Effects of T cell adoptive transfer into nude mice on alveolar bone resorption induced by endotoxin.

Using the method of reconstitution of nude mice with T cells, we examined the effects of T cell on alveolar bone resorption induced by repeated injections of Escherichia coli endotoxin into periodontal tissue. Three mice groups (normal, nude and T cell reconstituted nude mice) were used. Endotoxin derived from E coli was repeatedly injected into the gingiva of the mice left mandibles every 48 h and the mice were killed on the day after the 1st, 4th, 7th, 10th, 13th and 20th injections of endotoxin. Alveolar bone resorption was examined histopathologically and histomorphometrically. Bone surfac

Adoptive transfer of lymphocytes sensitized to the major surface glycoprotein of Pneumocystis carinii confers protection in the rat.

Pneumocystis carinii is a major opportunistic pathogen and a leading cause of morbidity in patients with AIDS. CD4+ cells have been shown to be important in host defenses against P. carinii, but the antigen(s) involved with this response have not been identified. We undertook the present study to determine whether the major surface glycoprotein (MSG) of P. carinii contains epitopes that can elicit a protective cellular immune response. Spleen cells and purified CD4+ cells isolated from Lewis rats, pulsed 1-4 d with MSG, and injected into corticosteroid-treated Lewis rats with pneumocystosis re

[Successful local adoptive immunotherapy for pleuritis carcinomatosa due to renal cell cancer. A case report].

A 54-year-old woman who had undergone radical nephrectomy for renal cell cancer nine months before was admitted to our hospital because of difficulty in breathing. X-ray films of the chest showed massive pleural effusion on the left side and cytological examination of the effusion revealed malignant cells which may have originated from renal cell cancer. Intrapleural instillations of interleukin-2 and of tumor infiltrating lymphocytes isolated from the pleural effusion were performed. After the initiation of the treatment, the pleural effusion decreased and malignant cells disappeared from the

Adoptive transfer of experimental autoimmune hepatitis in mice--cellular interaction between donor and recipient mice.

This report extends our previous study on experimental autoimmune hepatitis in C57BL/6(B6) mice. Cellular immunity involved in the induction of liver injury in this model was studied by transfer of primed spleen cells from hepatitis donor mice to syngeneic normal recipient mice. The most prominent liver damage in recipient B6 mice was induced by transfer of nylon wool adherent spleen cells from hepatitis donor mice, and T cells in this fraction were the essential requirement for the liver damage in the recipient mice. Nylon wool adherent spleen cells from hepatitis donor mice after depletion o

Antibody levels in adoptively immunized mice after infection with Babesia microti or injection with antigen fractions.

Specific IgG anti-Babesia antibodies could be detected by an ELISA test in the serum of adoptively immunized mice which subsequently were infected with Babesia microti. Initially, higher IgG antibody levels were present in the recipients of either untreated immune spleen cells or mitomycin C-treated immune spleen cells compared with the values in control mice which received the infection alone. Subsequently, a marked anamnestic response occurred only in mice which received untreated immune spleen cells and not in the recipients of mitomycin C-treated cells, demonstrating a requirement for prol

Adoptive transfer of anti-syphilis immunity with lymphocytes from Treponema pallidum-infected guinea pigs.

Spleen and lymph node cells taken from strain 2 and strain 13 guinea pigs at the peak of their primary immune response to cutaneous syphilitic infection could transfer partial protection to symptomatic disease to normal syngeneic recipients challenged with the Nichols strain of Treponema pallidum. These recipients of immune cells had significantly fewer treponemes disseminating to the regional lymph nodes and developed fewer and less severe cutaneous lesions that resolved faster than those in guinea pigs that had been infused with normal lymphoid cells. Immune donor cells also had the capacity

Phase I study of the adoptive immunotherapy of human cancer with lectin activated autologous mononuclear cells.

In previous in vitro studies, the authors showed that phytohemagglutinin (PHA) stimulated peripheral blood lymphocytes (PBL) from cancer patients to generate cells that were lytic for fresh autologous tumor but not for lymphocytes or lymphoblasts. Thus, after IRB approval, a phase I clinical protocol was instituted in cancer patients who had failed all other therapy to determine the toxicity and effects, in vivo, of the infusion of large numbers of such PHA activated autologous PBL. Ten patients were treated on the protocol, six with sarcoma, one with melanoma, and three with colorectal cancer

Restriction in adoptive transfer of resistance to Listeria monocytogenes. I. Influence of non-H-2 loci.

In vivo adoptive transfer of T-cell-mediated immunity to the facultative intracellular bacterium Listeria monocytogenes is restricted, not only by the H-2 haplotype of the mice, but also by incompatibilities at non-H-2 loci. Thus, transfer between H-2 identical strains of mice with different background genes was reproducibly and significantly less efficient than transfer between completely syngeneic mice, although the restriction was less marked than that across the H-2 barrier. Restriction also occurred when parental cells were injected into semisyngeneic F1 hybrids and when cells from F1 hyb

Mechanisms of immunologic eradication of a syngeneic guinea pig tumor. I. Quantitative analysis of adoptive immunity.

