSN, we conducted a multi-omics analysis integrating bulk segregant analysis sequencing (BSA-seq), phytohormone, and transcriptome profilings in a soybean mutant, LSD914, which exhibits a significantly increased MSN number compared to its wild-type parent, HN48. BSA-seq of an F2 population identified 27 candidate genomic regions spanning 2.92 Mb, primarily on chromosome 18. Within these regions

Ifitm3 positively regulates homeostasis of long-term cultured oogonial stem cells derived from Paralichthys olivaceus†.

Fish oogonial stem cells (OSCs) are vital for fish reproduction research and germplasm conservation, yet stable long-term in vitro culture of fish OSCs remains a major technical bottleneck. Herein, we established an efficient in vitro culture system (termed L15SP) for OSCs derived from Paralichthys olivaceus, consisting of L-15 medium supplemented with15% fetal bovine serum (FBS), 2 μg/L basic fibroblast growth factor (bFGF), 2 μg/L leukemia inhibitory factor (LIF), 50 μmol/L β-mercaptoethanol (β-ME), 1% fish serum, and 18 g/L embryo extract protein, cultured at 23 °C. This system enabled stab

Physiological roles of phosphoinositides and inositol phosphates: Implications for metabolic dysfunction-associated steatotic liver disease.

Phosphoinositides and inositol phosphates (IPs) are integral to numerous cellular processes, including membrane trafficking, signal transduction and calcium dynamics. These lipid-derived signalling mediators orchestrate the spatial and temporal regulation of many signalling cascades, largely through interactions with specific effector proteins. Recent studies have highlighted their critical roles in metabolic homeostasis and the pathogenesis of metabolic dysfunction-associated steatotic liver disease (MASLD). In this review, we examine the pathways important for phosphoinositide and IP synthes

Новые знания о генетической основе и транскриптомном освещении вирулентности в вариантах Pseudomonas shiringae pv. actinidiae.

Sex-Specific Transcriptome Signatures in Pacific Oyster Hemolymph.

BACKGROUND/OBJECTIVES: Sex determination and differentiation exhibit remarkable molecular diversity across taxa, driven by genetic, epigenetic, and environmental factors. Invertebrates with sequential hermaphroditism, such as the Pacific oyster (Magallana gigas), represent a poorly understood system despite their role as keystone species and contribution to a substantial aquaculture industry. METHODS: To identify sex-related molecular markers during gametogenesis, we repeatedly sampled hemolymph from artificially conditioned oysters over two months, and sex phenotypes were assigned at the end

Pediatric T-cell lymphoblastic lymphomas but not leukemias harbor TRB::NOTCH1 fusions with unfavorable outcome.

T-cell lymphoblastic lymphoma (T-LBL) and T-cell acute lymphoblastic leukemia (T-ALL) have common and distinguishing clinical and molecular features. Molecular prognostic factors are needed for T-LBL. We assessed the prevalence and prognostic impact of the T-cell receptor β (TRB)::NOTCH1 fusion in 192 pediatric patients with T-LBL and 167 pediatric patients with T-ALL, using novel multiplex polymerase chain reaction and genomic capture high-throughput sequencing techniques. The fusion was detected in 12 patients with T-LBL (6.3%) but in none of the patients with T-ALL (P = .0006, Fisher exact

Lentiviral Vector Production from a Stable Packaging Cell Line Using a Packed Bed Bioreactor.

Self-inactivating lentiviral vectors (LVVs) are used regularly for genetic modification of cells, including T cells and hematopoietic stem cells for cellular gene therapy. As vector demand grows, scalable and controllable methods are needed for production. LVVs are typically produced in HEK293T cells in suspension bioreactors using serum-free media or adherent cultures with serum. The iCELLis® is a packed-bed bioreactor for adherent or entrained cells with surface areas from 0.53 to 500 m2. Media are pumped through the fixed bed and overflows, creating a thin film that is replenished with oxyg

Some mutations in the xeroderma pigmentosum D gene may lead to moderate but significant radiosensitivity associated with a delayed radiation-induced ATM nuclear localization.

Purpose: Xeroderma Pigmentosum (XP) is a rare, recessive genetic disease associated with photosensitivity, skin cancer proneness, neurological abnormalities and impaired nucleotide excision repair of the UV-induced DNA damage. Less frequently, XP can be associated with sensitivity to ionizing radiation (IR). Here, a complete radiobiological characterization was performed on a panel of fibroblasts derived from XP-group D patients (XPD).Materials and methods: Cellular radiosensitivity and the functionality of the recognition and repair of chromosome breaks and DNA double-strand breaks (DSB) was

Regulation of hedgehog Ligand Expression by the N-End Rule Ubiquitin-Protein Ligase Hyperplastic Discs and the Drosophila GSK3β Homologue, Shaggy.

Hedgehog (Hh) morphogen signalling plays an essential role in tissue development and homeostasis. While much is known about the Hh signal transduction pathway, far less is known about the molecules that regulate the expression of the hedgehog (hh) ligand itself. Here we reveal that Shaggy (Sgg), the Drosophila melanogaster orthologue of GSK3β, and the N-end Rule Ubiquitin-protein ligase Hyperplastic Discs (Hyd) act together to co-ordinate Hedgehog signalling through regulating hh ligand expression and Cubitus interruptus (Ci) expression. Increased hh and Ci expression within hyd mutant clones

Tax unleashed: fulminant Tax-positive Adult T-cell Leukemia/Lymphoma after failed allogeneic stem cell transplantation.

The human retrovirus HTLV-1 causes Adult T-cell Leukemia/Lymphoma (ATLL), a malignant lymphoproliferative disease of CD4+ T cells of dismal prognosis, in 3-5% of the 20 million infected individuals (Proietti et al.(1) and Bazarbachi et al.(2)). Infection with HTLV-1 represents a prototypical model of virus-mediated oncogenesis by virtue of the viral transactivator Tax, a potent oncogenic protein that exerts pleiotropic effects through its ability to deregulate the transcription of various cellular genes and signal transduction pathways and inhibit DNA repair enzymes, which are critical for T-c

Assessment of F/HN-pseudotyped lentiviral vector following intravenous delivery to mice.

In pursuit of a gene transfer agent with efficient pulmonary transduction, the UK Respiratory Gene Therapy Consortium has developed a lentiviral vector pseudotyped with the envelope proteins, F and HN from Sendai virus (rSIV.F/HN). In contrast to other viral vectors, pulmonary rSIV.F/HN delivery achieves sustained gene expression ( ~ 2 years in mice) in the lungs and systemic circulation following a single dose. Here, we investigate the application of the rSIV.F/HN vector-platform for wider indications, including systemic disorders that require serum expression of therapeutic proteins. To asse
