The hemolymph of Biomphalaria snail vectors of schistosomiasis supports a diverse microbiome.

The microbiome - the microorganism community that is found on or within an organism's body - is increasingly recognized to shape many aspects of its host biology and is a key determinant of health and disease. Microbiomes modulate the capacity of insect disease vectors (mosquitoes, tsetse flies, sandflies) to transmit parasites and disease. We investigate the diversity and abundance of microorganisms within the hemolymph (i.e. blood) of Biomphalaria snails, the intermediate host for Schistosoma mansoni, using Illumina MiSeq sequencing of the bacterial 16S V4 rDNA. We sampled hemolymph from fiv

Transmission of Corynosoma australe (Acanthocephala: Polymorphidae) from fishes to South American sea lions Otaria flavescens in Patagonia, Argentina.

Acanthocephalans display a two-host life cycle that involves arthropods as intermediate hosts and vertebrates as definitive hosts. Some species also use paratenic hosts to bridge the trophic gap between both obligatory hosts. However, the relative role of these paratenic hosts in the transmission to definitive hosts has seldom been assessed quantitatively. We report on infection patterns of cystacanths of Corynosoma australe Johnston, 1937 in 20 common teleost species and the Argentine shortfin squid Illex argentinus (Castellanos) from the Patagonian shelf of Argentina. We also explore the rol

Bacterial Pathogen Emergence Requires More than Direct Contact with a Novel Passerine Host.

While direct contact may sometimes be sufficient to allow a pathogen to jump into a new host species, in other cases, fortuitously adaptive mutations that arise in the original donor host are also necessary. Viruses have been the focus of most host shift studies, so less is known about the importance of ecological versus evolutionary processes to successful bacterial host shifts. Here we tested whether direct contact with the novel host was sufficient to enable the mid-1990s jump of the bacterium Mycoplasma gallisepticum from domestic poultry to house finches (Haemorhous mexicanus). We experim

Mitochondrial gene heterogeneity of the bat soft tick Argas vespertilionis (Ixodida: Argasidae) in the Palaearctic.

BACKGROUND: Recently, a high degree of mitochondrial gene heterogeneity was demonstrated between conspecific ixodid ticks of bats in Eurasia. Argas vespertilionis is a soft tick species of mainly vespertilionid bats, also with a wide distribution in the Old World. The aim of this study was to investigate the morphology, mitochondrial gene heterogeneity and host range of A. vespertilionis in the Old World. RESULTS: Altogether 318 soft tick larvae were collected from 17 bat species (belonging to six genera) in seven countries. Based on the general morphology (setal arrangement) of 314 A. vesper

Analysis of synonymous codon usage in Zika virus.

Zika virus is a zoonotic pathogen, which have made frequent incursion into the human population in Africa and South East Asia over the course of several decades but never reached to the pandemic proportions until the most recent outbreak. Viruses are solely dependent on host synthetic machinery for their replication cycle; therefore, replication and persistence in a host species of different genetic background requires certain degree of adaptation. These adaptations are necessary to avoid detection from host immune surveillance and maximize the utilization of available resources for efficient

A guard-killer phage cocktail effectively lyses the host and inhibits the development of phage-resistant strains of Escherichia coli.

In recent years, after the emergence of a large number of multidrug-resistant bacteria, phages and phage-associated products for the prevention and control of bacterial disease have revealed prominent advantages as compared with antibiotics. However, bacteria are susceptible to becoming phage-resistant, thus severely limiting the application of phage therapy. In this study, Escherichia coli cells were incubated with lytic bacteriophages to obtain mutants that were resistant to the lytic phages. Then, bacteriophages against the phage-resistant variants were isolated and subsequently mixed with

Estimating trematode prevalence in snail hosts using a single-step duplex PCR: how badly does cercarial shedding underestimate infection rates?

BACKGROUND: Trematode communities often consist of different species exploiting the same host population, with two or more trematodes sometimes co-occuring in the same host. A commonly used diagnostic method to detect larval trematode infections in snails has been based on cercarial shedding, though it is often criticized as inaccurate. In the present study we compare infection prevalences determined by cercarial emission with those determined, for the first time, by molecular methods, allowing us to quantify the underestimation of single and double infections based on cercarial emission. We t

An LED-based UV-B irradiation system for tiny organisms: System description and demonstration experiment to determine the hatchability of eggs from four Tetranychus spider mite species from Okinawa.

We developed a computer-based system for controlling the photoperiod and irradiance of UV-B and white light from a 5×5 light-emitting diode (LED) matrix (100×100mm). In this system, the LED matrix was installed in each of four irradiation boxes and controlled by pulse-width modulators so that each box can independently emit UV-B and white light at irradiances of up to 1.5 and 4.0Wm(-2), respectively, or a combination of both light types. We used this system to examine the hatchabilities of the eggs of four Tetranychus spider mite species (T. urticae, T. kanzawai, T. piercei and T. okinawanus)

Eilat virus host range restriction is present at multiple levels of the virus life cycle.

UNLABELLED: Most alphaviruses are mosquito-borne and exhibit a broad host range, infecting many different vertebrates, including birds, rodents, equids, humans, and nonhuman primates. This ability of most alphaviruses to infect arthropods and vertebrates is essential for their maintenance in nature. Recently, a new alphavirus, Eilat virus (EILV), was described, and in contrast to all other mosquito-borne viruses, it is unable to replicate in vertebrate cell lines. Investigations into the nature of its host range restriction showed the inability of genomic EILV RNA to replicate in vertebrate ce

Isolation and characterisation of lytic bacteriophages of Klebsiella pneumoniae and Klebsiella oxytoca.

