Tandem Kinase WTK1 Triggers Waves of Transcriptional Reprogramming Underlying Wheat Resistance to Stripe Rust.

Tandem kinases are emerging regulators of plant immunity, but the downstream defense programs they activate remain poorly understood. Stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is a major constraint on global wheat production. The Yr15 resistance gene, derived from wild emmer wheat, encodes the tandem kinase protein WTK1. To define the temporal basis of this resistance, we analysed resistant and susceptible near-isogenic lines across a 7-day infection time course using histopathology, ROS phenotyping, RNA sequencing, co-expression network analysis, and qRT-PCR validation

Infection with Tomato Mosaic Virus in Nicotiana tabacum cv. Samsun-Physiological and Molecular Approach During Early Stage of Infection.

Although tomato mosaic virus (ToMV) is an economically important tobamovirus, the physiological and molecular events occurring during the asymptomatic phase of infection remain poorly understood. In this study, we investigated the early responses of Nicotiana tabacum cv. Samsun to ToMV infection using an integrated physiological, biochemical, photosynthetic, and molecular approach. Viral accumulation was quantified via RT-PCR. Oxidative stress markers, antioxidant systems, photosynthetic performance, and gene expression were analyzed at 7 days post inoculation (DPI), before visible symptoms de

The AcLFYL1-AcCSE module enhances kiwifruit resistance to Pseudomonas syringae pv. actinidiae by regulating cell density and lignin biosynthesis.

Canker disease caused by Pseudomonas syringae pv. actinidiae (Psa) is a severe bacterial infection threatening global kiwifruit production. Psa causes lignin degradation, cell wall rupture, leaf wilting, and canker formation on branches and trunks, often leading to plant death. The plant cell wall serves as a structural barrier against pathogens, with its thickness, composition, and cell density influencing disease resistance. Comparative studies between resistant germplasms Actinidia eriantha "Maohuaxiong" (A. eriantha 'MHX') and Actinidia latifolia "Kuoye" (A. latifolia 'KY') and susceptible

Label-Free Proteomics Reveals the Response of Oat (Avena sativa L.) Seedling Root Respiratory Metabolism to Salt Stress.

Soil salinity is among the crucial factors influencing agricultural productivity of crops, including oat. The respiratory metabolic pathways are of great significance for plants to adapt to salt stress, but current research is limited and there are few reports on salt-tolerant crops such as oat, which is necessary to conduct in-depth research. In this study, we conducted a pot experiment to determine the effects of salt stress on oat root growth and respiratory metabolism. Three salt stress levels-control (CK), moderate, and severe-were applied to compare the salt tolerance of the salt-toleran

Identification of sugar transporter MrSTP20 responding to high-salt stress and hexoses induction in apple rootstock (Malus robusta Rehd).

BACKGROUND: Sugar distribution regulation, especially in the roots, is an important pathway for plants to cope with abiotic stress. Sugar transport proteins (STPs) mediate the transmembrane transport of hexoses such as glucose and fructose; however, the molecular mechanism underlying the response to salt stress remains unclear. RESULTS: Here, we identified 22 STP genes in apple (Malus domestica) using genome-wide screening. Under salt stress, 17 genes were detected in the roots of the apple rootstock Malus robusta Rehd; of them, MrSTP3, MrSTP7, MrSTP14, and MrSTP20 showed stronger positive re

The DYW-type pentatricopeptide repeat protein DEK618 interacts with ZmMORFs to modulate mitochondrial RNA editing and kernel development in maize.

C-to-U editing is a critical post-transcriptional modification process in plant mitochondrial RNAs, involving the conversion of specific cytidine (C) residues to uridine (U). Pentatricopeptide repeat (PPR) proteins serve as site-specific recognition factors in this process. Although certain DYW-type PPR proteins (DYW-PPR) are known to participate in the post-transcriptional processing of mitochondrial RNAs, the functions of more than 60 DYW-PPR proteins in maize remain uncharacterized. In this study, we identify a novel DYW-PPR protein, DEK618, which is associated with defective kernel develop

Exogenous Hydrogen sulfide attenuates cadmium toxicity to Chrysanthemum (Chrysanthemum indicum) by modulating glutathione synthesis and cadmium adsorption capacity in the cell wall.

