Phenotypic conversion leads to structural and functional changes of smooth muscle sarcolemma.

Continuous changes in the length of smooth muscles require a highly organized sarcolemmal structure. Yet, smooth muscle cells also adapt rapidly to altered environmental cues. Their sarcolemmal plasticity must lead to profound changes which affect transmembrane signal transduction as well as contractility. We have established porcine vascular and human visceral smooth muscle cultures of epithelioid and spindle-shaped morphology and determined their plasma membrane properties. Epithelioid cells from both sources contain a higher ratio of cholesterol to glycerophospholipids, and express a less d

Relevance of platelet-independent effects of aspirin to its salutary effect in atherosclerosis-related events.

There is a close inter-relationship between oxidative stress, coagulation, inflammation, and smooth muscle cell growth as key components of atherosclerosis (Fig. 1). As an analgesic and anti-pyretic, aspirin has been in use for over a century. It acetylates the COX enzyme, irreversibly inhibiting the formation of prostaglandin. Its action on platelet TxA2 has highlighted its role as an anti-thrombotic agent in cardiovascular patients. Over the last two decades, unique anti-inflammatory properties of aspirin not shared by other non-steroidals have been discovered. Aspirin biotransforms into sal

Determination of Ca2+/calmodulin-stimulated phosphodiesterase activity in intact cells.

Ca(2+)/calmodulin (CaM)-stimulated phosphodiesterases (PDEs) constitute a large family (PDE1 family) of enzymes. All members of the PDE1 family can be stimulated by Ca(2+) in the presence of CaM in vitro. It has been shown that the Ca(2+)/CaM-stimulated PDE activity present in the vessel wall or vascular smooth muscle cells can be stimulated in vivo by contracting reagents that increase intracellular Ca(2+) concentrations. We describe in detail a technique used to estimate the extent of PDE1 activation in vivo by measuring in vitro the PDE activity that represents the extent of association bet

Graphene oxide-reinforced thermosensitive hydrogel co-delivering paclitaxel and dexamethasone for restenosis prevention.

Restenosis remains a significant limitation of vascular intervention procedures, primarily driven by the proliferation and inflammation of human-derived vascular smooth muscle cells (hVSMCs). This study developed a thermosensitive chitosan/β-glycerophosphate (CS-β-GP) hydrogel reinforced with graphene oxide (GO) for the localized co-delivery of paclitaxel (PTX) and dexamethasone (DEX). The incorporation of GO increased the storage modulus from approximately 3.95 kPa to 6.96 kPa, enhancing mechanical strength and accelerating gelation near body temperature (~36°C-37°C). SEM analysis revealed a

Common Genes Between Hyperlipidemia and Aortic Dissection and Their Regulatory Mechanisms.

PURPOSE: Hyperlipidemia is a potential risk factor for aortic dissection (AD), a severe cardiovascular emergency with high mortality. However, the common molecular mechanisms linking these 2 conditions remain poorly understood. This study aimed to identify common hub genes and elucidate their regulatory mechanisms in both hyperlipidemia and AD. METHODS: Transcriptome sequencing of dissected and normal ascending aorta tissues was performed to identify differentially expressed genes (DEGs) associated with AD. DEGs related to hyperlipidemia were analyzed using publicly available datasets. Common

3D bioprinted human vascular organoid sheets promote functional ischemic repair and exhibit adaptive in vivo remodeling.

BACKGROUND: Ischemic vascular diseases remain a major clinical challenge, creating a need for engineered vascular tissues that can establish functional vascular networks and promote durable tissue repair. Conventional vascular organoids offer limited control over cellular composition and spatial organization, while the fate and adaptive remodeling of graft-derived human vascular cells after transplantation remain poorly understood. METHODS: Human vascular organoid sheets (hVOS) were constructed by extrusion-based three-dimensional (3D) bioprinting of human pluripotent stem cell (hPSC)-derived

Regulating Cell-Material Interfacial Interactions through Selective Cellular Resistance.

