Intrinsic neurons of Drosophila mushroom bodies express short neuropeptide F: relations to extrinsic neurons expressing different neurotransmitters.

Mushroom bodies constitute prominent paired neuropils in the brain of insects, known to be involved in higher olfactory processing and learning and memory. In Drosophila there are about 2,500 intrinsic mushroom body neurons, Kenyon cells, and a large number of different extrinsic neurons connecting the calyx, peduncle, and lobes to other portions of the brain. The neurotransmitter of the Kenyon cells has not been identified in any insect. Here we show expression of the gene snpf and its neuropeptide products (short neuropeptide F; sNPFs) in larval and adult Drosophila Kenyon cells by means of

OTU7B Modulates the Mosquito Immune Response to Beauveria bassiana Infection via Deubiquitination of the Toll Adaptor TRAF4.

The Aedes aegypti mosquito transmits devastating flaviviruses, such as Zika, dengue, and yellow fever viruses. For more effective control of the vector, the pathogenicity of Beauveria bassiana, a fungus commonly used for biological control of pest insects, may be enhanced based on in-depth knowledge of molecular interactions between the pathogen and its host. Here, we identified a mechanism employed by B. bassiana, which efficiently blocks the Ae. aegypti antifungal immune response by a protease that contains an ovarian tumor (OTU) domain. RNA-sequencing analysis showed that the depletion of O

Nutritional properties of extracellular vesicle-like particles from Sophora flavescens and Periplaneta americana with effects on streptozotocin-induced diabetic wound healing in rats.

INTRODUCTION: The medicinal plant Sophora flavescens (SF) and the insect Periplaneta americana (PA) have long been used in treating diabetic wounds. PA-derived extracellular vesicle-like particles (PA-EVLP) can accelerate diabetic wound healing. However, no evidence of the nutritional components and anti-diabetic wound effects of the combination of SF-EVLP with PA-EVLP (SF-PA-EVLPs) is available. METHODS: We separated SF-EVLP and PA-EVLP via ultracentrifugation and characterized them via nano flow cytometry, transmission electron microscopy, and atomic force microscopy. Ultra-high-performance

Edible Insects: Perceptions of Marketing, Economic, and Social Aspects among Citizens of Different Countries.

Because edible insects (EI) have been, in recent years, recommended as a nutritious animal protein food with enormous environmental advantages over other sources of animal protein for human consumption, studies aimed at investigating the consumer perspective have become more prominent. Hence, this study intended to examine the perceptions of participants from different countries about the commercialization and economic and social impacts of edible insects. The study was made using a questionnaire survey, and data were collected in Brazil, Croatia, Greece, Latvia, Lebanon, Lithuania, Mexico, Po

Lnc40766-TLR6-miR14-3p Regulation Pathway Mediates the Immunity of Plutella xylostella against Entomopathogenic Fungus Isaria cicadae.

The diamondback moth Plutella xylostella is a major agricultural pest with severe insecticide resistance. Entomopathogenic fungi (EPF) serve as eco-friendly biocontrol. Insects rely on immunity to resist fungal invasion, while the regulatory mechanism of long non-coding RNAs (lncRNAs) in insect antifungal immunity remains unclear. Here, lncRNA-mediated immune responses of P. xylostella against EPF Isaria cicadae were explored. RNA-seq identified 33,402 lncRNAs, 97 of which were predicted to cis-regulate 72 immune genes. The lnc40766 targets Toll-like receptor 6 (TLR6); both were upregulated po

Development of an RPA-CRISPR/Cas12a-based rapid visual detection method for two insecticide resistance-associated mutations in Aphis gossypii: A302S in the ace1 gene and M918L in the para gene.

The resistance of Aphis gossypii to pyrethroids and organophosphate/carbamate insecticides has become increasingly severe. The para M918L and ace1 A302S target-site mutations are the key mechanisms conferring resistance to these two types of insecticides, respectively. However, rapid and field-deployable methods for detecting these mutations are still lacking. In this study, the genotype and allele frequencies of eight field populations collected in 2023 were investigated, and a rapid visual detection method based on RPA-CRISPR/Cas12a was developed. By optimizing the concentration of the react

Functional characterization of PxUGT33W1 with abamectin response and reproduction in Plutella xylostella.

