Long-term cardiovascular complications following sepsis: is senescence the missing link?

Among the long-term consequences of sepsis (also termed "post-sepsis syndrome") the increased risk of unexplained cardiovascular complications, such as myocardial infarction, acute heart failure or stroke, is one of the emerging specific health concerns. The vascular accelerated ageing also named premature senescence is a potential mechanism contributing to atherothrombosis, consequently leading to cardiovascular events. Indeed, vascular senescence-associated major adverse cardiovascular events (MACE) are a potential feature in sepsis survivors and of the elderly at cardiovascular risk. In the

Senescent cell networks link matrix remodeling and vascular dysfunction in human fibroids.

Uterine fibroids (leiomyomas) are highly prevalent benign tumors defined by excessive extracellular matrix (ECM) deposition, altered vascular structure, and progressive tissue stiffening, yet the cellular programs that coordinate these features remain poorly understood. Cellular senescence has been implicated in fibroid biology, but whether senescence represents a uniform state or distinct, functionally specialized cell identities within fibroids is unknown. Here, we identify the distinct heterogeneous populations of senescent cells ("senotypes") present in human fibroids and characterize thei…

IRAP deficiency preserves renal function and attenuates pathology in aged mice.

Senescent cell accumulation and fibrosis play a fundamental role in kidney ageing. Our aim was to determine whether the angiotensin IV/insul … in 28-month-old WT mice similarly prevented age-related decline in GFR and attenuated expression of cellular senescence markers. Using an in vitro model of cultured proximal tubule cells, IRAP inhibition mitigated the pro-senescent effects of oxidative stress. This was associated with shifts in the metabolic phenotype of proximal tubular cells, indicating the senopro …

Two new species of Capitellidae (Annelida, Polychaeta) from the southern coasts of China.

Senescent cells, which arise due to damage-associated signals, are apoptosis-resistant and can express a pro-inflammatory, tissue-destructiv … s who died from acute SARS-CoV-2 infection than other causes. Our results suggest that induction of cellular senescence and SASP amplification through TLR-3 contribute to SARS-CoV-2 morbidity, indicating that clinical trials of senolytics and/or SASP/TLR-3 inhibitors for alleviating acute and long-term SARS-CoV-2 sequelae are warranted.

Exercise and p21 signaling in central nervous system aging and senescence: A context- and dose-dependent relationship.

Physical exercise is a potent modifier of CNS aging, but its relationship with the cyclin-dependent kinase inhibitor p21 (CDKN1A) is neither uniformly suppressive nor equivalent to cellular senescence. Direct evidence remains sparse and preclinical. Sustained physiological exercise reduced hippocampal or cortical p21-associated signatures in models of metabolic distress, natural aging, and amyloid pathology. Conversely, acute treadmill exercise transiently increased hippocampal Cdkn1a, while spatial transcriptomics identified increased endothelial Cdkn1a during broader exercise-associated vasc…

On-Demand Collaborative Delivery for Remodeling the Senescent Microenvironment in Intervertebral Disc Degeneration.

The senescent microenvironment, defined as the cellular environment surrounding senescent cells, plays a pivotal role in tissue degenerative diseases by promoting inflammation, disrupting extracellular matrix homeostasis, and inducing senescence in neighboring healthy cells. By analyzing the etiology of the senescent microenvironment in intervertebral disc degeneration (IVDD), senescence-associated secretory phenotype (SASP)-positive nucleus pulposus cells (NPCs) and pro-inflammatory macrophages were considered the most likely primary contributors to this pathological microenvironment. HMGB1 Box A gene therapy to alleviate bleomycin-induced pulmonary fibrosis in rats.

BACKGROUND: Pulmonary fibrosis is characterized by the destruction of normal lung tissue and then replacement by abnormal fibrous tissue, leading to an overall decrease in gas exchange function. The effective treatment for pulmonary fibrosis remains unknown. The upstream pathogenesis of pulmonary fibrosis may involve cellular senescence of the lung tissue. Previously, a new gene therapy technology using Box A of the HMGB1 plasmid (Box A) was used to reverse cellular senescence and cure liver fibrosis in aged rats. METHODS: Here, we show that Box A is a promising medicine for the treatment of …

Endoplasmic reticulum stress coordinates perfluorononanoic acid (PFNA)-induced dysfunction in murine preadipocytes.

Perfluorononanoic acid (PFNA), a persistent long-chain per- and polyfluoroalkyl substance (PFAS), is increasingly recognized as a potential metabolic disruptor; however, the cellular and molecular mechanisms underlying its obesogenic activity remain poorly understood. In the present study, we investigated the effects of low nanomolar PFNA concentrations on 3T3-L1 adipocyte differentiation and function, with particular emphasis on the exposure timing during adipogenesis process and the molecular mechanisms underlying PFNA-induced adipose dysfunction. PFNA exposure during the proliferative pread

Mitochondria-targeted esculetin and metformin delay endothelial senescence by promoting fatty acid β-oxidation: Relevance in age-associated atherosclerosis.

Impaired mitochondrial fatty acid β-oxidation (FAO) plays a role in the onset of several age-associated diseases, including atherosclerosis. In the current work, we investigated the efficacies of mitochondria-targeted esculetin (Mito-Esc) and metformin in enhancing FAO in human aortic endothelial cells (HAECs), and its relevance in the delay of cellular senescence and age-associated atherosclerotic plaque formation in Apoe-/- mice. Chronic culturing of HAECs with either Mito-Esc or metformin increased oxygen consumption rates (OCR), and caused delay in senescence features. Conversely, etomoxir…

Chemotherapy and Stem Cell Transplantation Increase p16INK4a Expression, a Biomarker of T-cell Aging.

