Effect of sympathetic and angiotensin-aldosterone systems on renal salt conservation in the rat.

During dietary salt deprivation, the sympathetic nervous system and the angiotensin-aldosterone system are stimulated. Both systems are thought to be essential for maximal salt conservation by the kidney. To study their relative contributions, we produced negative salt balance in rats by intraperitoneal injection of furosemide, followed by a low-salt diet (<0.008% NaCl). In a 1-wk metabolic study, the animals were unable to replace the drug-induced salt deficit. Six groups of rats were studied. A control group established baseline function, a second group of 6-hydroxydopamine (OHDA) rats were

Detection of dopaminergic neurotransmitter activity using pharmacologic MRI: correlation with PET, microdialysis, and behavioral data.

The metabolic activation resulting from direct dopaminergic stimulation can be detected using auto-radiography, positron emission tomography (PET) or, potentially, fMRI techniques. To establish the validity of the latter possibility, we have performed a number of experiments. We measured the regional selectivity of two different dopaminergic ligands: the dopamine release compound D-amphetamine and the dopamine transporter antagonist 2 beta-carbomethoxy-3 beta-(4-fluoropheny) tropane (CFT). Both compounds led to increased signal intensity in gradient echo images in regions of the brain with hig

The effects of axotomy and preganglionic denervation on the expression of pituitary adenylate cyclase activating peptide (PACAP), galanin and PACAP type 1 receptors in the rat superior cervical ganglion.

The effects of axotomy, chemical sympathectomy and preganglionic denervation on the expression of the neuropeptides, pituitary adenylate cyclase-activating peptide (PACAP), galanin (GAL), and the PACAP type 1 receptor in the rat superior cervical ganglion (SCG) were investigated by immunocytochemistry, in situ hybridization and receptor autoradiography. An antibody recognizing the rat vesicular acetylcholine transporter (VAChT) was used for the detection of preganglionic cholinergic fibers. In the normal SCG, PACAP-immunoreactivity (-IR) was present in numerous, basket-forming, preganglionic n

Dopaminergic transplants suppress L-DOPA-induced Fos expression in the dopamine-depleted striatum in a rat model of Parkinson's disease.

We examined the effects of MK-801, a non-competitive antagonist of the N-methyl-D-aspartate (NMDA) receptor, and fetal ventral mesencephalic (VM) transplants on L-3,4-dihydroxyphenylalanine (L-DOPA)-induced Fos protein expression in the dopamine (DA)-depleted striatum. Unilateral 6-hydroxydopamine (6-OHDA) lesions of the nigrostriatal pathway were produced in young adult female rats and grafting was performed 3 weeks later. Methamphetamine-induced rotational behavior recovered significantly on the 4th week after grafting. Immunohistochemical examinations of c-Fos and tyrosine hydroxylase (TH)

Reactive astrocyte formation in vivo is regulated by noradrenergic axons.

Beta adrenergic receptor antagonists greatly reduce reactive astrocyte formation induced by neuronal degeneration. To test the hypothesis that the density of noradrenergic innervation is a factor in the regulation of astrocytosis, we measured glial fibrillary acidic protein (GFAP) optical density after neuronal injury in central nervous system (CNS) regions with permanent noradrenergic sprouting or norepinephrine (NE) depletion. The injury model employs the injection of Ricinus communis lectin into a cranial or peripheral nerve to destroy CNS neurons without the blood-brain barrier disruption

L-dopa induced increases in brain uric acid in an animal model of Parkinson's disease: a relationship to behavioral activation.

Rats with severe unilateral dopamine denervation (> or = 95% dopamine deficit), produced by intracerebral injection of 6-hydroxydopamine (6-OHDA) into the ventral tegmentum nigrostriatal dopamine neurons, were administered 25 mg/kg L-dopa (3,4-dihydroxyphenylalanine) methyl ester/2 mg/kg carbidopa. The neurochemical effects of the L-dopa treatment on uric acid in the cortex and striatum of the intact and 6-OHDA hemisphere were measured. In comparison to saline animals, uric acid concentrations in brain were increased in the L-dopa treated animals. There were no interhemispheric differences in

Selective regeneration of photoreceptors in goldfish retina.