Regulating the behavior of different types of cells at the material-tissue interface is pivotal for inducing tissue regeneration. Traditional methods enhance target cell activity using specific ligands such as peptides and antibodies, which have stability issues within biological environments. Herein, we show that selective cell resistance can be realized by fine-tuning the material surface chemistry, achieving strong cell selectivity superior to that of extracellular matrix peptides. A certain degree of adsorption resistance differentially affects the adhesion of various types of cells on mat

Multiscale computational model of aortic remodeling following postnatal disruption of TGFβ signaling.

The healthy adult aorta is a remarkably resilient structure, able to resist relentless cardiac-induced and hemodynamic loads under normal conditions. Fundamental to such mechanical homeostasis is the mechano-sensitive cell signaling that controls gene products and thus the structural integrity of the wall. Mouse models have shown that smooth muscle cell-specific disruption of transforming growth factor-beta (TGFβ) signaling during postnatal development compromises this resiliency, rendering the aortic wall susceptible to aneurysm and dissection under normal mechanical loading. By contrast, dis

Arrangement into layers and mechanobiology of multi-cell co-culture models of the uterine wall.

STUDY QUESTION: Can a co-culture of three cell types mimic the in vivo layers of the uterine wall? SUMMARY ANSWER: Three protocols tested for co-culture of endometrial epithelial cells (EEC), endometrial stromal cells (ESC), and myometrial smooth muscle cells (MSMC) led to formation of the distinct layers that are characteristic of the structure of the uterine wall in vivo. WHAT IS KNOWN ALREADY: We previously showed that a layer-by-layer co-culture of EEC and MSMC responded to peristaltic wall shear stresses (WSS) by increasing the polymerization of F-actin in both layers. Other studies sho

Engineered vascular grafts lend unique insight to pathophysiology of aortic aneurysms.

Yang et al.1 generate tissue engineered blood vessels from hiPSC-derived smooth muscle cells harboring a mutation found in Loeys-Dietz syndrome. In vitro and in vivo data from these vessels provide new insight into the molecular physiology of aortic aneurysms and may create a paradigm for understanding a suite of vascular diseases.

Integrative single-cell meta-analysis reveals disease-relevant vascular cell states and markers in human atherosclerosis.

Coronary artery disease (CAD) is characterized by atherosclerotic plaque formation in the arterial wall. CAD progression involves complex interactions and phenotypic plasticity among vascular and immune cell lineages. Single-cell RNA-seq (scRNA-seq) studies have highlighted lineage-specific transcriptomic signatures, but human cell phenotypes remain controversial. Here, we perform an integrated meta-analysis of 22 scRNA-seq libraries to generate a comprehensive map of human atherosclerosis with 118,578 cells. Besides characterizing granular cell-type diversity and communication, we leverage th

Smooth Muscle Cell Relaxation Worsens Aortic Dilatation and Clinical Presentation in a BAPN/Angiotensin II-Induced Aortic Dissection Model in Rats.

INTRODUCTION: Beta-aminopropionitrile (BAPN) administration is a chemically induced model for preclinical aortic pathologies research. Angiotensin II (AngII) has been widely used to promotes aortic dissections in mice. Here, we provide insight on a modified aortic dissection model in rats. The effect of smooth muscle cell (SMC) relaxation with vasodilators is studied in this model. METHODS: Forty Sprague-Dawley rats were divided in 4 groups: control, isosorbide dinitrate (ISDN, 30 mg/kg/day) in the drinking water, BAPN (0.02%) in the food, BAPN + ISDN (same doses). Thoracic and abdominal aort

SRSF1 promotes ASMC proliferation in asthma by competitively binding CCND2 with miRNA-135a.