Klebsiella bacteria have emerged as an increasingly important cause of community-acquired nosocomial infections. Extensive use of broad-spectrum antibiotics in hospitalised patients has led to both increased carriage of Klebsiella and the development of multidrug-resistant strains that frequently produce extended-spectrum β-lactamases and/or other defences against antibiotics. Many of these strains are highly virulent and exhibit a strong propensity to spread. In this study, six lytic Klebsiella bacteriophages were isolated from sewage-contaminated river water in Georgia and characterised as p

Immunological diagnosis of tuberculosis: problems and strategies for success.

Tuberculosis remains a serious social and public health problem, affecting millions of people annually. The bacille Calmette-Guerin (BCG) vaccine, used prophylactically, does not impede the progression of the disease, which usually manifests as decreased cellular immunity. Early diagnosis, together with polychemotherapy, can control the dissemination of the tuberculosis infection. The current diagnostic methods present certain problems. Such problems include the low sensitivity of sputum smear microscopy, the fact that performing microbiological cultures is quite time-consuming, and the low sp

Competitive displacement in Triatominae: the Triatoma infestans success.

Brazil has just been certificated by Pan American Health Organization as 'free of Chagas disease transmission due to Triatoma infestans'. During the early 1980s, this species of blood-sucking bug alone was considered responsible for approximately 80% of Chagas disease transmission. But it was not always so. The species originally abundant in houses of central and eastern Brazil was Panstrongylus megistus, which seems to have been progressively displaced from houses by T. infestans during the past century. Indeed, T. infestans seems able to displace other Triatominae in artificial environments.

The new fungal opportunists are coming.

Frequent and prolonged exposure of immunocompromised patients to a variety of environmental conditions has resulted in the recognition of infections with new fungal opportunists. Infections with fungi such as Fusarium species, Paecilomyces lilacinus, Acremonium species, Trichosporon beigelii, Blastoschizomyces capitatus, Malassezia furfur, Penicillium marneffei, Scedosporium prolificans, and other dematiaceous species have become significant problems in the treatment of immunocompromised hosts. This discussion addresses several issues of pathogenesis, epidemiology, diagnosis, and treatment wit

[Ticks (Ixodidae) in zoonotic propagation of piroplasma Babesia micorti and Babesia divergens].

Recent advance in the knowledge of the role of Ixodidae in the epidemiology of human piroplasmosis is reviewed, with special emphasis on Babesia species which cause diseases in man. The problem in Europe came to medical attention since 20 cases have been reported, and most of them due to B. divergens. However, contrary to other European countries, the zoonotic reservoir of Babesia spp. in Poland is unknown and further research is required to elucidate the importance of Ixodidae in the transmission of piroplasms.

Experimental infection of Anopheles gambiae s.s., Anopheles freeborni and Anopheles stephensi with Plasmodium malariae and Plasmodium brasilianum.

Susceptibility to infection of 2 strains of Anopheles gambiae s.s., An. freeborni and An. stephensi, was determined for 2 closely related malaria parasites, Plasmodium malariae and P. brasilianum. Neither strain of An. gambiae supported development of oocyst densities as great as the other 2 anopheline mosquitoes. The ZAN strain of An. gambiae s.s. from Zanzibar was more susceptible to infection with the strain of P. malariae from Uganda than the G-3 strain of An. gambiae s.s. from The Gambia. All species and strains of mosquitoes supported complete development to the presence of sporozoites i

Biology, host relations, and epidemiology of Sarcoptes scabiei.

Scabies continues to be an important parasitic disease of humans, and other mammals. Suprisingly for a disease that has afflicted humans since antiquity, little is directly known about the basic biology of the parasite, the host-parasite interactions, the host immune response, and host susceptibility. Much more research in these areas is needed if we are to understand fully the occurrence, transmission, and epidemiology of both human and animal scabies.

Identification of mosquito proteins that differentially interact with alphavirus nonstructural protein 3, a determinant of vector specificity.

Chikungunya virus (CHIKV) and the closely related onyong-nyong virus (ONNV) are arthritogenic arboviruses that have caused significant, often debilitating, disease in millions of people. However, despite their kinship, they are vectored by different mosquito subfamilies that diverged 180 million years ago (anopheline versus culicine subfamilies). Previous work indicated that the nonstructural protein 3 (nsP3) of these alphaviruses was partially responsible for this vector specificity. To better understand the cellular components controlling alphavirus vector specificity, a cell culture model s

Prokaryotic expression and immunogenicity of 56-kDa protein of Orientia tsutsugamushi strain Karp.

To express the 56-kDa protein of O. tsutsugamushi strain Karp, this protein gene was cloned into pET30a(+) before transforming into host bacteria, E. coli Rossetta. Specificity of the recombinant protein was assessed by ELISA using rabbit sera against common members of the order Rickettsiae and 10 other pathogenic bacteria. After IPTG induction, SDS-PAGE analysis of isolated protein demonstrated a band at approximately 46-kDa. Western blot and mass spectrometry analysis proved that the recombinant protein was expressed successfully. Specificity analysis demonstrated that all sera were negative