Soil cadmium (Cd) contamination leads to plant toxicity and poses a risk to human health both directly and indirectly through the food chain. Hydrogen sulfide (H2S), a novel gaseous signaling molecule, has been shown to enhance plant tolerance to various abiotic stresses. In this study, the potential of H2S in mitigating Cd toxicity in chrysanthemum (Chrysanthemum indicum) was investigated through physiological, biochemical and transcriptomic analyses. Results showed that the application of exogenous H2S resulted Cd accumulation in the roots by 21.15 %, while reducing Cd in the aboveground par

Hydrogen sulfide promotes lateral root formation in peach through persulfidation of SnRK1α kinase.

Root development is crucial for the growth and yield of horticultural crops. Hydrogen sulfide (H2S), an important gasotransmitter, has been shown to regulate lateral root (LR) formation in plants, including peach (Prunus persica). However, its specific regulatory mechanism remains largely unclear. Here, we show that the energy/metabolic sensor SUCROSE NON-FERMENTING RELATED KINASE 1 (SnRK1) mediates the control of peach LR growth by H2S. PpSnRK1 activity in peach roots is enhanced with H2S to promote LR generation. Cys419, Cys430 and Cys505 residues in the catalytic α-subunit of PpSnRK1 are mo

Mutation of TaNRAMP5 impacts cadmium transport in wheat.

Cadmium (Cd) pollution significantly impacts the normal growth, development, and food safety of wheat. Employing modern molecular biology techniques represents an effective strategy for cultivating low-Cd wheat. Natural resistance-associated macrophage protein 5 (NRAMP5) is a critical heavy metal transporter, however, its function in wheat, particularly in response to Cd stress, remains largely unexplored. Here, we employed the CRISPR/Cas9 gene-editing technology to generate TaNRAMP5 knockout lines (KO). Cd content in wheat was detected by inductively coupled plasma mass spectrometry (ICP-MS).

The inhibition of endo-β-1,4-glucanase is required for the resistance of Lilium regale Wilson to Fusarium wilt.

Fusarium wilt is a significant problem restricting the growth of the lily industry. However, Lilium regale Wilson is a valuable germplasm resource because of its Fusarium Wilt resistance. In this study, six endo-β-1,4-glucanase genes (Cel1-Cel6) in L. regale were identified among the differentially expressed genes reveled by omics data for L. regale infected Fusarium osysporum, which causes Fusarium wilt. Interestingly, the abundance of a few proteins and the expression of endo-β-1,4-glucanase genes, including LrCel1, decreased in L. regale roots infected with F. oxysporum. Thus, the function

Changes in primary metabolism and associated gene expression during host-pathogen interaction in clubroot resistance of Brassica napus.

The role of primary metabolism during Brassica napus-Plasmodiophora brassicae interaction leading to clubroot resistance has not yet been investigated thoroughly. In this study, we investigated some of the primary metabolites and their derivatives as well as expression of the genes involved in their biosynthesis to decipher this host-pathogen interaction. For this, two sets (clubroot resistant and susceptible) of canola lines were inoculated with P. brassicae pathotype 3A to investigate the endogenous levels of primary metabolites at 7-, 14-, and 21-days after inoculation (DAI). How Histone Acetyltransferases Shape Plant Photomorphogenesis and UV Response.

Higher plants have developed complex mechanisms to adapt to fluctuating environmental conditions with light playing a vital role in photosynthesis and influencing various developmental processes, including photomorphogenesis. Exposure to ultraviolet (UV) radiation can cause cellular damage, necessitating effective DNA repair mechanisms. Histone acetyltransferases (HATs) play a crucial role in regulating chromatin structure and gene expression, thereby contributing to the repair mechanisms. HATs facilitate chromatin relaxation, enabling transcriptional activation necessary for plant development

Hydrogen sulfide antagonizes cytokinin to change root system architecture through persulfidation of CKX2 in Arabidopsis.

