Voltage-sensor conformational microenvironments encode isoform-selective ion channel activation.

Isoform-selective opening of voltage-gated potassium (Kv) channels is desirable for its therapeutic potential yet mechanistically little-understood. Rosemary leaf metabolite carnosic acid (CA) is a high-efficacy opener of the neuronal Kv7.3 channel but exerts minimal effects on Kv7.2 or Kv7.2/3 heteromers. Here, using alanine-scanning mutagenesis, electrophysiology, radioligand binding, and all-atom molecular dynamics simulations, we found that CA achieves high-efficacy activation of Kv7.3 by binding at the extracellular cap of the voltage-sensing domain (VSD), independent of the canonical ret

Genetic engineering of somatic cells to study and improve cardiac function.

AIMS: To demonstrate the utility of genetically engineered excitable cells for studies of basic electrophysiology and cardiac cell therapy. METHODS AND RESULTS: 'Zig-zag' networks of neonatal rat ventricular myocytes (NRVMs) were micropatterned onto thin elastomeric films to mimic the slow action potential (AP) conduction found in fibrotic myocardium. Addition of genetically engineered excitable human embryonic kidney cells (HEK-293 cells) ('Ex-293' cells stably expressing Kir2.1, Na(v)1.5, and Cx43 channels) increased both cardiac conduction velocity by 370% and twitch force amplitude by 64%

Phasic activity of urinary bladder smooth muscle in the streptozotocin-induced diabetic rat: effect of potassium channel modulators.

Increased phasic activity in the bladder smooth muscle of animal models and patients with detrusor overactivity has been suggested to underlie the pathophysiology of overactive bladder. Potassium (K+) channels are key regulators of bladder smooth muscle tone and thus may play a role in this altered phasic activity. In this study the effects of K+ channel modulators on the phasic activity of bladder strips from the streptozotocin-induced diabetic rat model of bladder dysfunction were investigated. Bladder strips from rats 1 week following streptozotocin administration and age-matched controls w

Isolation and characterization of a high affinity peptide inhibitor of ClC-2 chloride channels.

The ClC protein family includes voltage-gated chloride channels and chloride/proton exchangers. In eukaryotes, ClC proteins regulate membrane potential of excitable cells, contribute to epithelial transport, and aid in lysosomal acidification. Although structure/function studies of ClC proteins have been aided greatly by the available crystal structures of a bacterial ClC chloride/proton exchanger, the availability of useful pharmacological tools, such as peptide toxin inhibitors, has lagged far behind that of their cation channel counterparts. Here we report the isolation, from Leiurus quinqu

Physiological significance of hyperpolarization-activated inward currents (Ih) in smooth muscle cells from the circular layers of pregnant rat myometrium.

The properties of hyperpolarization-activated current in pregnant rat uterus (17-19 days gestation) were investigated using microelectrode and patch-clamp techniques, and isometric tension recording. The resting membrane potentials were -58.4 mV and -48.5 mV in longitudinal and circular muscle cells, respectively. Application of hyperpolarizing current pulses produced a time-dependent anomalous inward rectification of membrane potential only in circular muscle cells. Under voltage-clamp conditions, inward currents (Ih) were activated by long hyperpolarizing pulses below -60 mV in circular but

Identification of residues lining the anthrax protective antigen channel.

In its activated 63 kDa form, the protective antigen (PA) component of anthrax toxin forms a heptameric prepore, which converts to a pore (channel) in endosomal membranes at low pH and mediates translocation of the toxin's enzymic moieties to the cytosol. It has been proposed that the prepore-to-pore conversion involves a conformational rearrangement of a disordered amphipathic loop (D2L2; residues 302-325), in which loops from the 7 protomers combine to form a transmembrane 14-stranded beta barrel. To test this model, we generated Cys substitutions in 24 consecutive residues of the D2L2 loop,

P2X4 Drives Sex-Specific neuroprotection in autoimmune neuroinflammation.