UDP-glycosyltransferases (UGTs) are key Phase II detoxification enzymes involved in insect xenobiotic metabolism; however, the functional contribution of individual UGT genes to abamectin response in Plutella xylostella remains poorly understood. In this study, we characterized a candidate gene, PxUGT33W1, using a combination of molecular, cellular, and genetic approaches. PxUGT33W1 exhibited broad expression across developmental stages and tissues, with elevated transcript levels in the midgut, epidermis, and reproductive organs. Its expression was significantly induced following abamectin ex

Comprehensive proteomic analysis of honeybee (Apis mellifera L.) intestine reveals nutrient-dependent molecular adaptation.

Western honeybees (Apis mellifera, L.) play a crucial role in the pollination of numerous plants, including many economically important crops, and are essential for food production. In this study, we first developed and optimized a protocol for the extraction and characterization of proteins from the intestinal tissue of worker honeybees, which allowed the identification of 1787 distinct components. By feeding honeybees with distinct diets in a caged bee assay and using a Tandem Mass Tag (TMT)-based proteomic approach, we also investigated how the proteome of A. mellifera intestine varies its

Sequence and expression analysis of potential spermatogenesis-specific gene cognates in the Caribbean fruit fly, Anastrepha suspensa.

The sterile insect technique (SIT) is a highly effective biologically-based method for the suppression of many insect pest populations. SIT efficacy could be improved by methods of male sterilization that avoid the use of irradiation that can result in diminished fitness and mating competitiveness. Alternative sterilization methods include conditional disruption of genes for male fertility, or using their sperm-specific promoters to drive the expression of genes for lethal effectors. Testing has begun for the testis-specific β2-tubulin gene, though additional male fertility genes are required

Hypoxanthine accumulation caused by mutations of Bombyx mori xanthine dehydrogenase triggers female sterility.

Insect reproductive capacity is a key determinant of population fitness, though its regulatory mechanisms remain limited. Here, we identified an op50 silkworm mutant (derived from p50 strain) exhibiting female-specific reproductive defects, which provided an important model for studying insect reproductive regulation mechanism. We found that Bombyx mori xanthine dehydrogenase 1 (BmXDH1) in op50 contained a single-base insertion causing premature termination, deleting 154 C-terminal amino acids and losing catalytic activity. ΔBmXDH1 mutants recapitulated the op50 phenotype with 50 % reduced ovi

Resistance reduction and cross-resistance of spinosad-resistant Rhyzopertha dominica (Coleoptera: Bostrichidae) and the association between spinosad resistance and maltase activity.

BACKGROUND: The lesser grain borer, Rhyzopertha dominica, is a serious stored-products pest mainly controlled by insecticides. Spinosad, an environmentally friendly biological insecticide with low mammalian toxicity, is considered a suitable candidate for R. dominica management. Given the structural similarity of spinosad and maltose, both containing a neutral sugar structure, it was hypothesized that maltase may hydrolyze spinosad, rendering R. dominica resistant to spinosad. In this study, multiple spinosad-resistant strains (RdSR) were used to test the hypothesis. RESULTS: The resistance r

Bicaudal-C plays a critical role in regulating oogenesis in the lepidopteran insect Bombyx mori.

Oogenesis is an intricate cellular specialization process that requires the precise expression of multiple genes. However, the functional genes and mechanisms contributing to lepidopteran oogenesis remain incompletely understood. In silkworm, the eggless mutant (smn) has a typical oogenesis disorder phenotype. In smn females, oocyte development is delayed, causing follicle degeneration and the absence of mature eggs in virgin ovarioles. Here, we revealed that BmBic-C, an ortholog of Drosophila melanogaster Bicaudal-C (Bic-C), is responsible for smn. BmBic-C expression was significantly downreg

Central role of squid gene during oocyte development in the Hemiptera Rhodnius prolixus.

Oocyte polarity establishment is a conserved and crucial phenomenon for embryonic development. It relies on the precise spatial localization of maternal factors deposited during oocyte development, which is essential for establishing and maintaining cell polarity and subsequently specifying embryonic axes. The heterogeneous nuclear ribonucleoprotein (hnRNP) encoded by the squid (sqd) gene has been implicated in mRNA localization and embryonic axis establishment in Drosophila melanogaster. Comparative genomics allowed for the identification of a homologue in Rhodnius prolixus. In this study, we

Osiris17 is indispensable for morphogenesis of intestinal tract in Locusta migratoria.

