The blood pressure in the cardinal vein, cardiac chambers and ventral and dorsal aortae have been recorded from conscious eels in water. At low doses, acetylcholine reduced the heart rate but increased the transbranchial differential blood pressure. The effects were abolished by atropine but not by tubocurarine. At doses in excess of 0.1 mug/kg, a secondary rise in arterial blood pressure was observed which could be abolished by bretylium, phentolamine or phenoxybenzamine. Catecholamines did not affect the heart rate although a transient bradycardia due to reflex inhibition could be observed w…

In receptor-rich membrane fragments from Torpedo, acetylcholine binds, in the presence of 70 muM Tetram, to a homogeneous population of high-affinity sites with Kd = (3.4 +/- 0.8) x 10(08) M. Dissolution of these membrane fragments by sodium cholate causes a decrease of affinity associated with the appearance of medium-affinity (Kd approximately 10(-7) M) and low-affinity (Kd greater than or equal to 10(-6) M) sites. Dissolution by neutral detergents Triton X-100 or Emulphogene preserves the high affinity of the acetylcholine binding sites. In all the soluble states of the receptor protein, Ca…

1. Studies of the synthesis and release of radioactive acetylcholine in rat brain-cortex slices incubated in Locke-bicarbonate-[U-(14)C]glucose media, containing paraoxon as cholinesterase inhibitor, revealed the following phenomena: (a) dependence of K(+)-or protoveratrine-stimulated acetylcholine synthesis and release on the presence of Na(+) and Ca(2+) in the incubation medium, (b) enhanced release of radioactive acetylcholine by substances that promote depolarization at the nerve cell membrane (e.g. high K(+), ouabain, protoveratrine, sodium l-glutamate, high concentration of acetylcholine…

1. Intracellular recording was made with glass micro-electrodes from cells of the longitudinal muscle of the guinea-pig ileum in isotonic and in hypertonic solution.2. In isotonic solution spontaneous bursts of electrical activity occurred; these consisted of a slow potential component which carried a burst of spike action potentials. Acetylcholine increased the size (and the frequency) of the slow potential component. This had the effect of first reducing and then abolishing the spike potentials; continuous slow wave activity was thus produced. Slow waves were about 1 sec in duration and up t…

Non-neuronal acetylcholine (ACh) has been shown to elevate intracellular calcium levels in the Malpighian tubules (MTs) of Mythimna separata. However, the physiological functions and mechanisms of this ACh-mediated signaling in MTs remain largely unknown. In this study, pharmacological analyses suggest that muscarinic receptors trigger ACh-induces Ca2+ signal via the phospholipase C (PLC) and inositol trisphosphate receptor (IP3R) pathway in M. separata MTs. This Ca2+ signal activates a Ca2+-dependent nitric oxide synthase (NOS1), which is specifically localized within the principal cells. NAD…

OBJECTIVE: This study aimed to identify factors associated with secondary generalization in patients with ocular-onset myasthenia gravis (OoMG) who were positive for anti-acetylcholine receptor (AChR) antibodies. Early identification of patients at higher risk of progression to generalized myasthenia gravis (gMG) is clinically relevant to optimize therapeutic strategies and improve long-term outcomes.
METHODS: We conducted a multicenter retrospective cohort study including patients with anti-AChR-positive OoMG recruited from three Italian neurology centers. Demographic, clinical, electrophysi…

BACKGROUND: FcRn antagonists have reshaped generalized myasthenia gravis (MG) treatment by enhancing IgG catabolism, complementing traditional antibody-depleting therapy (ADT). However, disrupting IgG homeostasis may paradoxically cause antibody overshoot, potentially worsening MG symptoms. The risk factors and optimal management of antibody overshoot remain inadequately characterized.
METHODS: We report two novel cases of anti-acetylcholine receptor (AChR) antibody overshoot and clinical exacerbation following FcRn antagonist efgartigimod treatment. To contextualize these findings, we perfor…

Myasthenia gravis (MG) is an autoimmune neuromuscular disorder mediated by autoantibodies targeting the nicotinic acetylcholine receptor (nAChR), which can lead to severe disability and life-threatening crises. Current therapies rely on broad immunosuppression and fail to achieve sustained remission in the majority of patients. Here, we report the development of AChR chimeric autoantibody receptor (CAAR) T cells engineered to selectively eliminate autoreactive B cells producing anti-AChR autoantibodies. T cells were co-transduced with CAARs expressing extracellular domains of the AChRα1 or β1 …

The crosstalk between the M1 muscarinic acetylcholine receptor and the endocannabinoid system was investigated during acetic acid-induced experimental colitis. In this research, male mice were treated with the agonist M1 receptor McN-A-343 (1.5 mg/kg; intraperitoneal) or dexamethasone (2.0 mg/kg; subcutaneous) 17 h or 17 h 30 min after the induction of acetic acid colitis, respectively. The cannabinoid receptor antagonists, CB1 (AM251, 3.0 mg/kg, intraperitoneal) and CB2 (AM630, 1.0 mg/kg, intraperitoneal), were administered 30 min before McN-A-343 and dimethyl sulfoxide (DMSO, 4 %, intraperit…

Cholinergic input initiates or regulates prolonged neuronal activity implicated in executive attention, learning and neuroendocrine control. In the hermaphroditic sea snail, Aplysia, ionotropic acetylcholine receptors on neuroendocrine bag cell neurons trigger an ∼30-min afterdischarge that leads to egg-laying hormone secretion and reproductive behaviour. The afterdischarge is followed by an ∼18-h refractory period, where acetylcholine fails to cause further bursting. Here, we report on a similar long-term depression of acetylcholine receptors in cultured bag cell neurons. Under whole-cell vol…

Myasthenia gravis (MG) is an autoimmune disorder caused mainly by autoantibodies against the acetylcholine receptor (AChR), leading to muscle weakness. While treatments targeting AChR autoantibodies benefit many, some patients remain refractory, highlighting the need for personalized therapies. This study evaluates the therapeutic potential of S-1117, a pan-IgG-specific protease, in AChR autoantibody-mediated pathology. Using live cell-based assays, we examined AChR-specific monoclonal IgG autoantibodies (mAbs) and patient-derived serum samples for their effects on receptor binding, blockade, …

Patients with pemphigus vulgaris (PV) develop IgG autoantibodies (AuAbs) binding to keratinocyte desmogleins (Dsg), acetylcholine (ACh) receptors, mitochondrial proteins, and some other self-antigens. In this study, we identified linear and discontinuous peptide tetrameric epitope segments (ES) of M3 muscarinic ACh receptor (M3AR) targeted by different anti-M3AR AuAbs. As positive controls, we identified Dsg1 and Dsg3 ES targeted by PV sera. Healthy individuals also possessed natural antibodies targeting M3AR, Dsg1 and Dsg3 epitopes that were different from those targeted by AuAbs produced by …

Vagus nerve regulates viral infection and inflammation via the alpha 7 nicotinic acetylcholine receptor (α7 nAChR); however, the role of α7 nAChR in ZIKA virus (ZIKV) infection, which can cause severe neurological diseases such as microcephaly and Guillain-Barré syndrome, remains unknown. Here, we first examined the role of α7 nAChR in ZIKV infection in vitro. A broad effect of α7 nAChR activation was identified in limiting ZIKV infection in multiple cell lines. Combined with transcriptomics analysis, we further demonstrated that α7 nAChR activation promoted autophagy and ferroptosis pathways …
