Changes in interstitial pressure during acute interstitial volume depletion in normally hydrated rats.

Interstitial fluid pressure was measured in normally hydrated rats during acute interstitial volume depletion by intravenous hyperoncotic bovine serum albumin infusion. Body fluid volumes, systemic arterial and venous pressure and selected blood and urine variables were also measured. The infusion increased plasma volume twice as much as do iso-oncotic infusions which cause comparable increases in mean central venous pressure. The kidneys responded with a diuresis and natriuresis closely resembling those which follow iso-oncotic infusion in normally hydrated rats. At the end of the elevated re

Compartments and barriers in the sciatic nerve of the rabbit.

Near constant concentration of various radioactively labelled polar nonelectrolytes were maintained in the blood plasma of renal ligated rabbits. The uptake of [14C]urea, [51Cr]EDTA and [14C]inulin into sciatic nerve was interpreted in terms of a two-compartment model. There was a rapidly equilibrating space representing about 20% of the tissue weight which is almost certainly the interstitial fluid of the epineurium and outer layers of the perineurium. Urea penetrated the blood-nerve barrier at a similar rate to the entry of this compound into brain. Sucrose, chromium-EDTA and sulphate passed

Infectious mononucleosis. Epstein-Barr-virus shedding in saliva and the oropharynx.

In an examination of excretion patterns of Epstein-Barr virus in 104 throat washings from 20 patients with infectious mononucleosis we found that three persons regularly shed virus from the second week through the third month after onset; 15 demonstrated intermittent excretion over three months, and in two cases, no virus was detected. In oral secretions, the virus appeared to be located extracellularly. Transforming activity was demonstrated in aliquots after centrifugation and filtration, in a sample in which cells were disrupted before filtration, and in specimens after two years' storage.

Hypertensive cardiomyopathy - histopathological and immunohistochemical aspects.

Arterial hypertension (AH) represents the main cause of morbidity and mortality all over the world. Approximately 40% of the adults aged over 25 years old and about 90% of the persons aged over 80 years old suffer from AH. It is a multifactorial condition, in whose etiopathogeny there are involved numerous genetic, metabolic and environment factors. In its turn, AH is one of the most important risk factors for heart disease, stroke, heart failure, kidney disease and peripheral vascular diseases. In hypertensive patients, it progresses into the left ventricle hypertrophy, as a result of some ma

Acid-base regulation in the toad Bufo marinus during environmental hypoxia.

Gas exchange and the correlated changes in blood and tissue metabolic and acid-base status were investigated during long term exposure of the toad Bufo marinus to graded levels of hypoxia. During hypoxia, PCO2 values in blood and tissues fell, leading to a transient alkalosis in the extracellular but not in the intracellular space. A reduction in blood perfusion of the skin during hypoxia may explain why PCO2 was low in sartorius muscle under normoxia, but approached the PCO2 values found in other tissues (gastrocnemius muscle, ventricle) under hypoxia. At PO2 values below the critical PO2, la

The role of the neutrophil in inflammatory diseases of the lung.

Under certain circumstances, the neutrophil has been implicated in causing disease by damaging normal host tissue. This may occur in the adult respiratory distress syndrome (ARDS). The neutrophil has been implicated since a) substances that activate neutrophils are produced in association with the predisposing risks that lead to ARDS; b) activated neutrophils migrate into the alveolar spaces and their toxic products can be found in lung lavage fluid and in the breath of patients with ARDS; and c) the magnitude of the physiologic alterations correlate with the number of neutrophils in the alveo

When Our Best Friend Becomes Our Worst Enemy: The Mitochondrion in Trauma, Surgery, and Critical Illness.

Common for major surgery, multitrauma, sepsis, and critical illness, is a whole-body inflammation. Tissue injury is able to trigger a generalized inflammatory reaction. Cell death causes release of endogenous structures termed damage associated molecular patterns (DAMPs) that initiate a sterile inflammation. Mitochondria are evolutionary endosymbionts originating from bacteria, containing molecular patterns similar to bacteria. These molecular patterns are termed mitochondrial DAMPs (mDAMPs). Mitochondrial debris released into the extracellular space or into the circulation is immunogenic and …

Extracellular Vesicles: Advanced Tools for Disease Diagnosis, Monitoring, and Therapies.

