Enfortumab Vedotin-Induced Cutaneous Toxic Effects and Survival in Urothelial Carcinoma.

IMPORTANCE: Enfortumab vedotin (EV) is an antibody-drug conjugate approved for the treatment of locally advanced or metastatic urothelial cancer. Cutaneous adverse events (cAEs) are common during EV therapy, with prior studies suggesting an association between EV-related cAEs and improved survival; however, there are insufficient data to delineate the survival benefit of EV-induced cAEs from those associated with concurrent immune checkpoint inhibitors (ICIs). OBJECTIVE: To evaluate the association of EV-induced cAEs with survival and to characterize the timing and morphologic mechanisms of E

[Use of sacituzumab govitecan in HR+/HER2- metastatic breast cancer following progression on endocrine therapy and chemotherapy: a case report in line with the results of the TROPiCS-02 study.].

Metastatic hormone receptor-positive HR+/HER2- breast cancer represents the most common biological subtype of advanced breast cancer. Despite the significant improvements achieved with the introduction of CDK4/6 inhibitors combined with endocrine therapy, disease progression eventually occurs in a substantial proportion of patients, requiring additional therapeutic strategies. In this context, antibody-drug conjugates have emerged as one of the most relevant therapeutic innovations in recent years. We report the case of a 60-year-old woman with metastatic HR+/HER2- breast cancer with liver, no

[NECTIN4 amplification as a predictive biomarker for enfortumab vedotin response].

Enfortumab vedotin (EV) is a targeted antibody-drug conjugate (ADC) directed against NECTIN4 and approved for treatment of metastatic urothelial carcinoma (mUC) after failure of platinum-based chemotherapy and immune checkpoint inhibitors. However, not all patients benefit equally from EV, highlighting the need for a predictive biomarker. In this multicenter study, 108 EV-treated mUC patients were analyzed for genomic NECTIN4 amplifications using fluorescence in situ hybridization (FISH). Approximately one-quarter of tumors showed NECTIN4 amplification, which was strongly associated with incre

OBI-992, a Novel TROP2-Targeted Antibody-Drug Conjugate, Demonstrates Antitumor Activity in Multiple Cancer Models.

Trophoblast cell surface antigen 2 (TROP2) is highly expressed in multiple cancers relative to normal tissues, supporting its role as a target for cancer therapy. OBI-992 is an antibody-drug conjugate (ADC) derived from a novel TROP2-targeted antibody linked to the topoisomerase 1 (TOP1) inhibitor exatecan via an enzyme-cleavable hydrophilic linker, with a drug-antibody ratio of 4. This study evaluated and compared the antitumor activity of OBI-992 with that of benchmark TROP2-targeted ADCs datopotamab deruxtecan (Dato-DXd) and sacituzumab govitecan (SG) in cell line-derived xenograft (CDX) an

Future directions for antibody-drug conjugates in urothelial cancer.

INTRODUCTION: Urothelial carcinoma (UC) is a highly aggressive malignancy that has traditionally been treated with platinum-based chemotherapy. In recent years, the therapeutic landscape has undergone substantial transformation with the advent of precision oncology, particularly through the introduction of immune checkpoint inhibitors, targeted agents, and antibody-drug conjugates (ADCs). AREAS COVERED: We searched MEDLINE (via PubMed), Embase, and ClinicalTrials.gov and reviewed practice guidelines from ASCO, ESMO, AIOM, and EAU in order to describe the current landscape of ADC-based therapy

A humanized anaplastic lymphoma kinase (ALK)-directed antibody-drug conjugate with pyrrolobenzodiazepine payload demonstrates efficacy in ALK-expressing cancers.

The Anaplastic Lymphoma Kinase (ALK) gene is a receptor tyrosine kinase (RTK) with expression restricted to the developing nervous system. Most neuroblastomas express native ALK protein on the cell surface and ALK is uniformly overexpressed in fusion-positive rhabdomyosarcoma and in subsets of metastatic colorectal carcinoma, melanoma, ovarian carcinoma, and breast carcinoma. Here, we first confirm that ALK RNA, protein, and tumor cell surface expression is elevated in multiple pediatric and adult malignancies with minimal expression in childhood normal tissues. We then demonstrate that a huma

Site-specific Antibody-Nitric Oxide Conjugate HN02 Possesses Improved Antineoplastic and Safety Properties.

Antibody-drug conjugates (ADCs) combine the high specificity of antibodies with the cytotoxicity of payloads and have great potential in pan-cancer immunotherapy. However, the current payloads for clinical uses have limited the therapeutic window due to their uncontrollable off-site toxicity. There is unmet needs to develop more potent ADC payloads with better safety and efficacy profiles. Nitric oxide (NO) is a special molecule that has low toxicity itself, which can kill tumor cells effectively when highly concentrated, has broad application prospects. Previously, we prepared for the first t

Case report: The utilization of crizotinib and brentuximab vedotin as a bridge to autologous stem cell transplantation and followed by CD30-directed CAR-T cell therapy in relapsed/refractory ALK+ ALCL.

BACKGROUND: Anaplastic lymphoma kinase-positive anaplastic large cell lymphoma (ALK+ ALCL) is a rare, mature T-cell non-Hodgkin lymphoma. The prognosis of patients with relapsed or refractory ALCL following first-line chemotherapy is extremely poor. NCCN guidelines recommend intensified chemotherapy with or without ASCT consolidation for r/r ALCL, however, this is not an effective treatment for all ALK+ALCL. CASE REPORT: Herein, we report a patient with relapsed/refractory ALK+ ALCL who received crizotinib and brentuximab vedotin as bridging therapy, followed by autologous stem cell transplan

New Advances in Metastatic Urothelial Cancer: A Narrative Review on Recent Developments and Future Perspectives.

The standard of care for advanced or metastatic urothelial carcinoma (mUC) was historically identified with platinum-based chemotherapy. Thanks to the advances in biological and genetic knowledge and technologies, new therapeutic agents have emerged in this setting recently: the immune checkpoint inhibitors and the fibroblast growth factor receptor inhibitors as the target therapy for patients harboring alterations in the fibroblast growth factor receptor (FGFR) pathway. However, chasing a tumor's tendency to recur and progress, a new class of agents has more recently entered the scene, with p

Anti-tumor effect of antibody-drug conjugate targeting cell adhesion molecule 1 on GIST cells representing small intestinal GIST.

Gastrointestinal stromal tumor (GIST) is the most common mesenchymal tumor of the alimentary tract. The prognosis depends on the primary site, and small intestinal GISTs have a worse prognosis than gastric GISTs. Molecularly targeted drugs to inhibit tyrosine kinase activity of KIT were used for unresectable or recurrent GISTs. However, secondary resistance to the drugs is often acquired, and treatments based on other mechanisms are needed. Previously, we reported that cell adhesion molecule 1 (CADM1) was highly expressed in most of small intestinal GISTs but not in most of gastric GISTs. In t
