An extracellular matrix mimicking alginate hydrogel scaffold manipulates an inflammatory microenvironment and improves peripheral nerve regeneration by controlled melatonin release.

Low efficiency of nerve growth and unstable release of loaded drugs have become a major problem in repairing peripheral nerve injury. Many intervention strategies were focused on simple drug loading, but have still been less effective. The key challenge is to establish a controlled release microenvironment to enable adequate nerve regeneration. In this study, we fabricate a multilayered compound nerve scaffold by electrospinning: with an anti-adhesive outer layer of polycaprolactone and an ECM-like inner layer consisting of a melatonin-loaded alginate hydrogel. We characterized the scaffold, a

Contralateral Astrocyte Response to Acute Optic Nerve Damage Is Mitigated by PANX1 Channel Activity.

Glial reactivity is considered a hallmark of damage-induced innate immune responses in the central nervous system. In the visual system, unilateral optic nerve damage elicits dramatic glial reactivity in the retina directly affected by the lesion and a similar, albeit more modest, effect in the contralateral eye. Evaluation of astrocyte changes in a mouse model of optic nerve crush indicates that astrocyte reactivity, as a function of retinal coverage and cellular hypertrophy, occurs within both the experimental and contralateral retinas, although the hypertrophic response of the astrocytes in

Actions of CNTF and neurotrophins on degenerating motoneurons: preclinical studies and clinical implications.

Spinal motoneurons innervating skeletal muscle were amongst the first neurons shown to require the presence of their target cells to develop appropriately. Isolated embryonic chick and rat motoneurons have been used to identify neurotrophic factors and cytokines capable of supporting the survival of developing motoneurons. Such factors include ciliary neurotrophic factor (CNTF), which is present physiologically in high amounts in myelinating Schwann cells of peripheral nerves, and brain-derived neurotrophic factor (BDNF) which is synthesized in skeletal muscle and, after peripheral nerve lesio

Challenging reconstruction of oncologic defects of the parotid region: a single-institution experience and decision-making framework proposal.

OBJECTIVES: Reconstruction of the parotid region presents challenges due to the area’s complex anatomy. A classification of defects from ablative surgery can simplify and systematize the reconstructive approach. This study aims to propose a structured decision-making framework to guide reconstructive planning. MATERIALS AND METHODS: A retrospective study was conducted on patients with malignant parotid tumors treated at Policlinico Umberto I between 2013 and 2023. Non-malignant tumors were excluded. The analysis focused on the involvement of skin, soft tissue, bone, and nerves, the type of par

The frontal branch of the facial nerve across the zygomatic arch: anatomical relevance of the high-SMAS technique.

BACKGROUND: The frontal branch has a defined course along the Pitanguy line from tragus to lateral brow, although its depth along this line is controversial. The high-superficial musculoaponeurotic system (SMAS) face-lift technique divides the SMAS above the arch, which conflicts with previous descriptions of the frontal nerve depth. This anatomical study defines the depth and fascial boundaries of the frontal branch of the facial nerve over the zygomatic arch. METHODS: Eight fresh cadaver heads were included in the study, with bilateral facial nerves studied (n = 16). The proximal frontal br

The innervation of the synovium of the knee joint in the guinea pig: an immunohistochemical and ultrastructural study.

The innervation of the knee joint synovial membrane of the guinea pig, i.e., the synoviocyte layer, the subjacent connective tissue and the connective tissue region beneath, was analyzed with immunohistofluorescence and electron microscopy. A screening of the innervation with antibodies against the general axon marker -- protein gene product (PGP) 9,5 -- revealed the presence of nerve fibers distributed in various regions of the knee joint synovial membrane. Confirming previous studies, some of these nerve fibers stained with antibodies to tyrosine hydroxylase (TH), neuropeptide Y (NPY), subst

Comparative distribution of misonidazole and its amine metabolite in female Swiss Webster mice.

The distribution of misonidazole and its terminal reduction product 1-(2-amino-1-imidazolyl)-3-methoxy-2-propanol (miso-amine) were compared in female Swiss Webster mice to determine if either misonidazole or miso-amine is distributed to peripheral nerves. Female Swiss Webster mice received a 100 mg/kg (5 microCi/mumole) i.p. dose of either 3H-misonidazole or 3H-miso-amine and the distribution of radioactivity was determined in various tissues including sciatic nerves and other myelinated nerves. Urine from misonidazole treated animals contained both miso-amine and misonidazole (8.4 and 20.4%,

Lower branches of the facial nerve.

Various aesthetic procedures have been developed over the past several decades for facial rejuvenation. One of the parallel goals is protection of vital structures such as the facial nerve. In this study, 10 cadaveric hemifacial dissections were performed, and the course of the lower portion of the facial nerve was studied with specific attention addressed toward the three-dimensional location of the nerve as it relates to the usual superficial tissue planes that are dissected during the course of an open facial rejuvenation procedure. The locations of the nerve as it runs within the different

Prenatal traces of aberrant neurofacial growth.