Extracellular vesicles (EVs) are a heterogeneous group of membrane-encapsulated vesicles released by cells into the extracellular space. They play a crucial role in intercellular communication by transporting bioactive molecules such as proteins, lipids, and nucleic acids. EVs can be detected in body fluids, including blood plasma, urine, saliva, amniotic fluid, breast milk, and pleural ascites. The complexity and diversity of EVs require a robust and standardized approach. By adhering to standardized protocols and guidelines, researchers can ensure the consistency, purity, and reproducibility…

Native and bioengineered extracellular vesicles for cardiovascular therapeutics.

Extracellular vesicles (EVs) are a heterogeneous group of natural particles that are relevant to the treatment of cardiovascular diseases. These endogenous vesicles have certain properties that allow them to survive in the extracellular space, bypass biological barriers and deliver their biologically active molecular cargo to recipient cells. Moreover, EVs can be bioengineered to increase their stability, bioactivity, presentation to acceptor cells and capacity for on-target binding at both cell-type-specific and tissue-specific levels. Bioengineering of EVs involves the modification of the do…

Post-guidance signaling by extracellular matrix-associated Slit/Slit-N maintains fasciculation and position of axon tracts in the nerve cord.

Axon-guidance by Slit-Roundabout (Robo) signaling at the midline initially guides growth cones to synaptic targets and positions longitudinal axon tracts in discrete bundles on either side of the midline. Following the formation of commissural tracts, Slit is found also in tracts of the commissures and longitudinal connectives, the purpose of which is not clear. The Slit protein is processed into a larger N-terminal peptide and a smaller C-terminal peptide. Here, I show that Slit and Slit-N in tracts interact with Robo to maintain the fasciculation, the inter-tract spacing between tracts and t

Activation of extracellular matrix metalloproteases by proteases and organomercurials.

Extracellular matrix metalloproteases are synthesized as proenzymes and are activated by certain physiological agents after secretion into the extracellular space. The identity of these agents and the stimulus that elicits their response in vivo is only recently becoming clear, but a variety of agents or stimuli are capable of activating these metalloproteases in vitro also. Of these, the most well studied and characterized are trypsin, plasmin and the organomercurials. These agents appear to have in common an ability to disrupt the structure of the stable latent enzyme in such a way as to all…

The "glymphatic" mechanism for solute clearance in Alzheimer's disease: game changer or unproven speculation?

How solutes and macromolecules are removed from brain tissue is of central importance in normal brain physiology and in how toxic protein aggregates are cleared in neurodegenerative conditions, including Alzheimer's disease (AD). Conventionally, solute transport in the narrow and tortuous extracellular space in brain parenchyma has been thought to be primarily diffusive and nondirectional. The recently proposed "glymphatic" (glial-lymphatic) hypothesis posits that solute clearance is convective and driven by active fluid transport from para-arterial to paravenous spaces though aquaporin-4 wate…

Amyloidosis of the submaxillary gland.

Amyloidosis can be defined as an extracellular deposit of the amyloid fibrillar protein in one or more locations in the organism. Multiple clinical and biochemical forms of amyloid exist, and are characterized according to their fibrillar structural characteristics. The Red Congo stain presents green fluorescence when histologic cuts are observed by polarized light. The deposits of this substance can be focal in nearly any area of the organism, although the perivascular space tends to be affected. We present the case of a woman operated on for myeloma at the base of the cranium who showed no w

Superficial siderosis of the central nervous system: a neglected cause of sensorineural hearing loss.

Superficial siderosis of the central nervous system is a rare, often unrecognized condition in which there is a deposition of intracellular and extracellular hemosiderin in the leptomeninges, subpial tissue, spinal cord, and cranial nerves as a result of chronic bleeding in the subarachnoid space. On the basis of our cases and a review of the literature, the majority of patients manifest sensorineural hearing loss in addition to cerebellar dysfunction, pyramidal tract signs, and progressive mental deterioration. Recent imaging techniques--particularly CT and MRI--can define the presence of iro

[Electron microscopic study of a tick-borne encephalitis virus-infected cell culture exposed to tunicamycin].

Ultrastructural features of tick-borne encephalitis virus-infected pig embryo cell culture treated with different concentrations of tunicamycin at various intervals after infection were studied electron microscopically. The inhibition of glycosylation did not prevent virion formation in the infected cells. At the same time, treatment with tunicamycin led to marked accumulation of virus particles in cisterns and vacuoles of the Golgi complex and to a decrease in the number of virions released into the extracellular space. It is assumed that inhibition of glycosylation leads to disorders in the …