Adoptive immunity against a syngeneic hepatoma (line-10) of Sewall-Wright inbred strain 2 guinea pigs was analyzed by a two-dimensional titration of iv transferred immune lymphoid cells versus intradermal tumor challenges. Tumor resistance increased exponentially as a function of the number of immune lymphoid cells transferred. Within the tumor challenge doses analyzed, suppression of tumor growth mediated by the transferred immune lymphoid cells appeared to be independent of the primary immune response in the recipient. Quantitatively, rejection of a given number of tumor cells reflected the

Adoptive transfer of resistance to Plasmodium berghei with spleen cells and serum from Fansidar-cured mice.

The ability of splenic leukocytes or serum to transfer immunity to Plasmodium berghei was studied in C57BL/6 mice. Splenic leukocytes or serum was removed from mice which had been inoculated previously 1 to 4 or 5 times with P. berghei and cured 1 to 4 or 5 times with Fansidar (pyrimethamine plus sulfadoxine) and transferred to syngeneic recipients 1 or 2 days before parasite challenge. Partial protection was observed in recipients of immune serum or unfractionated splenic leukocytes. Significantly more protection was observed in recipients of preparations from immune mice enriched for immunog

Suppressor cells in transplantation tolerance. I. Analysis of the suppressor status of neonatally and adoptively tolerized rats.

The lymphocytes from neonatally tolerant rats which adoptively transfer tolerance to sublethally irradiated recipients do so by specifically suppressing the regeneration of alloreactivity which normally occurs after irradiation. Although tolerant cells will only partially suppress normal alloreactive cells when the two are mixed in near equivalent numbers, experiments in which the interval between injection of tolerant and normal cells into irradiated recipients was gradually extended, indicated that total suppression of normally alloreactive cells was achieved after 8 weeks of prior residence

Adoptive transfer of immunity from mice immunized with ribosomes or live yeast cells of Histoplasma capsulatum.

This investigation was designed to compare the role of lymphoid cells and immune serum in protective immunity induced by immunization with ribosomes or live yeast cells of Histoplasma capsulatum. Spleen cells, peritoneal cells, and serum from C3H mice immunized with Histoplasma ribosomes or live cells were transferred intravenously to separate groups of syngeneic recipients. All recipients along with a set of immunized and control mice were challenged intravenously with 4 x 10(6) yeast cells of H. capsulatum, and protection was assessed. Immunization with ribosomes or live cells provided 90 to

Innovations and emerging trends in adoptive cell therapies for solid tumors: engineering strategies and clinical translation.

Cancer remains a formidable challenge in global oncology, with conventional treatments often failing to achieve durable responses in advanced stages. Adoptive cell therapy (ACT), despite its revolutionary success in hematological malignancies, faces critical bottlenecks in solid tumors, including a hostile immunosuppressive tumor microenvironment (TME), restricted cellular trafficking, and the rapid onset of T-cell exhaustion. This review systematically explores the evolution of ACT platforms, ranging from foundational tumor-infiltrating lymphocytes (TILs) to precision-engineered chimeric anti

Practical Considerations for Adopting AI Clinical Decision Support Tools and Large Language Models in Dermatology.

Artificial Intelligence (AI) in dermatology is a rapidly evolving field, offering promising advancements in the diagnosis and management of skin cancers. As global skin cancer incidence rises, tools like machine-learning based systems and large language models (LLMs) have demonstrated significant potential in supporting triage of skin cancer patients. Yet several key practical and ethical considerations stand in the way of widespread adoption. Limitations in dataset diversity, medicolegal ambiguity, and concerns regarding transparency of AI-generated outputs are challenges that must be address…

Enforced BATF expression via clinically approved LNPs enhances adoptive T-cell therapies.

Chimeric antigen receptor (CAR) T-cell therapy is highly effective in hematologic malignancies, yet its durability is limited by insufficient expansion, persistence, and T-cell exhaustion. Basic leucine zipper ATF-like transcription factor (BATF) promotes CD8+ T-cell effector differentiation but can drive exhaustion under chronic stimulation. Here, we developed a transient, non-viral strategy to modulate BATF expression in therapeutic T cells using clinically approved lipid nanoparticles (LNPs). Among 3 Food and Drug Administration (FDA)-approved ionizable lipids, SM-102-based LNPs achieved th

Modelling the impact of adopting new-generation insecticide-treated nets on malaria transmission and insecticide resistance.

BACKGROUND: Widespread insecticide resistance increasingly threatens malaria elimination, prompting a reassessment of vector control strategies. As Tanzania transitions from standard pyrethroid-only insecticide-treated nets (ITNs) to new-generation nets, evaluating the impact of this shift on malaria transmission and resistance is critical. METHODS: Using the agent-based malaria model, EMOD, we assessed the impact of three ITN types, standard pyrethroid-only nets, pyrethroid-PBO nets (Olyset® Plus/PermaNet® 3.0), and the dual-active Interceptor® G2 nets (IG2) on malaria transmission and on mo

Adoptive T Cell therapy for cancer and autoimmune disease: current advances, challenges, and future directions.

Adoptive T Cell Therapy (ACT) is a kind of immunotherapy that eliminates target cells by autologous T cells. Since the mechanism of adoptive immune was elucidated, ACT has been developed for more than half a century. As one of ACTs, chimeric antigen receptor (CAR)-T cell therapy produced remarkable clinical responses in some hematological malignancies that is considered revolutionary strategy of immunotherapy. However, the effects of CAR-T cell therapy in solid tumors are hindered by immunosuppressive environment and limit of CAR to only recognize antigens on the cell surface. T cell receptor-
