Multicenter surveillance of antifungal susceptibility of clinical Aspergillus isolates to conventional and novel antifungal agents in Taiwan, 2021-2023.

UNLABELLED: Periodic surveillance of antifungal susceptibility among Aspergillus isolates is essential to guide effective treatment. This multicenter study analyzed 550 clinical Aspergillus isolates collected in Taiwan from 2021 to 2023, primarily from the respiratory tract (69.6%) and ear (12.9%). Calmodulin-based sequencing identified 24 species across six Aspergillus sections: Flavi (28.7%), Fumigati (28.5%), Nigri (21.6%), Terrei (15.8%), Nidulantes (5.1%), and Circumdati (0.2%). The major sections were represented by Aspergillus fumigatus (99.4%), Aspergillus flavus (91.8%), and Aspergill

Reproductive toxicity induced by a repeated 28-day oral exposure to a commercial mixture of the herbicides S-metolachlor and flumioxazin in male rats.

Male reproductive health has become a growing concern, with evidence indicating a global decline in sperm quality. Among the contributing factors, pesticide exposure stands out. Flumioxazin and S-metolachlor are herbicides widely used in agriculture, and isolated studies report adverse effects on the reproductive physiology of rodents. Toxicological information on their combined exposure remains limited, even though these compounds are frequently applied together in agricultural settings. The objective of this study was to investigate the reproductive toxicity of a commercial formulation conta

Lenacapavir as pre-exposure prophylaxis for HIV prevention.

RATIONALE: Globally, there are 1.3 million new HIV infections annually, with a disproportionate burden on young women and girls, especially in sub-Saharan Africa (63% of new infections). Despite the demonstrated effectiveness of pre-exposure prophylaxis (PrEP), global uptake remains low, reaching only 16.5% of the UNAIDS 2025 target. PrEP adherence is suboptimal in vulnerable populations. There is an urgent need to develop and implement alternative, user-friendly PrEP strategies like long-acting formulations that minimise reliance on daily dosing or frequent injections. Lenacapavir is a first-

Modelling the 5-Year Impact of Twice-Yearly Injectable Lenacapavir for HIV Pre-Exposure Prophylaxis in the United States.

INTRODUCTION: The impact of pre-exposure prophylaxis (PrEP) on the US HIV-1 epidemic has not been fully maximized, potentially due to challenges in uptake, coverage, adherence and consistent use. Following the FDA (Food and Drug Adminstration) approval of twice-yearly lenacapavir, epidemiological projections are needed to understand the potential role of lenacapavir in overcoming PrEP barriers. The objective of this analysis was to estimate the impact of lenacapavir on HIV-1 incidence in the United States. METHODS: We used a static cohort model representing the US population aged 15 or older

Agomelatine normalizes region-specific, diurnal mGluR5 dysregulation in a chronic mild stress rat model of depression.

Desynchronization of circadian rhythms is a hallmark of major depressive disorder (MDD). Agomelatine is an atypical antidepressant that acts as a melatonin receptor agonist and serotonin receptor antagonist. It has shown efficacy in alleviating symptoms of MDD with a favorable side effect profile. In the brain, agomelatine also modulates the glutamatergic system and in the present study, we investigated the effects of chronic mild stress (CMS) and agomelatine treatment on metabotropic glutamate receptor 5 (mGluR5) and synaptic vesicle glycoprotein 2 A (SV2A) binding in the medial prefrontal co

Advances in treatment of chronic graft-versus-host disease.

Chronic graft-versus-host disease (cGVHD) is a leading cause of late morbidity and mortality after allogeneic hematopoietic cell transplantation. Corticosteroids remain the standard first-line therapy; however, many patients develop steroid-refractory or steroid-dependent disease, underscoring the need for more effective and better-tolerated treatments. Ruxolitinib has emerged as the most evidence-supported option for steroid-refractory cGVHD, with the phase 3 REACH3 trial demonstrating higher response rates, durable disease control, and clinically meaningful improvements in symptom burden com

A Comparative Study of the Effects of Nine CXCR3 Antagonists on Macrophage Function and the Treatment of Acute Lung Injury.

BACKGROUND: The C-X-C motif chemokine receptor 3 (CXCR3) antagonist AMG 487 has been shown to alleviate acute lung injury (ALI) in mice. Other CXCR3 antagonists, including NBI-74330, TAK-779, and SCH 546738, exhibit anti-inflammatory effects in various diseases, including apical periodontitis, arthritis, and acute respiratory distress syndrome (ARDS). However, with the exception of AMG 487, the roles of these antagonists in ALI remain poorly understood. Macrophages can differentiate into various phenotypes and play a crucial role in the progression of inflammatory and autoimmune diseases. MET

Effect of some prostanoid receptor modulators on cholinergic and purinergic pathways in rat model of hemorrhagic cystitis: potential beneficial action of selexipag and alprostadil.

Acetylcholine (ACh) and adenosine triphosphate (ATP) are the major neurotransmitters in bladder contraction. This study investigates the modulatory role of some prostanoids in cholinergic and purinergic pathways in normal and cystitis. Cystitis was induced in rats by cyclophosphamide single injection (300 mg/kg) and confirmed histopathologically. Organ bath experiments were implemented using isolated detrusor muscles. Bladder contraction was induced by electrical stimulation and by ACh or ATP direct addition. Cystitis attenuated bladder contractility. Alprostadil and selexipag amplified neurog

Agomelatine and tianeptine antidepressant activity in mice behavioral despair tests is enhanced by DMPX, a selective adenosine A2A receptor antagonist, but not DPCPX, a selective adenosine A1 receptor antagonist.

BACKGROUND: Adenosine, an endogenous nucleoside, modulates the release of monoamines, e.g., noradrenaline, serotonin, and dopamine in the brain. Both nonselective and selective stimulation of adenosine receptors produce symptoms of depression in some animal models. Therefore, the main objective of our study was to assess the influence of a selective adenosine A1 receptor antagonist (DPCPX) and a selective adenosine A2A receptor antagonist (DMPX) on the activity of agomelatine and tianeptine. METHODS: The forced swim test (FST) and tail suspension test (TST) were performed to assess the effect

Effects of selexipag and its active metabolite in contrasting the profibrotic myofibroblast activity in cultured scleroderma skin fibroblasts.

BACKGROUND: Myofibroblasts contribute to fibrosis through the overproduction of extracellular matrix (ECM) proteins, primarily type I collagen (COL-1) and fibronectin (FN), a process which is mediated in systemic sclerosis (SSc) by the activation of fibrogenic intracellular signaling transduction molecules, including extracellular signal-regulated kinases 1 and 2 (Erk1/2) and protein kinase B (Akt). Selexipag is a prostacyclin receptor agonist synthesized for the treatment of pulmonary arterial hypertension. The study investigated the possibility for selexipag and its active metabolite (ACT-33