Microglia critically influence multiple sclerosis (MS) pathophysiology through debris clearance, myelin repair, and modulation of neuroinflammation. These processes are partly regulated by ATP-gated ion channel P2X4, predominantly expressed in microglia. We previously reported that ivermectin (IVM), a positive allosteric modulator of P2X4, modulates microglia activation and function in myelin phagocytosis and promotes following lysolecithin-induced demyelination, and ameliorates neurological symptoms in experimental autoimmune encephalomyelitis (EAE). Here, we dissected the molecular and cellu

SmOBr Quasi-van der Waals Dielectric with High-κ and Ultralow Hysteresis for Two-Dimensional Electronics.

The integration of two-dimensional (2D) semiconductors into next-generation electronics critically depends on the availability of high-κ dielectrics that combine wide band gaps, strong breakdown fields, and pristine interfaces. Conventional oxides such as HfO2 suffer from interfacial incompatibility with 2D channels, while van der Waals dielectrics like h-BN offer excellent stability but limited capacitance. Here, we report the synthesis of quasi-vdW samarium oxybromide (SmOBr) nanosheets via a molten salt-assisted chemical vapor deposition method, establishing a previously unexplored member o

Scalable fabrication and mechanistic understanding of horizontally aligned MWCNT nanochannel devices for ionic current-based DNA biosensing.

Carbon nanotube (CNT)-based ionic nanochannels offer a powerful platform for controlling ion transport at the nanoscale, enabling highly sensitive biosensing modalities based on molecularly gated ion transport. In this work, we design, fabricate, simulate, and experimentally characterize a multi-walled carbon nanotube (MWCNT)-based ionic nanochannel functionalized with single-stranded DNA for label-free detection of complementary nucleic acids. Devices fabricated via CVD growth, mechanical transfer, and microfabrication of integrated reservoirs and electrodes exhibit stable and reproducible io

Structural insights into the human P2X1 receptor and ligand interactions.

The P2X1 receptor is a trimeric ligand-gated ion channel that plays an important role in urogenital and immune functions, offering the potential for new drug treatments. However, progress in this area has been hindered by limited structural information and a lack of well-characterised tool compounds. In this study, we employ cryogenic electron microscopy (cryo-EM) to elucidate the structures of the P2X1 receptor in an ATP-bound desensitised state and an NF449-bound closed state. NF449, a potent P2X1 receptor antagonist, engages the receptor distinctively, while ATP, the endogenous ligand, bind

The HERG K+ channel: progress in understanding the molecular basis of its unusual gating kinetics.

The HERG K+ channel has very unusual kinetic behaviour that includes slow activation but rapid inactivation. These features are critical for normal cardiac repolarisation as well as in preventing lethal ventricular arrhythmias. Extensive mutagenesis of the HERG K+ channel has allowed identification of which regions of the channel are important for the unusual kinetic behaviour of the channel. Furthermore, structural studies on scorpion toxins that potently inhibit HERG are beginning to provide clues as to the structural differences between HERG and other voltage-gated K+ channels.

Intracellular Ca2+ waves in mammalian cells.

Intracellular calcium waves refer to the coordinated propagation of increased free calcium ion (Ca2+) concentration in the cytoplasm. Ca2+ is one of the major intracellular second messengers which coordinates many cells function including gene transcription, division, and cell apoptosis. The spread of the ions in the cytoplasm is not the same in all cell types. Experiments indicate the strength of the stimuli, the site of the first Ca2+ entry and the localization of the organelles influence the Ca2+ propagation and may lead to functional compartmentalization. Polarized cells with complex anato

Cellular and behavioral effects of altered NaV1.2 sodium channel ion permeability in Scn2aK1422E mice.

Genetic variants in SCN2A, encoding the NaV1.2 voltage-gated sodium channel, are associated with a range of neurodevelopmental disorders with overlapping phenotypes. Some variants fit into a framework wherein gain-of-function missense variants that increase neuronal excitability lead to developmental and epileptic encephalopathy, while loss-of-function variants that reduce neuronal excitability lead to intellectual disability and/or autism spectrum disorder (ASD) with or without co-morbid seizures. One unique case less easily classified using this framework is the de novo missense variant SCN2

Functional characterization of acid-sensing ion channels in cultured neurons of rat inferior colliculus.

