Growth Differentiation Factor 15 Is a Novel Diagnostic Biomarker of Mitochondrial Diseases.

The present study aimed to investigate whether serum growth differentiation factor 15 concentration is a valuable and reliable diagnostic biomarker of mitochondrial diseases. We examined consecutive patients with mitochondrial diseases, in comparison with patients with non-mitochondrial disease neuromuscular disorders and healthy controls. The serum concentrations of growth differentiation factor 15 were measured by ELISA, and compared with those of FGF21, lactate, and creatine kinase. We also evaluated the correlations between growth differentiation factor 15 concentrations and the Newcastle

Spectrum of clinical neuroimaging in mitochondrial disorders: a neuroanatomical approach.

Mitochondrial disorders are a highly heterogeneous group of genetic diseases that impact pathways associated with the structure and function of the mitochondrion. Clinical presentations of mitochondrial disorders include a wide range of onset, progression, and spectrum of neurological symptoms - ranging from episodic, focal neurological deficits to gradual onset of developmental delays, sensorineural hearing loss, visual impairment, or ataxia. This variability provides clinicians with a diagnostic challenge in identifying suspicion of a mitochondrial disorder and prioritizing specific mitochon

[Spiroergometry in diagnosis of mitochondrial diseases].

Cardiopulmonary exercise testing (CPX) is a non-invasive method of recording quantitative data from gas exchange and ventilation for the evaluation of oxidative metabolism at rest and during exercise. Determination of oxygen uptake (VO2) and carbon dioxide output (VCO2) describes the activity of anaerobic vs aerobic metabolism. An incremental exercise test measuring gas exchange, ventilation and lactate release was performed in healthy volunteers and in patients suffering from mitochondrial disorders. At rest as well as during exercise patients with mitochondrial disorders differ from healthy

Leigh Syndrome due to MT-ATP6 Variants: A Case Presentation and the Review of the Literature.

INTRODUCTION: Leigh syndrome is a rare mitochondrial disorder characterized by subacute necrotizing encephalomyelopathy, resulting from defects in mitochondrial respiratory enzymes or pyruvate dehydrogenase complex. Symptoms can manifest in infancy, childhood, or adulthood. We present a case of a 7-month-old girl initially misdiagnosed with septic shock but was later found to have Leigh encephalomyelopathy due to MT-ATP6 deficiency. CASE PRESENTATION: A 7-month-old girl was admitted with fever, drowsiness, and wheezing, initially diagnosed with septic shock. She had a history of parental cons

Breastfeeding and the sexual abuse survivor.

In this paper, the possible effects of child sexual abuse on a mother's breastfeeding experience are described. The long-term effects of sexual abuse are divided into seven domains that may impact breastfeeding behavior: posttraumatic stress disorder, cognitive distortions, emotional distress, impaired sense of self, avoidance, interpersonal difficulties, and health problems. In each section, the potential impact of past sexual abuse on current breastfeeding behavior and the mother-infant relationship is described. Finally, specific suggestions and strategies for lactation consultants are offe

Limited nesting stress in mice as a model to study neuroimmune relationships in postpartum depression.

Postpartum depression (PPD) is a mood disorder that affects the ability of mothers to engage with and care for their infants. Stress exposure is known to be a major predisposing factor for PPD, while immune factors such as T regulatory cells (Tregs) are also hypothesized to influence the development of the disorder. Here we assessed limited nesting (LN) in mice, with or without depletion of Tregs through anti-CD25 antibody treatment, as a potential model to investigate the relationship between stress and the immune system in PPD. Dams and their pups were exposed to LN from post-natal day 3-10

Glycolytic Reprogramming in Parkinson's Disease: From Bioenergetic Crosstalk to Therapeutic Modulation.

Parkinson's disease (PD) is increasingly recognized not merely as a localized proteinopathy, but as a systemic metabolic disorder driven by bioenergetic failure. While mitochondrial dysfunction is a well-established pathological hallmark, the compensatory reprogramming of glycolysis has emerged as a critical, yet double-edged, determinant of neuronal survival. This review critically examines the molecular and spatial architecture of glucose metabolism in the parkinsonian brain. We systematically dissect how upstream genetic regulators, including PTEN and PARK7/DJ-1, alongside downstream rate-l

1H NMR-Based Metabolomic Signatures in Rodent Models of Sporadic Alzheimer's Disease and Metabolic Disorders.

Alzheimer's disease (AD) is a chronic neurological disorder that impacts the elderly population all over the globe. Evidence suggests association between AD and metabolic disorders such as diabetes mellitus (DM) and obesity (OB). The present study is an attempt to evaluate metabolic alterations in the serum and brain through NMR spectroscopy with the aim to identify shared metabolic signatures. AD was induced in rats by stereotactic intracerebroventricular injection of oligomerized Aβ-42 peptide into the brain. DM and OB were induced by intraperitoneal injection of streptozotocin and feeding r

[Influence of intranasally administered insulin on metabolic and hormonal parameters in adult male rats, impaired due to three-day fasting in the early postnatal period].

