Generalized Linear Mixed Models for Changepoint Analysis of Biomedical Time Series Data.

Changepoint analysis for time series data identifies the time points for which properties about the data shift. In this paper, we are motivated by two distinct types of biomedical time series data: digital health monitoring and neuroimaging. The neuroimaging study consists of multivariate time series obtained from resting-state functional magnetic resonance imaging (fMRI). The digital health monitoring data consists of binary univariate time series observed within an individual throughout the day across multiple days of activity. The goal of the analysis is to model how physical activity chang

A statistical framework for integrative imaging genomics with biclustering and ensemble penalized regression in Alzheimer's disease.

Imaging genomics provides a powerful paradigm for decoding the complex interplay between molecular variation and brain functional phenotypes in neurodegenerative disorders. In this study, the molecular modality is specifically gene expression data rather than time-invariant genetic variants. However, statistically integrating these high-dimensional modalities remains challenging, as existing methods often rely on incomplete biological priors or overlook the latent modular structure of the data. To address this, we propose BRIDGE (Biclustering and ensemble penalized Regression for Integrative D

An innovation scalp acupuncture prescription for post-stroke aphasia: A neuroimaging-based validation study.

BACKGROUND AND OBJECTIVE: The coexistence of speech disorders in stroke patients can negatively impact their quality of life and rehabilitation outcomes. Scalp acupuncture (SA) has shown potential as a non-pharmacological treatment for post-stroke aphasia (PSA). As the location of SA in PSA treatment is controversial, this study aims to utilize neuroimaging techniques for identifying and validation the promising target. METHODS: The study was divided into two phases. In phase Ⅰ, three pipelines, including lesion mapping, meta-analysis, and resting-state functional connectivity, were integrate

Early Alzheimer's disease classification via structure and feature-based graph attention network from multi-center data.

With the increasing Alzheimer's disease (AD) prevalence, morbidity, and mortality, its early diagnosis is particularly important. The multi-modal data can be used to analyze intermodal feature relationships and enhance the feature expression ability related to AD. The stable and discriminative features are obtained by fusing multiple regions of interest among different modalities. In this paper, a novel graph attention network is proposed that integrates image as well as non-image information for each subject, leveraging a structure-based, and feature-based approach. Initially, we devise a gro

Development and validation of a machine learning model to predict cognitive behavioral therapy outcome in obsessive-compulsive disorder using clinical and neuroimaging data.

BACKGROUND: Cognitive behavioral therapy (CBT) is a first-line treatment for obsessive-compulsive disorder (OCD), but clinical response is difficult to predict. In this study, we aimed to develop predictive models using clinical and neuroimaging data from the multicenter Enhancing Neuro-Imaging and Genetics through Meta-Analysis (ENIGMA)-OCD consortium. METHODS: Baseline clinical and resting-state functional magnetic imaging (rs-fMRI) data from 159 adult patients aged 18-60 years (88 female) with OCD who received CBT at four treatment/neuroimaging sites were included. Fractional amplitude of

Neural correlates of body size estimation: A systematic review and narrative synthesis.

Inaccurate body size estimation (BSE), the discrepancy between an individual's actual and perceived body size and shape, is observed not onl … to identify brain regions associated with BSE across studies of healthy individuals. Studies using functional neuroimaging were selected if they elicited BSE with a task, contrasted BSE with a control task, and used whole-brain analyses (rather than being restricted to a priori regions of interest). This yielded a set of nine functional magnetic resonance imaging papers …

Alterations in dynamic regional homogeneity and functional connectivity in lung cancer patients with bone metastasis pain.

BACKGROUND: Although previous neuroimaging studies have characterized static brain activity in bone metastasis pain (BMP), its dynamic functional properties remain largely unexplored. This study aimed to investigate the dynamic brain activity in BMP patients. METHODS: We analyzed dynamic regional homogeneity (dReHo) and dynamic functional connectivity (dFC) in 50 right-handed lung cancer patients with BMP(+), 36 without BMP(-), and 32 healthy controls (HCs). Spearman's correlation was used to evaluate associations with clinical variables. RESULTS: Compared to BMP(-) patients, BMP(+) patients

Causal functional maps of brain rhythms in working memory.

Human working memory is a key cognitive process that engages multiple functional anatomical nodes across the brain. Despite a plethora of correlative neuroimaging evidence regarding the working memory architecture, our understanding of critical hubs causally controlling overall performance is incomplete. Causal interpretation requires cognitive testing following safe, temporal, and controllable neuromodulation of specific functional anatomical nodes. Such experiments became available in healthy humans with the advance of transcranial alternating current stimulation (tACS). Here, we synthesize

Abnormal spontaneous neural activity in hippocampal-cortical system of patients with obsessive-compulsive disorder and its potential for diagnosis and prediction of early treatment response.

Early brain functional changes induced by pharmacotherapy in patients with obsessive-compulsive disorder (OCD) in relation to drugs per se or because of the impact of such drugs on the improvement of OCD remain unclear. Moreover, no neuroimaging biomarkers are available for diagnosis of OCD and prediction of early treatment response. We performed a longitudinal study involving 34 patients with OCD and 36 healthy controls (HCs). Patients with OCD received 5-week treatment with paroxetine (40 mg/d). Resting-state functional magnetic resonance imaging (fMRI), regional homogeneity (ReHo), support

Unpacking common and distinct neuroanatomical alterations in cocaine dependent versus pathological gambling.