Acid-sensing ion channels (ASICs), which are widely distributed in the mammalian brain, the spinal cord and the peripheral sensory organs, are ligand-gated cation channels activated by extracellular protons. Abundant experimental evidence shows that ASICs play important roles in physiological/pathological conditions, such as sensory transduction, learning/memory, retinal function, seizure and ischemia. In the auditory system, however, there are only a few studies available describing ASICs in hair cells, the spiral ganglion and the vestibular ganglion. In particular, functional ASICs have not

Two HCN4 Channels Play Functional Roles in the Zebrafish Heart.

The HCN4 channel is essential for heart rate regulation in vertebrates by generating pacemaker potentials in the sinoatrial node. HCN4 channel abnormality may cause bradycardia and sick sinus syndrome, making it an important target for clinical research and drug discovery. The zebrafish is a popular animal model for cardiovascular research. They are potentially suitable for studying inherited heart diseases, including cardiac arrhythmia. However, it has not been determined how similar the ion channels that underlie cardiac automaticity are in zebrafish and humans. In the case of HCN4, humans h

Actions of a hydrogen sulfide donor (NaHS) on transient sodium, persistent sodium, and voltage-gated calcium currents in neurons of the subfornical organ.

Hydrogen sulfide (H2S) is an endogenously found gasotransmitter that has been implicated in a variety of beneficial physiological functions. This study was performed to investigate the cellular mechanisms underlying actions of H2S previously observed in subfornical organ (SFO), where H2S acts to regulate blood pressure through a depolarization of the membrane and an overall increase in the excitability of SFO neurons. We used whole cell patch-clamp electrophysiology in the voltage-clamp configuration to analyze the effect of 1 mM NaHS, an H2S donor, on voltage-gated potassium, sodium, and calc

Erg currents support electrical bursting in murine anterior pituitary corticotrophs.

The regulation of electrical excitability of anterior pituitary corticotrophs is critical for an appropriate response of the hypothalamic-pituitary-adrenal (HPA) axis in the face of diverse physiological challenges in health and disease. However ion channels that control corticotroph excitability remain poorly characterised. Members of the mammalian ether-à-go-go (EAG) channel family are voltage-gated potassium channels with diverse functions in the endocrine, cardiovascular and nervous systems. Expression of Kcnh2 mRNA, which encodes for the Eag-related potassium channel Erg1, is enriched in

Modulation of human Nav1.7 channel gating by synthetic α-scorpion toxin OD1 and its analogs.

Nine different voltage-gated sodium channel isoforms are responsible for inducing and propagating action potentials in the mammalian nervous system. The Nav1.7 channel isoform plays an important role in conducting nociceptive signals. Specific mutations of this isoform may impair gating behavior of the channel resulting in several pain syndromes. In addition to channel mutations, similar or opposite changes in gating may be produced by spider and scorpion toxins binding to different parts of the voltage-gated sodium channel. In the present study, we analyzed the effects of the α-scorpion toxin

Lipopolysaccharide-induced pulpitis up-regulates TRPV1 in trigeminal ganglia.

Tooth pain often accompanies pulpitis. Accumulation of lipopolysaccharides (LPS), a product of Gram-negative bacteria, is associated with painful clinical symptoms. However, the mechanisms underlying LPS-induced tooth pain are not clearly understood. TRPV1 is a capsaicin- and heat-gated nociceptive ion channel implicated in thermosensation and hyperalgesia under inflammation or injury. Although TRPV1 is expressed in pulpal afferents, it is not known whether the application of LPS to teeth modulates TRPV1 in trigeminal nociceptors. By assessing the levels of protein and transcript of TRPV1 in m

The gap junction: a channel for multiple functions?

Gap junctions are specialized membrane structures that enable the intercytoplasmic exchange of small molecules and ions between contacting cells. During the past decade, biophysical and structural analyses of the junctional channel have considerably increased our understanding of the pharmacological properties and gating mechanisms of gap junctions. Despite this impressive amount of work, until recently the physiological role of these ubiquitous intercellular pathways has remained speculative in most tissues. This review summarizes the most recent information obtained on the structure of the g