The interrelation between the development of the brain/peripheral nerves and that of the surrounding bone tissue is termed neuro-osteology. In orthodontic and pediatric practice the development of the hard tissues is evaluated radiographically, but the development of the neural tissue within the bone tissue is not evaluated. In this review the emphasis is placed on two neuro-osteologic interrelations that can be observed on profile radiographs and orthopantomograms, respectively. One is the connection between the pituitary gland of the central nervous system and the sella turcica (profile radi

Primary and secondary microvascular reconstruction of the upper extremity.

Tissue defects of the upper extremity may result from trauma, tumor resection, infection, and congenital malformation. Restoration of anatomy and functional integrity may require microsurgical free flap transfer for coverage of bones, nerves, blood vessels, or tendons. Microsurgical tissue transfer also may be required prior to secondary reconstruction, such as tendon transfers or nerve or bone grafts. This article addresses indications for upper extremity reconstruction using microsurgical tissue transfer flap selection and strategies including primary and secondary reconstruction.

Olfactory marker protein in the human olfactory pathway.

The presence of olfactory marker protein (OMP) in the olfactory tissue of rats, gerbils, and humans was demonstrated with goat antiserum to rat OMP. In control studies on rat neural tissue, OMP was found to be located in the olfactory receptor cells, nerves, and bulbs. Likewise, staining of OMP was found in the olfactory tissues of gerbils. Olfactory marker protein reactivity was also demonstrated in human olfactory receptor cells, nerves, and bulbs. The reactivity of OMP in human olfactory neurons to goat antiserum has allowed determination of the exact junction of olfactory and respiratory e

Mechanical tension promotes skin nerve regeneration by upregulating nerve growth factor expression.

This study aimed to explore the role of mechanical tension in hypertrophic scars and the change in nerve density using hematoxylin-eosin staining and S100 immunohistochemistry, and to observe the expression of nerve growth factor by western blot analysis. The results demonstrated that mechanical tension contributed to the formation of a hyperplastic scar in the back skin of rats, in conjunction with increases in both nerve density and nerve growth factor expression in the scar tissue. These experimental findings indicate that the cutaneous nervous system plays a role in hypertrophic scar forma

An immunohistochemical study on the postnatal changes in the C-terminal flanking peptide of neuropeptide Y (CPON)--positive nerve fibers in pineal gland of the pig.

Postnatal development of innervation of the porcine pineal gland by C-terminal flanking peptide of neuropeptide Y (CPON)-immunoreactive nerve fibers was examined using the immunohistochemical technique. CPON-positive nerve fibers appeared in the pineal gland of the new-born piglets and formed plexus in the capsula and connective tissue septa of the gland. The nerve terminals branched off from the plexus and penetrated into adjacent parenchyma. The number of nerve fibers immunoreactive to CPON in parenchyma is very low in new-born and 20-day-old piglets. In 7-month-old pigs they created a dense

Juvenile xanthogranuloma of nerve root origin.

A case of juvenile xanthogranuloma arising from the first sacral nerve root is reported together with a review of the literature. The patient presented with symptoms of low back pain and sciatica, and with S1 nerve root tensions signs. Computed tomography and magnetic resonance imaging revealed a mass arising from the right S1 nerve root. The tumour was resected, and the morphological and immunocytochemical appearances of the lesion confirmed the diagnosis of juvenile xanthogranuloma. Soft tissue juvenile xanthogranulomas have been described but are extremely rare. However, we believe this is

A rare choristoma of the optic nerve and chiasm.

Two patients with a heretofore undescribed unilateral choristomatous malformation of the intracranial optic nerve and chiasm underwent craniotomy for partial resection of the optic nerve in the belief that the tumor was either a glioma or a meningioma. Visual acuity in the affected eye was reduced to 20/200 in one patient and to finger counting in the other. Chiasmal involvement in the first case was associated with a superotemporal field defect in the opposite eye. Ophthalmoscopy disclosed atrophic nerve heads in both affected eyes. Heteroplastic masses of smooth muscle and adipose tissue rep

Ganglia of the nerve. Presentation of two unusual cases, a review of the literature, and a discussion of pathogenesis.

Two cases of intraneural ganglia are reported: the first reported involvement of the sciatic nerve and the first reported involvement of the tibial nerve in the tarsal tunnel. Both were successfully treated by excision with sparing the neural elements leading to neurological recovery. A review of the literature fails to reveal any consensus that lesions arise from normal synovial cavities. Incision, evacuation, and complete excision under magnification sparing neural elements gives satisfactory results. Excision of the nerve is not indicated. Pathologic examination suggests that the lesion ari

Prox1 induces new lymphatic vessel formation and promotes nerve reconstruction in a mouse model of sciatic nerve crush injury.

The peripheral nervous system lacks lymphatic vessels and is protected by the blood-nerve barrier, which prevents lymphocytes and antibodies from entering the neural parenchyma. Peripheral nerve injury results in degeneration of the distal nerve and myelin degeneration causes macrophage aggregation, T lymphocyte infiltration, major histocompatibility complex class II antigen expression, and immunoglobulin G deposition in the nerve membrane, which together result in nerve edema and therefore affect nerve regeneration. In the present paper, we show myelin expression was absent from the sciatic n
