Lactose-functionalized photosensitizer nanoparticles derived from thiophene-fused BODIPY for NIR-imaging-guided photodynamic treatment of hepatocellular carcinoma.

Four near-infrared (NIR) photosensitizers (PSs) (PHTB, PBrTB, THTB, and TBrTB) based on thiophene-fused, BODIPY-conjugated, and liver-tumor-targeting lactose were synthesized and characterized. Brominating the BODIPY core of PHTB and the terminal thiophene rings of THTB yielded PBrTB and TBrTB, respectively. All derivatives exhibited strong NIR absorption (λab = 640-674 nm) as well as moderate fluorescence quantum yields (ΦF = 0.21-0.35) and self-assembled into stable, uniform nanoparticles (∼30 nm) in aqueous solution, driven by the hydrophobic interactions of the BODIPY core and hydrophilic

Hexavalent/Tetravalent Tellurium-Based Chalcogen Bond Catalysis on the Cyclization of 1,6-Diyne: Mechanistic Insights and Computational Design.

Recently, the noncovalent organocatalysis on the alkyne has garnered considerable interest. Hexavalent tellurium-based chalcogen bond catalysis is a novel and efficient catalytic strategy in noncovalent organocatalysis, and tetravalent tellurium-based chalcogen bond catalysis has been effectively used in the metal-chloride bond activation, bromination, aza-Diels-Alder, and Povarov reactions. In this work, we perform an investigation on the intramolecular cyclization of 1,6-diyne catalyzed by hexavalent and tetravalent tellurium-based catalysts. The hexavalent and tetravalent tellurium···π chal

The association between blood trihalomethane concentrations and high risk for prostate cancer: A nationally representative cross-sectional study.

Trihalomethanes (THMs), a class of disinfection byproducts, have been implicated in various health risks. However, their association with prostate cancer (PCa) has not been systematically evaluated in biomarker-based studies. This cross-sectional study investigated associations between blood THMs exposure and both total prostate-specific antigen (tPSA) levels and high risk for PCa status using data from 2319 male participants (≥40 years) in the 2003-2010 National Health and Nutrition Examination Survey (NHANES) cycles. Blood concentrations of bromoform (TBM), chloroform (TCM), bromodichloromet

The compound risk of decabromodiphenyl ethane and cadmium to soil nutrient cycling in agroecosystems: Linking the enzyme, microbial community, gene networks to plant stress.

Soil serves as the habitat for numerous organisms and is increasingly threatened from co-pollution of novel brominated flame retardant (NBFRs) and heavy metals (HMs). Focusing on Deca-bromodiphenyl ethane (DBDPE) and cadmium (Cd) as the targets, we constructed a soil-lettuce-earthworm microcosm to explore co-pollution effects in rhizosphere soils. Results showed that DBDPE increased bioavailable Cd fraction to amplified its ecological risks. Co-pollution significantly inhibited the expression of soil functional genes, especially C/N cycling genes, including pgu and abfA in C-cycling and narG,

Environmental levels of bis(2-ethylhexyl)-tetrabromophthalate (TBPH) cause thyroid toxicity and early carcinogenic signals in juvenile fish.

Exposure to novel brominated flame retardants (NBFRs) may engender thyroid dysfunction and even increase thyroid cancer risk. However, information regarding their thyroid toxic or disorder potential is primarily limited to thyroid hormone homeostasis. Here, we conducted a 28-day exposure experiment with juvenile rare minnows (Gobiocypris rarus) to 0.1-1000 μg/L bis(2-ethylhexyl)-2,3,4,5-tetrabromophthalate (TBPH, a NBFR), which preferentially accumulates in freshwater fish. First, molecular docking revealed the binding potential of TBPH to the thyroid receptor of the rare minnow. Subsequently,

Thyroid Function Effects of Mixed Exposure to Urinary Trihalomethanes and Haloacetic Acids: Based on an Integrated Framework of Exposure Assessment, Qualitative Association, and Quantitative Attribution.

