Genetic evidence for the effect of prepregnancy maternal metabolic status on the risk of congenital heart disease in offspring: A Mendelian randomization study.

Congenital heart disease (CHD) is the most common birth defect in the world and is still one of the most common causes of illness and death in babies. There are links between maternal metabolic factors, like glycemic and lipid profiles, and the disease, but it is not clear if these links are causal. Using a two-sample Mendelian randomization (MR) method, this study looked at whether maternal HbA1c, fasting glucose, and triglyceride levels could cause an increase in the risk of CHD in offspring. We selected independent genome-wide significant single-nucleotide polymorphisms (SNPs) (P < 5 × 10-8

Dietary factors mediated the causal association between educational attainment and chronic periodontitis: A 2-sample Mendelian randomization study.

Dietary factors, influenced by educational attainment (EA) have the potential to affect development and course of chronic periodontitis (CP), whereby, the potential mediating role of dietary factors in the casual associations remain unclear. This study employed Mendelian randomization (MR) to explore the causal effect of EA on CP and to identify and quantify the mediating role of dietary factors, aiming to provide evidence for improving CP management. EA was considered as exposure, dietary factors as mediator, and CP as outcome. Genome-wide association study summary statistics were obtained fr

A complex mechanism translating variation of a simple genetic architecture into alternative life histories.

Understanding the processes that link genotype to phenotype is a central challenge in biology. Despite progress in discovering genes associated with ecologically relevant traits, a poor understanding of the processes and functions via which molecules mediate evolutionary differences leaves us critically far from linking proximate and ultimate causes of evolution. This knowledge gap is particularly large in multifaceted phenotypes of ecological relevance such as life histories where multiple traits covary and influence fitness. In Atlantic salmon (Salmo salar), variation in a key life-history t

Mediators of the association between depression and migraine: a mendelian randomization study.

BACKGROUND: An association between depression and migraine has been reported in observational studies; however, conventional observational studies are prone to bias. This study aims to investigate the causal relationship between depression and migraine and to quantify the mediating effects of known risk factors. METHODS: We applied two-sample Mendelian randomization and utilized single nucleotide polymorphisms as genetic instruments for exposure (depression) and mediators (sleep traits). We utilized summary data on genome-wide association studies for depression, sleep-related traits mediators

Causal associations of antioxidants with Alzheimer's disease and cognitive function: a Mendelian randomisation study.

BACKGROUND: Circulating antioxidants are associated with a lower risk of Alzheimer's disease (AD) in observational studies, suggesting potential target areas for intervention. However, whether the associations are causal remains unclear. Here, we studied the causality between antioxidants and AD or cognitive function using two-sample Mendelian randomisation (MR). METHODS: Single nucleotide polymorphisms strongly (p<5×10-8) associated with antioxidants (vitamin A, vitamin C, zinc, selenium, β-carotene and urate) and outcomes (AD, cognitive performance and reaction time) were obtained from the

No evidence for a causal contribution of bioavailable testosterone to ADHD in sex-combined and sex-specific two-sample Mendelian randomization studies.

The higher prevalence of attention-deficit/hyperactivity disorder (ADHD) in males raises the question of whether testosterone is implicated in ADHD risk. However, cross-sectional studies did not identify an association between ADHD and testosterone levels. Mendelian randomization (MR) studies can overcome limitations inherent to association studies, especially of reverse causation and residual confounding. In the current study, sex-combined and sex-specific two-sample MR analyses were conducted to address whether testosterone has a causal influence on ADHD risk. Sex-combined as well as sex-spe

Impact of SGLT2 inhibitors on lower limb complications: a mendelian randomization perspective.

BACKGROUND: While Sodium-glucose cotransporter 2 (SGLT2) inhibitors are effective in managing diabetes and reducing cardiovascular risk, concerns about their association with lower limb complications, including, osteomyelitis, ulcers, and peripheral artery disease (PAD), persist. This study employs Mendelian Randomization (MR) to assess the causal relationship between SGLT2 inhibitors and these lower limb safety outcomes. METHODS: A two-sample drug-target MR approach was used, complemented by a one-sample MR and genetic association analysis. Six SNPs were selected as instrumental variables to

Causal relationships between leukocyte telomere length and female reproductive system diseases: a bidirectional Mendelian randomization study.

OBJECTIVE: Although numerous observational studies have revealed a correlation between leukocyte telomere length (LTL) and female reproductive system diseases (RSDs), the findings of these studies have tended to be consistent. In this study, we accordingly sought to clarify the causal relationships between LTL and RSDs. METHODS: We performed a bidirectional two-sample Mendelian randomization (MR) analysis using pooled statistics from genome-wide association studies of LTL and nine female RSDs. The final results were analyzed using five MR methods, with the inverse variance weighted (IVW) meth

Autoimmune Diseases and Risk of Non-Hodgkin Lymphoma: A Mendelian Randomisation Study.