Temporary cessation or restriction of breastfeeding can lead to metabolic disorders in adulthood. However, data on the effect of fasting in the early postnatal period on the functions of the endocrine system in adulthood are rare and contradictory. Approaches for the correction of metabolic and hormonal disorders caused by premature cessation of breastfeeding have not been developed yet. The aim of the work was to study the metabolic and hormonal parameters and changes in the hormonal status of the gonadal and thyroid systems in 10-month-old male rats with interruption of breastfeeding on days

Peripartal rumination dynamics and health status in cows calving in hot and cool seasons.

Our objective was to evaluate the effect of season of calving, associated with variable levels of heat stress, on the dynamics of rumination during the prepartum period and early lactation of cows that were healthy or affected by peripartal health disorders. Three weeks before the estimated due date, 210 multiparous Holstein cows at the University of Florida Dairy Unit were affixed with a neck collar containing rumination loggers, providing rumination time (RT) in 2-h periods. One blood sample was collected in a subpopulation of cows (n=76) at 12 to 48h postcalving to assess metabolic status b

Residual analysis for the identification of potential mid-infrared-derived biomarkers of heat stress in dairy cattle.

Numerous prediction equations have been developed based on mid-infrared (MIR) spectra, and some could be potentially used as biomarkers of heat stress. However, practical experience shows that confusion can easily occur between the effect of heat stress and other effects, such as lactation stage or feeding variation over the year. On this basis, the objective of this study was to identify potential milk components predicted by MIR as biomarkers of heat stress based on a 2-step approach allowing correction for those effects. The first step consisted in the estimation of residuals from test-day

Late Embryogenesis Abundant Proteins Contribute to the Resistance of Toxoplasma gondii Oocysts against Environmental Stresses.

Toxoplasma gondii oocysts, which are shed in large quantities in the feces from infected felines, are very stable in the environment, resistant to most inactivation procedures, and highly infectious. The oocyst wall provides an important physical barrier for sporozoites contained inside oocysts, protecting them from many chemical and physical stressors, including most inactivation procedures. Furthermore, sporozoites can withstand large temperature changes, even freeze-thawing, as well as desiccation, high salinity, and other environmental insults; however, the genetic basis for this environme

Thermal performance of a low-profile cross-ventilated freestall dairy barn with evaporative cooling pads in a hot and humid climate.

A commercial low-profile cross-ventilated system (LPCV) free-stall barn with evaporative cooling pads on the air intake side was used to assess the potential of cross-ventilation with evaporative cooling to alleviate heat stress in lactating dairy cows. A total of 5712 records (238 days × 24 h) of air temperature (AT) and relative humidity (RH) values were collected. In order to state the effectiveness of the ventilation system and potential heat stress, the temperature humidity index (THI) and enthalpy (h) were calculated outside and inside the LPCV barn. The cooling capacity of the LPCV syst

The effects of heat stress on milk production and the grazing behavior of dairy Holstein cows milked by an automatic milking system.

The objective of the present study was to evaluate the productive performance and grazing behavior of 25 primiparous and 44 multiparous Holstein cows in a pasture-based automatic milking system (AMS) while experiencing heat stress (defined as a temperature-humidity index [THI] ≥ 68). Productive traits were analyzed according to the THI from days 0, -1, -2, and -3 in relation to the milking day, and grazing behaviors (expressed as the percentage of daily observation time) were related to the average THI only on the day of observation. Milk yield was not associated with the THI on day 0, but a s

Voluntary heat stress abatement system for dairy cows: Does it mitigate the effects of heat stress on physiology and behavior?

Many cooling strategies are used to keep cows in thermal homeostasis; however, most of them are applied to the group level, commonly at the feed bunk or milking parlor. The variance of heat stress effects on animals are well known, but with more individualized management in dairy farms, group cooling opportunities are becoming restricted. It is known that dairy cattle are variable in their responses to an increase in heat load. Thus, the first objective of this study was to investigate the effect of 2 mandatory soakings at the exit of the milking parlor and free access to a voluntary soaking s

TRIT1 defect leads to a recognizable phenotype of myoclonic epilepsy, speech delay, strabismus, progressive spasticity, and normal lactate levels.

TRIT1 defect is a rare, autosomal-recessive disorder of transcription, initially described as a condition with developmental delay, myoclonic seizures, and abnormal mitochondrial function. Currently, only 13 patients have been reported. We reviewed the genetic, clinical, and metabolic aspects of the disease in all known patients, including two novel, unrelated TRIT1 cases with abnormalities in oxidative phosphorylation complexes I and IV in fibroblasts. Taken together the features of all 15 patients, TRIT1 defect could be identified as a potentially recognizable syndrome including myoclonic ep