Toxicological studies have demonstrated that disinfection byproducts (DBPs) can disrupt thyroid function; however, human epidemiological evidence remains limited. The existing studies focus on a limited number of compounds and lack detailed investigation of mixed exposure effects. To address these gaps, we developed a three-tier analytical approach that includes exposure assessment, qualitative association, and quantitative attribution. A total of 435 community-dwelling adults in Shanghai, China, were enrolled, and urinary concentrations of 16 DBPs, including halomethanes (HMs) and haloacetic

Network toxicological and single-cell sequencing reveals the potential mechanisms of psoriatic toxicity of polybrominated diphenyl ethers.

Polybrominated diphenyl ethers (PBDEs), major brominated flame retardants, have been implicated in various health issues due to their environmental persistence and bioaccumulation. PBDEs preferentially accumulate in the skin due to their low hydrophobicity and may contribute to the onset of psoriasis. In this study, we aimed to investigate the potential mechanisms of PBDE-induced psoriatic toxicity by utilizing network toxicology at single-cell resolution. Initially, 139 overlapping targets between PBDEs and psoriasis were identified, and their protein-protein interactions (PPIs) were mapped.

Proinflammatory microglial response is a common mechanism of Aroclor 1254- and Tetrabromobisphenol-A-induced neurotoxicity in immature chronically exposed rats.

Polychlorinated biphenyls (PCBs) and brominated flame retardants (BFRs) are dominant environmental and food contaminants. Tetrabromobisphenol A (TBBPA) is the most widely used BFR in the world to improve the fire safety of laminates in electrical and electronic equipment. Aroclor 1254, one of the PCBs, is widely distributed in the environment due to its extensive use in industrial applications around the world. Both groups of substances are potent toxicants. There is also increasing evidence that they have neurotoxic effects. In this study we tested the pro-inflammatory effects of Aroclor 1254

Network analysis to reveal the most commonly detected compounds in predator-prey pairs in freshwater and marine mammals and fish in Europe.

Marine and freshwater mammalian predators and fish samples, retrieved from environmental specimen banks (ESBs), natural history museum (NHMs) and other scientific collections, were analysed by LIFE APEX partners for a wide range of legacy and emerging contaminants (2545 in total). Network analysis was used to visualize the chemical occurrence data and reveal the predominant chemical mixtures for the freshwater and marine environments. For this purpose, a web tool was created to explore these chemical mixtures in predator-prey pairs. Predominant chemicals, defined as the most prevalent substanc

Sex-specific effects on elements of the social brain neural network in Wistar rats from perinatal exposure to FireMaster 550 or its components.

Developmental exposure to chemical flame retardants (FRs) has been linked to a variety of neurodevelopmental disorders and abnormal socioemotional behaviors in human and laboratory animal studies. We have previously shown in Wistar rats that gestational and lactational exposure to the FR mixture Firemaster 550 (FM 550) or its brominated or organophosphate ester (OPFR) components (at 2000 µg, 1000 µg, and 1000 µg oral to the dam respectively (absolute and not by bodyweight)) results in increased anxiety-like behaviors in females and decreased sociality in both sexes. Reprotoxic Effect of Tris(2,3-Dibromopropyl) Isocyanurate (TBC) on Spermatogenic Cells In Vitro.

Tris(2,3-dibromopropyl) isocyanurate (TBC) belongs to the class of novel brominated flame retardants (NFBRs) that are widely used in industry. It has commonly been found in the environment, and its presence has been discovered in living organisms as well. TBC is also described as an endocrine disruptor that is able to affect male reproductive processes through the estrogen receptors (ERs) engaged in the male reproductive processes. With the worsening problem of male infertility in humans, a mechanism is being sought to explain such reproductive difficulties. However, so far, little is known ab

Association of blood trihalomethane concentrations with hypertension in US adults: A nationwide cross-sectional study.

Trihalomethanes (THMs), as the most common species of disinfection byproducts in chlorinated water, have been associated with hypertensive disorders in pregnancy. However, there is sparse epidemiological evidence regarding the possible link between THMs exposure and hypertension in general adults. In the present study, we aimed to characterize the associations between THMs exposure and hypertension in general adults. We performed cross-sectional analyses of 15,135 adults from the 1999-2018 National Health and Nutrition Examination Survey. In the general US adults, the median blood concentratio

Intracellular Bacterial Targeting by a Thiazolyl Benzenesulfonamide and Octaarginine Peptide Complex.