BACKGROUND: Non-Hodgkin lymphoma (NHL) is one of the most common haematologic malignancies in the world. Despite substantial efforts to identify causes and risk factors for NHL, its aetiology is largely unclear. Autoimmune diseases have long been considered potential risk factors for NHL. We carried out Mendelian randomisation (MR) analyses to examine whether genetically predicted susceptibility to ten autoimmune diseases (Behçet's disease, coeliac disease, dermatitis herpetiformis, lupus, psoriasis, rheumatoid arthritis, sarcoidosis, Sjögren's syndrome, systemic sclerosis, and type 1 diabetes

No causal link between herpes zoster and ischemic stroke: evidence from Mendelian randomization study.

OBJECTIVE: The association between herpes zoster (HZ) and stroke has been the subject of much previous research. Nevertheless, the connection remains ambiguous. A two-sample Mendelian randomisation study was conducted to explore the potential causal link between HZ and ischaemic stroke, including its subtypes. METHODS: For our MR analysis, we identified genetic instrumental variables related to both HZ and stroke by screening two prominent publicly accessible genome-wide association study databases. The primary approach involved using the inverse variance weighting method. To supplement this,

The causal role of circulating vitamin D concentrations in human complex traits and diseases: a large-scale Mendelian randomization study.

Vitamin D has been associated with a variety of human complex traits and diseases in observational studies, but a causal relationship remains unclear. To examine a putative causal effect of vitamin D across phenotypic domains and disease categories, we conducted Mendelian randomization (MR) analyses using genetic instruments associated with circulating 25-hydroxyvitamin D [25(OH)D] concentrations. We leveraged genome-wide significant 25(OH)D-associated SNPs (N = 138) from a meta-analysis combining a vitamin D GWAS conducted in 401,460 white British UK Biobank (UKBB) participants and an indepen

Does Maternal Normal Range Thyroid Function Play a Role in Offspring Birth Weight? Evidence From a Mendelian Randomization Analysis.

BACKGROUND: The association between normal range thyroid function and offspring birth weight has been postulated, but evidence from observational studies is prone to be confounded. We conducted a two-sample Mendelian randomization (MR) study to explore the causal effects of maternal thyroid stimulating hormone (TSH) and free thyroxine (FT4) on birth weight. METHODS: We utilized public shared summary-level statistics from European-ancestry genome wide association studies. We obtained 40 and 21 single nucleotide polymorphisms as instrumental variables, which were associated with TSH and FT4 lev

Association between clonal hematopoiesis and periodontitis: a two-sample mendelian randomization study.

Clonal hematopoiesis (CH) is an age-associated phenomenon characterized by the proliferation of blood cells carrying somatic mutations. Recent studies suggest a potential link between CH and periodontitis, but the causal relationship remains unclear. A two-sample Mendelian randomization (MR) analysis was implemented, examining whether CH phenotypes are causally associated with periodontitis. CH phenotypes included overall clonal hematopoiesis (CH-overall), CH with a DNMT3A mutation (CH-DNMT3A), CH with a TET2 mutation (CH-TET2), large clones (CH-large), and small clones (CH-small). Genetic ins

Unraveling the Association Between Cheese Consumption and Non-Alcoholic Fatty Liver Disease: Insights From a Two-Sample Mendelian Randomization Analysis.

Non-alcoholic fatty liver disease (NAFLD) is the most common chronic liver disease globally, and diet plays a crucial role in its progression. While dietary fats impact NAFLD, the specific effect of cheese consumption remains unclear. This study employs a two-sample Mendelian randomization (MR) approach to explore the causal relationship between cheese intake and NAFLD, liver fat content, and liver fat proportion. Using summary-level data from large genome-wide association studies, we applied a two-sample MR approach. Genetic variants linked to cheese consumption served as instrumental variabl

The causal relationship between inflammatory bowel disease and primary biliary cholangitis: A bidirectional two-sample Mendelian randomization study.

The cause-and-effect relationship between inflammatory bowel disease (IBD) and primary biliary cholangitis (PBC) has not been fully elucidated. Thus, this study explored their causal relationship through bidirectional 2-sample Mendelian randomization (MR). Genome-wide association studies were used to obtain data. Single-nucleotide polymorphisms (SNPs) closely related to IBD (including ulcerative colitis [UC] and Crohn disease [CD]) and PBC were screened, and the methods used included inverse variance weighted and the weighted median method. MR analysis was performed using maximum likelihood an
