Chili peppers and chili products are ingredients used throughout the world. However, safety and quality concerns persist. This study established an ultra-high-performance liquid chromatography coupled with triple quadrupole mass spectrometry method for the simultaneous analysis of pesticide residues, mycotoxins, and major capsaicinoids (capsaicin, dihydrocapsaicin, nordihydrocapsaicin) in fresh chili peppers and chili powders. Chemical analysis of capsaicinoids using the Scoville Heat Unit was used to determine the pungency levels of the chilies. With this method, Czech retail chili samples we

Given the extreme toxicity and carcinogenicity of aflatoxin B1 (AFB1) in foodstuffs and medicinal products, ultrasensitive detection for it is critically important. Herein, we engineered a type-II dual WO3/g-C3N4/Cu2O-Au heterojunction via hydrothermal synthesis and photoreduction, which synergistically integrates the structural stability of WO3, the visible-light response of Cu2O, the optimal band alignment of g-C3N4, and the plasmonic enhancement effect of Au nanoparticles. Leveraging the band structure differences among these constituent semiconductor components, this architecture enables e

Lycopene is a type of carotenoid pigment widely distributed in various plants and fruits, with tomatoes, carrots, and guava being the most abundant sources. Due to its high content of beneficial active components, lycopene has been used in medicine, where it is employed as a dietary additive for cancer therapy, immune modulator, and feed additive to improve livestock productivity. Lycopene is a lipophilic substance that can act as either a prooxidant or a free radical scavenger and is particularly efficient in enhancing broiler performance. Furthermore, lycopene can alleviate heat stress by im

It is well documented that liver is the primary target organ of aflatoxin B1 (AFB1) and curcumin proved to be effective against AFB1-induced liver injury. In the present study, we investigated the preventive effects of curcumin against AFB1-induced apoptosis through the molecular regulation of p53, caspase-3, Bax, caspase-9, Bcl-2 and cytochrome-C associated with mitochondrial pathway. Liver antioxidant levels were measured. The hallmarks of apoptosis were analysed by methyl green-pyronin-Y staining, transmission electron microscopy, RT-PCR and western blot. Results revealed that dietary curcu

Liver cancer is one of the world's most common cancers and the second leading cause of cancer deaths. Hepatocellular carcinoma (HCC), a primary hepatic cancer, accounts for 90%-95% of liver cancer cases. The pathogenesis of HCC consists of a stepwise process of liver damage that extends over decades, due to hepatitis, fatty liver, fibrosis, and cirrhosis before developing fully into HCC. Multiple risk factors are highly correlated with HCC, including infection with the hepatitis B or C viruses, alcohol abuse, aflatoxin exposure, and metabolic diseases. Over the last decade, genetic alterations

The distinct physiology of the liver makes it a unique ground with respect to its cross talk with p53, the "guardian of the genome." The stressful environment in the liver frequently leads to the activation of p53, which is associated with alterations in metabolic pathways and induction of apoptosis. The latter serves as a mechanism that controls the deposal of DNA-damaged cells. However, accentuated apoptosis may eventually lead to liver pathologies, mainly steatosis, which can develop into a more severe disease such as steatohepatitis, fibrosis, and cirrhosis. These pathologies, together wit

Genomic analysis of hepatocellular carcinoma (HCC) has been shown to provide clues about local risk factors. In the last decades, the mortality from malignant liver tumors increased sharply in Romania, where both hepatitis viruses and environmental pollutants are known to be highly prevalent. To date, HCC from this country has not been subject to molecular characterization. We analyzed a series of 48 consecutive HCC cases. Point mutations were searched in 9 nuclear genes and the mitochondrial D-loop. Oxidative stress response was monitored through measurement of gene expression (NRF2, KEAP1, S

Autophagy is an evolutionarily conserved pathway for degradation of cytoplasmic proteins and organelles via lysosome. Proteins coded by the autophagy-related genes (Atgs) are the core molecular machinery in control of autophagy. Among the various biological functions of autophagy identified so far, the link between autophagy and cancer is probably among the most extensively studied and is often viewed as controversial. Autophagy might exert a dual role in cancer development: autophagy can serve as an anti-tumor mechanism, as defective autophagy (e.g., heterozygous knockdown Beclin 1 and Atg7 i

Aflatoxin B(1) (AFB(1)) is a risk factor for hepatocellular carcinoma (HCC) in many low-resource countries. Although its metabolites bind at several positions in TP53, a mutation at codon 249 (AGG to AGT, arginine to serine, p.R249S) accounts for 90% of TP53 mutations in AFB(1)-related HCC. This specificity suggests that p.R249S confers a selective advantage during hepatocarcinogenesis. Using HCC cell lines, we show that p.R249S has lost the capacity to bind to p53 response elements and to transactivate p53 target genes. In p53-null Hep3B cells, stable transfection of p.R249S or of another mut

It has been shown that hepatocytes cultured in a collagen sandwich configuration maintain cell viability, morphology, and drug metabolizing activities for several weeks. The purpose of this study was to characterize chemically induced general toxicity in this system by exposing hepatocytes to eight different hepatotoxic compounds. Cell function and viability was measured by analyzing the secretions of urea and albumin and the release of lactate dehydrogenase. Significant decreases in urea and albumin secretions were detected after treatments with 32 nM aflatoxin B(1) and 1 mM doses of cadmium