Pathological gambling and cocaine dependence are highly pervasive disorders. Functional neuroimaging evidence implicates aberrant activity of prefrontal striatal pathways in both disorders. It is unclear if the neuroanatomy of these areas is also affected. Participants with pathological gambling (n = 18), cocaine dependence (n = 19) and controls (n = 21) underwent high-resolution structural MRI scan and cognitive assessments. In line with emerging functional neuroimaging findings, we hypothesised (i) lower volumes of corticostriatal areas ascribed to decision-making/inhibitory control, craving…

Binary Classification Using Neural and Clinical Features: An Application in Fibromyalgia With Likelihood-Based Decision Level Fusion.

Among several features used for clinical binary classification, behavioral performance, questionnaire scores, test results, and physical exam reports can be counted. Attempts to include neuroimaging findings to support clinical diagnosis are scarce due to difficulties in collecting such data, as well as problems in integration of neuroimaging findings with other features. The binary classification method proposed here aims to merge small samples from multiple sites so that a large cohort, which better describes the features of the disease can be built. We implemented a simple and robust framew

Neural dysfunction in ADHD with Reading Disability during a word rhyming Continuous Performance Task.

BACKGROUND: Attention-Deficit/Hyperactivity Disorder (ADHD) is a heterogeneous, neurodevelopmental disorder which co-occurs often with Reading Disability (RD). ADHD with and without RD consistently have higher inattentive ratings compared with typically developing controls, with co-occurring ADHD and RD also demonstrating impaired phonological processing. Accordingly, inattention has been associated with greater phonological impairment, though the neural correlates of the association are poorly understood from a functional neuroimaging perspective. It was postulated that only the co-occurring …

Neuroimaging of mobility in aging: a targeted review.

BACKGROUND: The relationship between mobility and cognition in aging is well established, but the relationship between mobility and the structure and function of the aging brain is relatively unknown. This, in part, is attributed to the technological limitations of most neuroimaging procedures, which require the individual to be immobile or in a supine position. Herein, we provide a targeted review of neuroimaging studies of mobility in aging to promote (i) a better understanding of this relationship, (ii) future research in this area, and (iii) development of applications for improving mobili

Brain connectivity and visual attention.

Emerging hypotheses suggest that efficient cognitive functioning requires the integration of separate, but interconnected cortical networks in the brain. Although task-related measures of brain activity suggest that a frontoparietal network is associated with the control of attention, little is known regarding how components within this distributed network act together or with other networks to achieve various attentional functions. This review considers both functional and structural studies of brain connectivity, as complemented by behavioral and task-related neuroimaging data. These studies

Spontaneous brain activity in combat related PTSD.

Posttraumatic stress disorder (PTSD) is a prevalent psychiatric disorder, especially in combat veterans. Existing functional neuroimaging studies have provided important insights into the neural mechanisms of PTSD using various experimental paradigms involving trauma recollection or other forms of emotion provocation. However it is not clear whether the abnormal brain activity is specific to the mental processes related to the experimental tasks or reflects general patterns across different brain states. Thus, studying intrinsic spontaneous brain activity without the influence of external task…

Normal amygdala activation but deficient ventrolateral prefrontal activation in adults with bipolar disorder during euthymia.

Functional neuroimaging studies have implicated the involvement of the amygdala and ventrolateral prefrontal cortex (vlPFC) in the pathophysiology of bipolar disorder. Hyperactivity in the amygdala and hypoactivity in the vlPFC have been reported in manic bipolar patients scanned during the performance of an affective faces task. Whether this pattern of dysfunction persists during euthymia is unclear. Using functional magnetic resonance imaging (fMRI), 24 euthymic bipolar and 26 demographically matched healthy control subjects were scanned while performing an affective task paradigm involving …

Neuropsychiatric symptoms in the dementias.

PURPOSE OF REVIEW: Neuropsychiatric, or non-cognitive symptoms are increasingly recognized as manifestations of dementias. RECENT FINDINGS: In Alzheimer's disease, recent advances have included the identification of behavioral profiles, differentiation of apathy and depression, characterization of risk factors for psychosis and its links to agitation and aggression, and an analysis of depressive symptoms in the absence of major depression. Functional neuroimaging data mainly supported the role of the anterior cingulate in apathy. The orbitofrontal and anterior cingulate tangle burden were ass…

The Mediating Roles of Neurobiomarkers in the Relationship Between Education and Late-Life Cognition.

BACKGROUND: The mediating roles of neuropathologies and neurovascular damage in the relationship between early-life education and later-life cognitive function are unknown. OBJECTIVE: To examine whether Alzheimer's and neurovascular biomarkers mediate the relationships between education and cognitive functions. METHODS: Data were from 537 adults aged 55-94 in the Alzheimer's Disease Neuroimaging Initiative 3. We tested whether the relationships between education (continuous, years) and cognitive function (memory, executive functioning, and language composites) were mediated by neuroimaging b

Daytime impairment and neurodegeneration in OSAS.

Controversy surrounds the pathogenesis of neurocognitive daytime dysfunction exhibited by patients with obstructive sleep apnea syndrome (OSAS). Underlying brain dysfunctions and damage have long been suspected as a cause of some of this impairment. Neuroimaging has enabled scientists to test these long-held theories. This paper is based on a comprehensive review of recent publications on neuroimaging studies in this area. It seeks to highlight results of recent research, which suggest connections between persistent neurocognitive daytime impairment of executive functions, underlying signs of