Previous work has shown that the neural retina in adult goldfish can regenerate. Following retinal damage elicited by surgical or cytotoxic lesions, missing neurons are replaced by foci of proliferating neuroepithelial cells, which previous studies have suggested are derived from rod precursors. In the intact retina, rod precursors proliferate but produce only new rods. The regenerative responses observed previously have involved replacement of neurons in all retinal layers; selective regeneration of specific neuronal types (except for rod photoreceptors) has not been reported. In the experime

Alpha 2-adrenoceptors are not involved in the regulation of striatal glutamate release: comparison to dopaminergic inhibition.

Striatal slices from the rat were loaded with [3H]glutamate ([3H]Glu) and superfused in order to measure release of radioactivity at rest and during potassium-evoked depolarization. Addition of KCl (22-40 mmol/liter) to the perfusion fluid enhanced the release of tritium in a concentration-dependent manner, and this release was abolished by omission of CaCl2. High-performance liquid chromatography (HPLC) separation coupled with radiochemical detection revealed that 23% and 41% of the tritium efflux detected in the perfusion fluid under resting conditions and during potassium stimulation, respe

Ultrastructural localization of sympathetic axons in experimental rat sciatic nerve neuromas.

The ultrastructural localization of sympathetic axons was investigated in normal rat sciatic nerves and experimental sciatic nerve neuromas. The best ultrastructural localization of noradrenaline in the dense-cored vesicles of sympathetic axons was accomplished following pretreatment of rats with nialamide and 5-hydroxy dopamine, followed by fixation according to the modified chromaffin technique of Tranzer and Richards (1976). After such preparation, sympathetic axons containing 5-hydroxy dopamine-labelled dense-cored vesicles could be identified in normal sciatic nerve. Large accumulations o

Intrathecal bradykinin acts presynaptically on spinal noradrenergic terminals to produce antinociception in the rat.

In the awake restrained rat the intrathecal (i.th.) administration of 8.1 pmol-8.1 nmol of bradykinin (BK) and kallidin (KD) enhanced the reaction time (RT) to a noxious radiant heat stimulus in a dose-dependent manner. The fragments BK-(1-8) and BK-(1-7) were active only at doses higher than 10 nmol and the following rank order of potency was observed: BK greater than KD much greater than BK-(1-8) greater than BK-(1-7). The increment of tail-flick latency was greatest at 1 (BK) or 6 (KD) min and the RT returned to basal levels within 15 min post-administration. The effect of BK (81 pmol) was

Agonism of FFA4/GPR120 activates tyrosine hydroxylase and confers neuroprotection from 6-OHDA-induced cytotoxicity in PC12 cells and in a rat 6-OHDA model of Parkinson's disease.

Free-fatty acid receptor 4 (FFA4/GPR120), a G-protein-coupled receptor activated by medium to long-chain free-fatty acids (FFA) including ω-3 polyunsaturated fatty acids, regulates many metabolic and anti-inflammatory processes, and has also been linked to diverse neurophysiological functions. Here, we investigated the expression and functional role of FFA4 in dopaminergic systems using PC12 cells, rat striatal tissue, and an in vivo 6-hydroxydopamine (6-OHDA) model of Parkinson's disease (PD). FFA4 mRNA and protein were expressed in rat caudate-putamen and colocalized with tyrosine hydroxylas

Limosilactobacillus reuteri Alleviates Parkinson's Disease by Regulating CDCA Secretion.

To explore potential therapeutic agents for Parkinson's disease (PD), we investigated the impact of Limosilactobacillus reuteri (L. reuteri), a probiotic found to be significantly depleted in 6-hydroxydopamine (6-OHDA)-induced PD rat model. Despite its known benefits, the specific effects and underlying mechanisms of L. reuteri in PD remain poorly understood. In this study, we demonstrated that supplementation with L. reuteri alleviated motor deficits and attenuated dopamine (DA) neuron damage in 6-OHDA-induced PD rats. 16 S rRNA sequencing and untargeted metabolomics revealed that L. reuteri

Neuroprotection Against a Panel of Toxicants via a Novel Analog of the Natural Product Fraxinellone.

