Copper-dependent enzymes catalyze some of nature's most challenging oxidative transformations, yet many copper cofactors lack distinctive spectroscopic signatures or are kinetically labile, complicating their discovery and mechanistic characterization. Recent advances in genome mining, heterologous expression, and protein structural prediction have accelerated the identification of previously unrecognized copper enzymes. This Current Opinion highlights two emerging families of binuclear type 2 copper enzymes: plant-specific BURP domain peptide cyclases (BpCs) and fungal DUF3328 dicopper oxidat

Intestinal stem cells (ISCs) sustain epithelial homeostasis and regeneration in a niche that is shaped by epithelial, stromal, immune, and microbial inputs. This review examines how bacterial and fungal signals influence ISC activity and epithelial repair through microbial metabolites, immune relays, and epithelial sensing. Bacterial metabolites and structural ligands act through Wnt/β-catenin, AHR, IL-22-STAT3, Hippo-YAP, and related pathways, and the effects vary according to the ligand involved, responding cell type, and tissue state. Evidence for fungi is more limited: some fungal products

Deep mucosal and organ infections caused by the infestation of Candida albicans in immunocompromised patients represent a significant cause of mortality in hospitalized patients. The rise in fungal resistance is a consequence of the overuse of antibiotics. Therefore, innovative immunostimulants must be developed to combat pathogenic fungal infections. We used urolithin A (UA), an intestinal metabolite rich in the naturally occurring polyphenolic antioxidants ellagic acid (EA) or ellagitannin (ET), as a lead compound for structural modification. Through liquid screening of 17 synthesized compou

To combat the ineffectiveness of currently available pharmaceutical medications, caused by the emergence of increasingly resistant bacterial and fungal strains, novel antibacterial and antifungal medications are urgently needed. Novel natural compounds with antimicrobial activities can be obtained by exploring underexplored habitats such as the world's oceans. The oceans represent the largest ecosystem on earth, with a high diversity of organisms. Oceans have received some attention in the past few years, and promising compounds with antimicrobial activities were isolated from marine organisms

With the rapid growth of fungal infections and the emergence of multi-drug resistant (MDR) fungal strains, new antifungals with novel mechanisms are a pressing need to tackle this emerging health problem. Herein it is reported for the first time that hyperbranched polylysine (HPL) shows antifungal activities against Candida, especially for drug-sensitive and MDR C. albicans strains, and broad-spectrum antibacterial activities against both Gram-negative and Gram-positive bacteria. The high antimicrobial activities are ascribed to the high charge density and compact size of the globular structur

Fungal urinary tract infection (UTI) represents a high-risk event in severely ill patients. Its increasing incidence in recent years is associated with extensive and prolonged use of broad-spectrum antimicrobial agents, corticosteroids, immunosuppressive and cytotoxic drugs. Other important risk factors comprise higher age, diabetes mellitus, chronic renal failure, hemodialysis, renal transplantation, structural or functional abnormalities of urinary tract with indwelling urinary catheter or nephrostomy. Fifty hospitalized symptomatic patients with funguria of > 10(5) CFU/ml and leucocyturia w

Genomic studies of Candida auris are underpinned by the generation of high-quality genome assemblies. These reference genomes have been essential for investigations of the evolution and epidemiology of this emerging fungal pathogen. In addition to genomic epidemiology studies of local outbreaks and analysis of the global emergence of this species, comparisons of genomes of isolates from the five major clades have revealed differences in gene content and genomic structure. Here, we provide a detailed protocol for generating complete genome assemblies for C. auris.

Fungal secretomes are prolific sources of structurally diverse bioactive natural products, including peptaibols, lipopeptides, and polar metabolites. Their chemical complexity makes comprehensive untargeted profiling a persistent analytical challenge, particularly for highly polar compounds that are poorly retained in conventional reversed-phase liquid chromatography (RPLC). This study presents the development and optimization of a supercritical fluid chromatography (SFC) method coupled to high-resolution tandem mass spectrometry (HRMS/MS) for the untargeted metabolite profiling of a Trichoder

Candida albicans is the pathogenic fungus that most frequently causes infections in humans. It is part of the microbiota commonly found in the skin, gastrointestinal tract, and vaginal mucosa. However, certain conditions, including immunosuppression, excessive use of antibiotics, hormonal changes, the use of medical devices in patients, and individual nutritional status, promote the development of opportunistic infections caused by this fungus. One of the main fungal structures interacting with the host is the cell wall, which is principally composed of chitin, glucan, and proteins. The cell w…