The synthetic monocyclic phenolics (MPs), acetaminophen (APAP), butylated hydroxyanisole (BHA), and butylated hydroxytoluene (BHT) are antimutagenic or anticarcinogenic against a diversity of chemical carcinogens affecting a variety of tissues in experimental animals. In studies in this laboratory of the anticarcinogenicity of MPs, the focus has been on delineating efficacy at low levels of MPs that do not elicit adaptive or toxic responses. To accomplish this, we are studying anticarcinogenicity against the neoplastic initiating activity of lower doses of carcinogens than have previously been

Mutational inactivation of the tumor suppressor gene p53 is common in hepatocellular carcinomas (HCC). AGG to AGT transversion in codon 249 of exon 7 of the p53 gene occurs in over 50% of HCC from endemic regions, where both chronic infection with the hepatitis B virus (HBV) and exposure to carcinogens such as aflatoxin B1 (AFB1) prevail. In this study, we report the effect of the HBV x protein (HBx) on carcinogen-induced cytotoxicity and AGG to AGT mutation in codon 249 of the p53 gene in the human liver cell line CCL13. Expression of HBx, as revealed by its transactivation function, results

Hepatobiliary neoplasms comprise a significant portion of the worldwide cancer burden. Advances in basic science research have led to rapid progress in our understanding of the molecular events responsible for these dreaded diseases. The genetic changes associated with hepatocellular carcinoma (HCC) have received the most attention. Aflatoxin B1 exposure leads to mutations in the p53 tumor suppressor gene, most commonly a transversion in codon 249 that leads to a substitution of serine for arginine in the p53 protein. Numerous other tumor suppressor genes, oncogenes, and tumor gene pathways ar

There are two ways of connecting Epstein-Barr virus (EBV) with the uncontrolled growth of EBV infected B lymphocytes: in case of evident immunosuppression when the control by cellular immunity is missing or in the case of pathological growth of malignant clone as a result of genetic translocations. Today, EBV is linked with the development of lymphomas in immunosuppressed patients, Hodgkin's and Burkitt's lymphoma and nasopharyngeal carcinoma. The presence of EBV genome in these patients can be confirmed in malignant cells, in lower or higher percent, as well as the high titers of antibodies a

Dietary aflatoxin and hepatitis B virus infection may play a role in generating the p53 tumor suppressor gene codon 249 hotspot mutation found in human hepatocellular carcinomas (HCCs) from Qidong (China) and southern Africa. No data are available on the HCC site-specific mutation of the p53 gene in hepadnavirus-infected animals exposed to AFB1. We have searched for the presence of p53 gene codon 249 mutations in both duck hepatitis B virus (DHBV) positive and negative HCCs of domestic ducks from Qidong, where the human p53 hotspot is so prevalent, as well as in duck HCCs experimentally induce

Mutations in the p53 tumour suppressor gene have been recently described in hepatocellular carcinomas (HCC) from high risk areas such as China and South Africa. Our study was designed to assess the importance of p53 aberrations in HCCs from Europe, where the major risk factors in hepatocarcinogenesis, aflatoxin exposure and chronic hepatitis B virus (HBV) infection, do not play a dominant role. We investigated 22 HCCs and, as controls, their corresponding tumour-free liver tissues, seven livers with primary biliary cirrhosis and four morphologically normal livers. p53 overexpression, which is

To explore further the mechanism underlying the alteration in expression of P450 enzymes in hepatic preneoplastic lesions, expression of CYP2B1/2 and 3A1 in individual hepatic hyperplastic nodules induced by an aflatoxin B1 (AFB) administration protocol and the Solt-Farber resistance protocol in male F344 rats was examined via immunohistology. In nodules induced by the resistance protocol, expression of both CYP2B1/2 and 3A1 proteins was highly variable among different nodules, whereas these P450 proteins were expressed in all nodules induced by the AFB protocol. Nodules induced by the resista

INTRODUCTION: Gut microbiota are vulnerable to foreign chemicals (xenobiotics) including pharmaceuticals, environmental pollutants, and dietary contaminants such as aflatoxin B1 (AFB1) and fumonisin B1 (FB1). Assessing the effect of these xenobiotics in the laboratory requires their dissolution in a solvent vehicle, such as dimethyl sulfoxide (DMSO). While DMSO is typically used at low concentrations under the assumption of neutrality, its independent impact on microbial dynamics is a potential experimental confounder that has not been fully explored.
METHODS: Human fecal microbiota were cult

Mycotoxin exposure is a major public health concern in Africa due to food insecurity that compromises food safety. Infants and young children are highly vulnerable due to immature immunity and higher food intake per body weight. However, data on mycotoxin exposure in Africa remain limited. This study investigated mycotoxin exposure in Nigerian infant-mother pairs and toddlers using biological samples (breast milk, infant/toddler urine) collected from three agro-ecological zones. Overall, 75.6% of all the samples tested positive for at least one mycotoxin. Zearalenone, aflatoxin M1, ochratoxin