The expression of markers of cellular senescence increases exponentially in multiple tissues with aging. Age-related physiological changes may contribute to adverse outcomes in cancer survivors. To investigate the impact of high dose chemotherapy and stem cell transplantation on senescence markers in vivo, we collected blood and clinical data from a cohort of 63 patients undergoing hematopoietic cell transplantation. The expression of p16INK4a, a well-established senescence marker, was determined in T-cells before and 6months after transplant. RNA sequencing was performed on paired samples fro…

LSD1-mediated demethylation of the DNA damage response factor ATM promotes senescence and organ aging.

Aging occurs heterogeneously across organs, leading to progressive tissue dysfunction. Cellular senescence is a stress response triggered by age-associated insults, yet the mechanisms regulating senescence and organ aging remain incompletely understood. Here, we defined a role for lysine-specific demethylase 1 (LSD1) in DNA damage-mediated senescence and organ aging. LSD1 was upregulated in aged organs and senescent cells. In response to natural aging or ionizing radiation-induced DNA damage, LSD1 interacted with and demethylated ATM at lysine 3,016, as confirmed using a newly generated ATM-K3…

Defining Microbiota-Derived Metabolite Butyrate as a Senomorphic: Therapeutic Potential in the Age-Related T Cell Senescence.

Advancing age is accompanied by an accumulation of senescent T cells that secrete pro-inflammatory senescence-associated secretory phenotype (SASP) molecules. Gut-microbiota-derived signals are increasingly recognised as immunomodulators. In the current study, we demonstrated that ageing and the accumulation of senescent T cells are accompanied by a reduction in microbial-derived short-chain fatty acids (SCFAs). Culturing aged T cells in the presence of butyrate suppresses the induction of a senescence phenotype and inhibits the secretion of pro-inflammatory SASP factors, such as IL6 and IL8.

Characterization of senescence-associated transcripts in the human placenta.

INTRODUCTION: Fusion of mononucleated cytotrophoblasts into syncytium leads to trophoblast senescence. Yet, premature senescence is associated with preeclampsia, fetal growth restriction (FGR), and related obstetrical syndromes. A set of 28 transcripts that comprise senescence-associated secretory phenotype (SASP) was recently described in placentas from women with preeclampsia. We posited that this transcript set is uniquely regulated in late-term placentas or in placentas derived from participants with major obstetrical syndromes. METHODS: Using our large placental RNAseq bank, we analyzed

Senescence-associated secretory phenotype (SASP) and uterine fibroids: Association with PD-L1 activation and collagen deposition.

Uterine fibroids (or uterine leiomyoma, UFs) are one of the most prevalent benign uterine tumors with high proliferation and collagen synthesis capabilities. UFs are a significant worldwide health issue for women, affecting their physical and financial well-being. Risk factors for UFs include age, racial disparities, obesity, uterine infections, hormonal variation, and lifestyle (i.e., diet, exercise, stress, and smoking). Senescence and its associated secretory phenotypes (SASPs) are among the most salient changes accompanying the aging process. As a result, SASPs are suggested to be one of t

MAPK15 Attenuates Intestinal Epithelial Cell Senescence-Like Phenotype to Regulate Gut Homeostasis and Inflammation by Phosphorylating Suz12 to Suppress Drp1-Mediated Mitochondrial Fission Damage.

The senescence of intestinal epithelial cells (IECs) is closely associated with the development of ulcerative colitis (UC), but the underlying regulatory mechanism remains unclear. Although mitogen activated protein kinase 15 (MAPK15) has been shown to inhibit cellular senescence, its specific role in UC has not yet been clarified. This study investigates the role of MAPK15 in modulating IEC senescence during UC and explores the molecular mechanisms involved in this regulatory process. The senescence phenotype and MAPK15 expression in colon tissues from UC patients, dextran sodium sulfate (DSS…

DHCR24 Insufficiency Promotes Vascular Endothelial Cell Senescence and Endothelial Dysfunction via Inhibition of Caveolin-1/ERK Signaling.

Endothelial cells (ECs) senescence is critical for vascular dysfunction, which leads to age-related disease. DHCR24, a 3β-hydroxysterol δ 24 reductase with multiple functions other than enzymatic activity, has been involved in age-related disease. However, little is known about the relationship between DHCR24 and vascular ECs senescence. We revealed that DHCR24 expression is chronologically decreased in senescent human umbilical vein endothelial cells (HUVECs) and the aortas of aged mice. ECs senescence in endothelium-specific DHCR24 knockout mice was characterized by increased P16 and senesce

Gut microbiota-skin axis in melasma: microbial metabolites and hormonal crosstalk.

The gut microbiota is increasingly recognized as a systemic regulator of host physiology, extending its influence to the skin. Microbial metabolites-including short-chain fatty acids, exopolysaccharides, β-glucuronidase, and S-equol-modulate key processes such as inflammation, estrogen metabolism, and cellular senescence. In melasma, a chronic hyperpigmentation disorder with inflammatory features, dysbiosis alters immunometabolic pathways that sustain disease persistence. Beneficial taxa such as Faecalibacterium prausnitzii, Bacteroides thetaiotaomicron, Lactobacillus, and Bifidobacterium gene…

ISRIB inhibits the senescence of type II pulmonary epithelial cells to alleviate pulmonary fibrosis induced by silica in mice.

The role and mechanisms of integrated stress response inhibitor (ISRIB) on silicosis are still not well defined. In the present study, the effects of ISRIB on cellular senescence and pulmonary fibrosis in silicosis were evaluated by RNA sequencing, micro-computed tomography, pulmonary function assessment, histological examination, and Western blot analysis. The results showed that ISRIB significantly reduced the degree of pulmonary fibrosis in mice with silicosis and reduced the expression of type I collagen, fibronectin, α-smooth muscle actin, and transforming growth factor-β1. Both in vivo a…