Humans are exposed to a myriad of environmental pollutants, with recent evidence indicating several of these toxicants serve as risk factors for neurodevelopmental disorders and neurodegenerative diseases. Given this, there is a need for both interventional and protective strategies; however, of concern, the mechanistic targets of these environmental pollutants are variable or unknown in some cases. A prior report indicated that analogs of the natural product fraxinellone act as potent NRF2 activators, mitigating excessive reactive oxygen species (ROS) generation and Glu toxicity in vitro. Usi

Modulation of Oxidative Stress and Apoptosis by Antrodia cinnamomea-Loaded Citrate-Stabilized Silver Nanoparticles in Experimental Parkinsonism.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by dopaminergic neuron loss in the substantia nigra pars compacta, accompanied by oxidative stress and neuroinflammation. Novel multitarget neuroprotective strategies are required to overcome the limitations of current symptomatic treatments. The neuroprotective effects of Antrodia cinnamomea (AC) and citrate-stabilized silver nanoparticles (AgNPs), alone and in combination, were evaluated using a 6-hydroxydopamine (6-OHDA)-induced SH-SY5Y cell model and a unilateral 6-OHDA rat model. Sixty-three rats were divid

Ghrelin-based interventions in preclinical models of Parkinson's disease: a systematic review.

Ghrelin plays a crucial role in metabolism and gastrointestinal function. In the central nervous system, ghrelin modulates both hedonic and homeostatic control of eating behavior. Ghrelin promotes neuron survival by reducing apoptosis, inflammation, and oxidative stress, making it a potential therapeutic agent for neurodegenerative diseases. Parkinson's Disease (PD) is a neurodegenerative disease characterized by motor and non-motor symptoms. The motor impairments result primarily from the progressive degeneration of dopaminergic neurons in the substantia nigra pars compacta. Individuals with

Mitochondria-targeted delivery of bromocriptine via cerium vanadate nanocarriers for Parkinson's disease.

Parkinson's disease (PD) is a therapeutically challenging neurodegenerative disease marked by profound disruptions in mitochondrial-redox axis. Current therapeutic agents such as Bromocriptine mesylate (BM) exhibit neuroprotective potential, however their clinical benefit remains largely symptomatic as they do not address the underlying cause of neurodegeneration. Moreover, the therapeutic efficacy of BM is compromised due to its poor solubility, low bioavailability, and inadequate brain penetration. To overcome these biological barriers and provide mechanistic interventions, we report the fir

Role of Mitochondrial Protein, mitoNEET, in Impaired Intracellular Cholesterol Metabolism-Induced Neuronal Cell Death.

Parkinson's disease (PD) is a progressive neurological disorder with an unclear etiology. Nonetheless, abnormal cholesterol metabolism is considered an environmental risk factor. MitoNEET (mNT) is an iron-sulfur cluster protein located in the outer mitochondrial membrane. mNT dysfunction is involved in the pathology of several diseases, including PD. However, the cause of mNT dysfunction remains unclear. Therefore, we hypothesized that increased intracellular cholesterol levels may reduce mNT function and increase reactive oxygen species (ROS) levels, resulting in neuronal cell death and PD pr

Characterization of the electrode-tissue interface during long-term deep brain stimulation in the 6-OHDA rat model of Parkinson's disease.

Objective.Deep brain stimulation is an established therapy for neurological disorders such as Parkinson's disease, but its underlying mechanisms and tissue effects remain incompletely understood. A particular challenge arises from the foreign body response to implanted electrodes, which leads to scar formation and encapsulation, altering the electrical properties of the surrounding tissue. This study aims to characterize the electrode-tissue interface during long-term stimulation and to improve model-based estimation of the stimulation volume using subject-specific dielectric properties.Approa

Elevated sympathetic tone exacerbates myocardial ischemia reperfusion injury through enhancing endothelial cell inflammation and apoptosis in chronic pain mice.

Although extensive evidence links chronic pain (CP) to an increased cardiovascular risk, its direct impact on myocardial ischemia-reperfusion (MI/R) remains poorly understood. In mice, CP was induced using chronic constriction injury (CCI) of the brachial plexus. We found that CP significantly increased myocardial infarction area, endothelial cell apoptosis, and inflammation, accompanied by the increase in sympathetic nerve tone around the endothelial cells. Viral tracer results demonstrated the existence of anatomical connections between the superior cervical ganglion (SCG) and the heart. The

Activation of the dopamine D2 receptor enhances colonic motility via disinhibition of somatostatinergic signalling in rodents.

Dopamine (DA) plays an important role in regulating gastrointestinal (GI) motility; however, the specific contribution of colonic dopamine D2 receptor (D2 receptor) remains incompletely understood. This study investigated the role of D2 receptor in colonic motility using pharmacological approaches in D2 receptor-deficient (D2R-/-) mice and a 6-hydroxydopamine (6-OHDA) rats model of Parkinson's disease (PD). In wild-type mice, low-dose DA and D2 receptor agonist quinpirole enhanced colonic spontaneous contractility. This pro-contractile effect was completely abolished by tetrodotoxin, indicatin