Hydrogen sulfide (H2S) is an endogenous gaseous signaling molecule, which has been shown to play an important role in plant growth and development by coupling with various phytohormones. However, the relationship between H2S and cytokinin (CTK) and the mechanisms by which H2S and CTK affect root growth remain poorly understood. Endogenous CTK was analyzed by UHPLC-ESI-MS/MS. Persulfidation of cytokinin oxidase/dehydrogenases (CKXs) was analyzed by mass spectrometry (MS). ckx2/CKX2wild-type (WT), OE CKX2 and ckx2/CKX2Cys(C)62alanine(A) transgenic lines were isolated with the ckx2 background. A novel OsGST gene encoding 9glutathione reductase negatively regulates cadmium accumulation in rice.

Cadmium (Cd) accumulates in rice and then moves up the food chain, causing serious health problems for humans. Glutathione S-transferase (GST) binds exogenous hazardous compounds to glutathione (GSH), which performs a variety of roles in plant responses to Cd stress. Here, Cd stimulated the transcripts of a novel OsGST gene, and the OsGST protein, which was localized in the nucleus and cytoplasm, was also induced by Cd. In OsGST deletion mutant lines generated by CRISPR/Cas9, more Cd was accumulated, and Cd hypersensitive phenotypes were observed, while transgenic lines overexpressing OsGST ex

TaNAM-6A is essential for nitrogen remobilisation and regulates grain protein content in wheat (Triticum aestivum L.).

Grain protein content (GPC) is a crucial quality trait in bread wheat, which is influenced by the key transcription factor TaNAM. However, the regulatory mechanisms of TaNAM have remained largely elusive. In this study, a new role of TaNAM was unveiled in regulating nitrogen remobilisation which impacts GPC. The TaNAM knockout mutants generated by clustered regularly interspaced short palindromic repeats/Cas9 exhibited significantly delayed senescence and lower GPC, while overexpression of TaNAM-6A resulted in premature senility and much higher GPC. Further analysis revealed that TaNAM directl

The D14-SDEL1-SPX4 cascade integrates the strigolactone and phosphate signalling networks in rice.

Modern agriculture needs large quantities of phosphate (Pi) fertilisers to obtain high yields. Information on how plants sense and adapt to Pi is required to enhance phosphorus-use efficiency (PUE) and thereby promote agricultural sustainability. Here, we show that strigolactones (SLs) regulate rice root developmental and metabolic adaptations to low Pi, by promoting efficient Pi uptake and translocation from roots to shoots. Low Pi stress triggers the synthesis of SLs, which dissociate the Pi central signalling module of SPX domain-containing protein (SPX4) and PHOSPHATE STARVATION RESPONSE p

Physiological and proteomic dissection of the rice roots in response to iron deficiency and excess.

Iron (Fe) disorder is a pivotal factor that limits rice yields in many parts of the world. Extensive research has been devoted to studying how rice molecularly copes with the stresses of Fe deficiency or excess. However, a comprehensive dissection of the whole Fe-responsive atlas at the protein level is still lacking. Here, different concentrations of Fe (0, 40, 350, and 500 μM) were supplied to rice to demonstrate its response differences to Fe deficiency and excess via physiological and proteomic analysis. Results showed that compared with the normal condition, the seedling growth and conten

Phosphate-induced resistance to pathogen infection in Arabidopsis.

In nature, plants are concurrently exposed to a number of abiotic and biotic stresses. Our understanding of convergence points between responses to combined biotic/abiotic stress pathways remains, however, rudimentary. Here we show that MIR399 overexpression, loss-of-function of PHOSPHATE2 (PHO2), or treatment with high phosphate (Pi) levels is accompanied by an increase in Pi content and accumulation of reactive oxygen species (ROS) in Arabidopsis thaliana. High Pi plants (e.g., miR399 overexpressors, pho2 mutants, and plants grown under high Pi supply) exhibited resistance to infection by ne