The Osiris gene family is believed to play important roles in insect biology. Previous studies mainly focused on the roles of Osiris in Drorophila, how Osiris operates during the development of other species remains largely unknown. Here, we investigated the role of LmOsi17 in development of the hemimetabolous insect Locusta migratoria. LmOsi17 was highly expressed in the intestinal tract of nymphs. Knockdown of LmOsi17 by RNA interference (RNAi) in nymphs resulted in growth defects. The dsLmOsi17-injected nymphs did not increase in body weight or size and eventually died. Immunohistochemical

CRISPR/Cas9-based functional characterization of the pigmentation gene ebony in Plutella xylostella.

Body pigmentation is an important character of insects in adapting to biotic and abiotic environmental challenges. Additionally, based on the relative ease of screening, several genes involved in insect melanization have been used in classic genetic studies or as visual markers in constructing transgenic insects. Here, a homologue of the Bombyx mori melanization-inhibiting gene ebony, associated with the conversion of dopamine to N-β-alanyl dopamine, was identified in a global pest, Plutella xylostella. The CRISPR/Cas9 system was applied to generate multiple Pxebony knockout alleles which were

Effects of sublethal phoxim exposure and lower food intake on nutrient metabolism in the midguts of Bombyx mori.

Silkworm (Bombyx mori) is an economically important insect. However, the survival of silkworms has been significantly affected by the assault of chemical pesticides on mulberry trees through aerial application and water currents. Phoxim is a broad-spectrum organophosphorus insecticide widely used in China. Currently, very little is known about the non-neuronal effects of sublethal exposure to phoxim. The purpose of this study was to investigate the non-neuronal effects of sublethal phoxim exposure in the silkworm midgut, with a focus on nutrient metabolism. After phoxim treatment, lipase activ

Molecular composition of the paralyzing venom of three solitary wasps (Hymenoptera: Pompilidae) collected in southeast Mexico.

The chemical and biological characterization of peptide and protein components of the paralyzing venom from three Pompilidae solitary spider wasps (Pepsis mexicana, Pepsis terminata, and Anoplius nigritus) is described for the first time. The molecular masses of the most abundant peptides were determined. The N-terminal sequences of two cysteine-rich peptides were obtained from Pepsis. Metalloproteinase and hyaluronidase activities were identified in the venom of P. mexicana. A novel non-lethal method to collect venom is described.

Exploring the hidden honeybee (Apis mellifera) venom proteome by integrating a combinatorial peptide ligand library approach with FTMS.

UNLABELLED: At present, 30 compounds have been described in the venom of the honeybee, and 16 of them were confirmed by mass spectrometry. Previous studies typically combined 2-D PAGE with MALDI-TOF/TOF MS, a technology which now appears to lack sensitivity to detect additional venom compounds. Here, we report an in-depth study of the honeybee venom proteome using a combinatorial peptide ligand library sample pretreatment to enrich for minor components followed by shotgun LC-FT-ICR MS analysis. This strategy revealed an unexpectedly rich venom composition: in total 102 proteins and peptides we

On the venom system of centipedes (Chilopoda), a neglected group of venomous animals.

Centipedes are among the oldest extant terrestrial arthropods and are an ecologically important group of soil and leaf litter predators. Despite their abundance and frequent, often painful, encounters with humans, little is known about the venom and venom apparatus of centipedes, although it is apparent that these are both quite different from other venomous lineages. The venom gland can be regarded as an invaginated cuticle and epidermis, consisting of numerous epithelial secretory units each with its own unique valve-like excretory system. The venom contains several different enzymes, but is

Relation between the outer cover of the egg case of Argiope aurantia (Araneae: Araneidae) and the emergence of its spiderlings.

To emerge from the egg case, Argiope aurantia spiderlings must penetrate a tightly woven outer cover composed primarily of large-diameter cylindrical gland fibers and small-diameter fibers, likely of aciniform gland origin. They accomplish this using enzymatic digestion and mastication to form a communal hole in the outer cover. The involvement of proteolytic enzymes in this process was demonstrated by zymography of spiderling homogenates and washes made from the edges of holes. The specific source(s) of the proteases is unknown, but histological examination of spiderling sections indicates th