BACKGROUND: Asthma is an inflammatory syndrome characterized by airway hyperresponsiveness, bronchial inflammation, and airway remodeling. Abnormal proliferation of airway smooth muscle cells (ASMCs) is the main pathological feature of asthma. This study investigated the function and mechanism of serine arginine-rich splicing factor 1 (SRSF1) in ASMC proliferation in asthma. METHODS: SRSF1 expressions in the bronchi of ovalbumin-induced asthmatic mice and IgE-treated mouse ASMCs (mASMCs) were evaluated using quantitative real-time PCR and Western blot. The localization and expression of SRSF1

Hypertensive Effect of Downregulation of the Opioid System in Mouse Model of Different Activity of the Endogenous Opioid System.

The opioid system is well-known for its role in modulating nociception and addiction development. However, there are premises that the endogenous opioid system may also affect blood pressure. The main goal of the present study was to determine the impact of different endogenous opioid system activity and its pharmacological blockade on blood pressure. Moreover, we examined the vascular function in hyper- and hypoactive states of the opioid system and its pharmacological modification. In our study, we used two mouse lines which are divergently bred for high (HA) and low (LA) swim stress-induced

Beyond Self-Recycling: Cell-Specific Role of Autophagy in Atherosclerosis.

Atherosclerosis is a chronic inflammatory disease of the arterial vessel wall and underlies the development of cardiovascular diseases, such as myocardial infarction and ischemic stroke. As such, atherosclerosis stands as the leading cause of death and disability worldwide and intensive scientific efforts are made to investigate its complex pathophysiology, which involves the deregulation of crucial intracellular pathways and intricate interactions between diverse cell types. A growing body of evidence, including in vitro and in vivo studies involving cell-specific deletion of autophagy-relate

Urethral reconstruction using an amphiphilic tissue-engineered autologous polyurethane nanofiber scaffold with rapid vascularization function.

Reconstruction and functional rehabilitation of the long urethra in males is one of the difficult tasks in urological treatment. Although many kinds of tissue-engineered urethra scaffold grafts have been successfully used in animals and even clinical research of urethra reconstruction, they all have the disadvantages of slow vascularization in scaffolds, which may lead to complications such as stricture and blockage of the urethra. Here, an amphiphilic polyurethane tubular nanofiber scaffold with a hierarchical structure was designed as a urethral scaffold. The scaffold can regulate the phenot

Oxytocin can regulate myometrial smooth muscle excitability by inhibiting the Na+ -activated K+ channel, Slo2.1.

KEY POINTS: At the end of pregnancy, the uterus transitions from a quiescent state to a highly contractile state. This transition requires that the uterine (myometrial) smooth muscle cells increase their excitability, although how this occurs is not fully understood. We identified SLO2.1, a potassium channel previously unknown in uterine smooth muscle, as a potential significant contributor to the electrical excitability of myometrial smooth muscle cells. We found that activity of the SLO2.1 channel is negatively regulated by oxytocin via Gαq-protein-coupled receptor activation of protein kina

The effects of stenting on coronary endothelium from a molecular biological view: Time for improvement?

Coronary artery stenting following balloon angioplasty represents the gold standard in revascularization of coronary artery stenoses. However, stent deployment as well as percutaneous transluminal coronary angioplasty (PTCA) alone causes severe injury of vascular endothelium. The damaged endothelium is intrinsically repaired by locally derived endothelial cells and by circulating endothelial progenitor cells from the blood, leading to re-population of the denuded regions within several weeks to months. However, the process of re-endothelialization is often incomplete or dysfunctional, promotin

Human Pluripotent Stem Cell-Derived TSC2-Haploinsufficient Smooth Muscle Cells Recapitulate Features of Lymphangioleiomyomatosis.

Lymphangioleiomyomatosis (LAM) is a progressive destructive neoplasm of the lung associated with inactivating mutations in the TSC1 or TSC2 tumor suppressor genes. Cell or animal models that accurately reflect the pathology of LAM have been challenging to develop. Here, we generated a robust human cell model of LAM by reprogramming TSC2 mutation-bearing fibroblasts from a patient with both tuberous sclerosis complex (TSC) and LAM (TSC-LAM) into induced pluripotent stem cells (iPSC), followed by selection of cells that resemble those found in LAM tumors by unbiased in vivo differentiation.