Neuraminidases (NAs) are glycoside hydrolase enzymes pivotal in carbohydrate metabolism, ubiquitously present in viruses, bacteria, fungi, and mammals. These enzymes catalyze the cleavage of terminal sialic acid residues from glycoproteins and glycolipids, impacting various biological processes, including pathogen infections and cancer cell proliferation. In our study, we employed advanced in silico strategies to repurpose existing drugs, aiming to provide a rapid response to health emergencies posed by multi-drug-resistant bacteria and fungi, as well as expanding the arsenal of antiviral ther

Xylobolus subpileatus is a widely distributed crust fungus reported from all continents except Antarctica, although considered a rare species in several European countries. Profound mycochemical analysis of the methanol extract of X. subpileatus resulted in the isolation of seven compounds (1-7). Among them, (3β,22E)-3-methoxy-ergosta-4,6,814,22-tetraene (1) is a new natural product, while the NMR assignment of its already known epimer (2) has been revised. In addition to a benzohydrofuran derivative fomannoxin (3), four ergostane-type triterpenes 4-7 were identified. The structure elucidation

Application of synthetic herbicides is currently the most widely used and cost-effective methods to assist with revegetation programs. However, the effects of short-term application of herbicides such as Roundup®, acetic acid, BioWeed™ and Slasher® as compared with mulch, on soil microbial biomass and microbial diversity remain unknown. This study examined the effects of short-term herbicide application on soil microbial biomass, C:N ratio, and fungal and bacterial communities at months 2 and 8 following initiation of treatment application. No effects of treatments on soil pH, C:N and microbia

There is a critical need for new antifungal drugs; however, the lack of available fungus-specific targets is a major hurdle in the development of antifungal therapeutics. Spore germination is a differentiation process absent in humans that could harbor uncharacterized fungus-specific targets. To capitalize on this possibility, we developed novel phenotypic assays to identify and characterize inhibitors of spore germination of the human fungal pathogen Cryptococcus. Using these assays, we carried out a high-throughput screen of ∼75,000 drug-like small molecules and identified and characterized

Wheat (Triticum aestivum L.) is a major crop grown on all continents. Due to environmental concerns, it is desirable to reduce the inputs of both chemical pesticides and inorganic fertilizers. However, yield reduction must be expected when switching to low-input systems. To mitigate such losses, the use of natural or introduced microbiomes may provide the key to maintaining sustainable yield. Phyllosphere fungi, both endophytic and phylloplane-associated, colonize aboveground plant structures, some of which have the potential to mitigate biotic and abiotic stressors. A first step toward realiz

Alternative proteins from plant, algal, fungal, and microbial sources are increasingly recognized for their potential to influence gut microbiota. However, the role of protein structure in shaping these interactions remains poorly understood. This review examines how key structural attributes of alternative proteins-including amino acid composition, folding characteristics, aggregation behavior, and enzymatic accessibility, govern gastrointestinal digestion. It also explores how these structural features affect microbial fermentation and the subsequent nutritional and functional outcomes. Stru

UNLABELLED: The identification of enzymes responsible for oxidation of lignin in lignin-degrading bacteria is of interest for biotechnological valorization of lignin to renewable chemical products. The genome sequences of two lignin-degrading bacteria, Ochrobactrum sp., and Paenibacillus sp., contain no B-type DyP peroxidases implicated in lignin degradation in other bacteria, but contain putative multicopper oxidase genes. Multi-copper oxidase CueO from Ochrobactrum sp. was expressed and reconstituted as a recombinant laccase-like enzyme, and kinetically characterized. Ochrobactrum CueO shows

The urgent need for sustainable therapeutic nanomaterials has driven interest in green synthesis routes. In this study, zinc oxide-manganese oxide nanocomposites were mycosynthesized by employing the fungal extracellular filtrate of Aspergillus terreus. Characterization based on different spectroscopical analysis confirmed their crystalline structure, associated functional groups, and nanoscale morphology. The obtained composites showed nanoscales with average particle sizes of 75 nm and 99 nm as determined by TEM and DLS analysis, respectively. Additionally, microbiological assays revealed th

We present a case of acute Candida glabrata prostatitis complicated by secondary candidemia in a 72-year-old man with poorly controlled Type 2 diabetes mellitus and benign prostatic hyperplasia, without classical healthcare-associated risk factors for invasive candidiasis. The patient presented with fever, urinary symptoms, and markedly elevated prostate-specific antigen. Urine and blood cultures confirmed disseminated C. glabrata infection. Despite a comprehensive diagnostic workup, no alternative source of candidemia was identified, supporting primary prostatic infection with secondary hemat