A brominated thiazolyl benzenesulfonamide (BTB) derivative is conjugated with the cell-penetrating peptide octaarginine (R8) in an effort to construct innovative antibacterial products. The noncovalent complex of BTB and R8 is characterized by Fourier transform infrared (FTIR) spectroscopy, which indicates hydrogen bonding between the two constituents. Attachment of the peptide moiety renders aqueous solubility to the hydrophobic benzenesulfonamide drug and bestows bactericidal activity. Confocal imaging in conjunction with dye probes shows successful clearance of intracellular Staphylococcus

Synthesis and anti-inflammatory activity of paeonol derivatives with etherized aryl urea by regulating TLR4/MyD88 signaling pathway in RAW264.7 cell.

Thirty-three novel paeonol etherized aryl urea derivatives (PEUs) were synthesized via a bromination-Williamson Ether Synthesis-deprotection-nucleophilic addition reaction sequence. The structures of PEUs were characterized by LC-MS, HRMS, 1H NMR and 13C NMR spectra. The levels of nitric oxide (NO), tumor necrosis factor-alpha (TNF-α) and interleukin-1 beta (IL-1β) in lipopolysaccharide (LPS)-induced RAW264.7 macrophages were initially employed to evaluate the anti-inflammatory effects of all compounds. Remarkably, b16 exhibited a good anti-inflammatory activity at 2.5 μm which is the same as

Synthesis of N-phenyl- and N-thiazolyl-1H-indazoles by copper-catalyzed intramolecular N-arylation of ortho-chlorinated arylhydrazones.

The broad application of 1H-indazoles has prompted the development of several approaches for the synthesis of such compounds, including metal-free, palladium-, or copper-promoted intramolecular N-arylation of in situ-generated or isolated o-haloarylhydrazones. Such methods mainly start from o-bromo derivatives due to the better yield observed when compared to those obtained from o-chloroarylhydrazones. However, the o-chloroarylaldehydes and o-chloroarylketones used to prepare the arylhydrazones are more commercially available and less expensive than brominated analogs. Seeking to cover a lack

The Binding of Monoclonal and Polyclonal Anti-Z-DNA Antibodies to DNA of Various Species Origin.

DNA is a polymeric macromolecule that can display a variety of backbone conformations. While the classical B-DNA is a right-handed double helix, Z-DNA is a left-handed helix with a zig-zag orientation. The Z conformation depends upon the base sequence, base modification and supercoiling and is considered to be transient. To determine whether the presence of Z-DNA can be detected immunochemically, the binding of monoclonal and polyclonal anti-Z-DNA antibodies to a panel of natural DNA antigens was assessed by an ELISA using brominated poly(dG-dC) as a control for Z-DNA. As these studies showed,

Influence of anions on ozonation of bisphenol AF: Kinetics, reaction pathways, and toxicity assessment.

In this article, the degradation of 4, 4'-(hexafluoroisopropylidene) diphenol (bisphenol AF, BPAF) by ozone was studied and toxicity of the degradation products was evaluated. Kinetic studies showed that acidic conditions were more conducive to the ozone degradation of BPAF than alkaline conditions. In the presence of common anions, Br- and SO42- promoted the degradation of BPAF, whereas NO2-, NO3-, HSO3- inhibited the degradation, and the other anions and cations had no significant effect. The degradation products were analyzed by mass spectrometry, and were mainly manifested in hydroxylation

Excretion characteristics and tissue accumulation of tetrabromobisphenol-A in male rats after sub-chronic inhalation exposure.

Tetrabromobisphenol-A (TBBPA) is an emerging organic pollutant and a commonly used brominated flame retardant that has received much attention owing to its toxicity. Although TBBPA is ubiquitously detected in atmospheric particulate matter and dust, few studies have investigated the sub-chronic inhalation exposure to TBBPA. To further understand the excretion characteristics and tissue accumulation of TBBPA after inhalation exposure, we used the rat model to conduct a sub-chronic inhalation exposure study. Male rats were administered with different doses of aerosol TBBPA (12.9, 54.6, 121.6, an
